The current study was conducted to evaluate the ability of Egyptian bentonite (EB) and montmorillonite (EM) for the prevention of genotoxicity, histochemical and biochemical changes induced by aflatoxin B(1) (AFB(1)) using the micronucleus (MN) assay, chromosomal aberrations and DNA fragmentation analysis in Tilapia fish. Six groups of fish were treated for 3 weeks and included the control group, AFB(1)-treated group and the groups treated with EB or EM alone or in combination with AFB(1). Formation of single strand breaks in nuclear DNA induced by hepatocarcinogens aflatoxin B1 and N-nitrosodimethylamine was observed to be more pronounced in rats maintained on a riboflavin-deficient diet compared to that on a normal diet. This increased damage was reversed on riboflavin supplementation. The effect of different vitamin A status on events following DNA damage by hepatocarcinogens was investigated in rats. Formation of single-strand breaks in nuclear DNA induced by aflatoxin B1 and N-nitrosodimethylamine was observed to be more pronounced after vitamin A-deficiency. Aflatoxin B1 is the most toxic mycotoxin and has strong carcinogenicity. In this study, coumarin was employed as the sole carbon source to isolate the microorganisms that had AFB1 detoxification activity. Among106 strains isolated from fermented foods, one potential strain was identified as Lactobacillus plantarum based on 16S rDNA sequence. Removal ratio of AFB1 was 89.5%, inhibitory ratio to A. flavus growth was 42.8% and inhibitory ratio to A. flavus spores growth was as high as 100%.
