Protocol for a primary human 2D intestinal epithelium-dendritic cell co-culture organoid model to study mucosal immunity and viral infections.

The intestinal mucosa is a major HIV-1 infection site, making immune-augmented intestinal models essential for mechanistic and therapeutic studies. We present a protocol that converts human intestinal organoids into polarized 2D epithelial monolayers and incorporates basolateral dendritic cells to create a biomimetic intestinal mucosal model. We describe steps for monolayer formation, integrity assessment, immune cell co-culture, virus inoculation, and analyses by flow cytometry and confocal microscopy. This immuno-organoid platform enables controlled HIV-1 exposure with immunological and viro

Dual-targeted glutathione-glutamate functionalized Bismuth-Niosomes hybrid nanosystem for co-delivery of doxorubicin and Pi3K inhibitor into U87 glioblastoma cells.

The simultaneous use of multiple therapeutic methods as combinational therapy could be a promising strategy to overcome glioblastoma resistance. We introduced a hybrid Bismuth-Niosomes nanocarriers containing Pi3K inhibitor and doxorubicin drug and targeted by glutathione and glutamate ligands (BiNPs-NISM-Dox/Pi3Ki@BSA-GSH-Glu). Finally, its anticancer effects were evaluated the in vitro on U87 glioblastoma cell line. The nanosystems were synthesized using a modified thin-film hydration technique and characterized using FTIR, UV-vis, DLS, TEM, FESEM, EDX, in vitro release, and hemolysis tests.

Chronic co-exposure to BPA and alcohol synergizes hepatotoxicity via dysregulation of glycerolipid metabolism and ceramide/HtrA2-driven apoptosis in HepG2 cells.

Bisphenol A (BPA) and alcohol (ALC) are recognized hepatotoxicants to which humans are chronically exposed through environmental sources, dietary habits, and preferences for alcoholic beverages. However, there is currently a lack of conclusive evidence concerning the synergistic hepatotoxic effects associated with co-exposure. This study provides mechanistic insights into BPA and ALC co-exposure-induced subcellular changes mimicking hepatotoxicity using acute and chronic 2D models and 3D HepG2/LX-2 spheroids. Results demonstrated that co-exposure amplified oxidative stress, DNA damage, and apo

High-Precision Synchronous Detection of Breath CO, CH4, and CO2 Using an NIR-WMS Sensor Based on a Dual-Channel Multipass Cell.

TDLAS-based breath sensors hold significant promise for clinical diagnostics because of their high selectivity, fast response, and operational simplicity. However, their application in high-precision, multicomponent breath analysis is often hindered by system complexity, high cost, and limited detection accuracy. To overcome these challenges, this study presents a near-infrared TDLAS system for the simultaneous monitoring of carbon monoxide (CO), carbon dioxide (CO2), and methane (CH4) in breath. The system integrates a spectral line optimization and selection strategy with a novel, miniaturiz

SOD1 at the Crossroads: Co-Overexpression of Canonical Antioxidant Response and Noncanonical Hydrogen Sulfide Generation Pathways in Down Syndrome, With Immune Cell Implications.

Accelerated immune cell aging is well-recognized feature of Down Syndrome (DS), a condition caused by trisomy of human chromosome 21 (Hsa21). DS predisposes individuals to recurrent infections, autoimmunity, low bone mass and leukemia. To investigate potential connections between immune cell dysfunction or disruption in DS, serum transcriptomic and proteomic datasets from DS and euploid individuals were examined. High DS superoxide dismutase 1 (SOD1) mRNA expression was consistently found and was strongly associated with an increased odds of inflammatory co-occurring conditions such as pharyng

Simultaneous induction of apoptosis and ferroptosis by a novel thiolato-bridged manganese(I)-based CO-releasing molecule in pancreatic and prostate cancer cells.

Due to the invasive nature of cancer cells, therapies targeting a single cell death pathway alone often encounter drug resistance; therefore, the development of novel therapeutic strategies, such as the dual induction of apoptosis and ferroptosis, is warranted. This study investigated the anticancer mechanism of the newly synthesised thiolato-bridged manganese-based CO-releasing molecule (S-MnC) in highly metastatic pancreatic (PANC-1) and prostate (PC-3) cancer cells. The myoglobin assay showed controlled liberation of approximately 1.5 CO equivalents per S-MnC molecule in the dark, and appro

From cell counts to cellular interactions: Cu-Cyto and the co-localization index as a spatial framework for the tumor immune microenvironment of rectal cancer.

The tumor immune microenvironment in colorectal cancer has emerged as a major determinant of patient outcome, with tumor-infiltrating lymphocytes, particularly CD8⁺ T cells, mediating antitumor immunity. Standardized density-based metrics such as the Immunoscore have demonstrated prognostic value but cannot capture the spatial interactions among cells that underlie immune-tumor biology. We introduce a deep learning framework that integrates two complementary axes of analysis: Cu-Cyto, a deep learning-based image cytometry platform that detects and classifies approximately twenty cell types fro

A rapid ex vivo co-culture of memory-like NK and activated T cells enhances anti-tumor response in gastric cancer.

BACKGROUND: Adoptive transfer of NK or T cells alone requires a long preparation time. We developed a rapid co-culture system, including cytokine-induced memory-like NK (CIML NK) cells and activated T cells (Ac-T), as well as tumor-penetrating peptide iRGD. METHODS: PBMCs were pretreated with IL-12/15/18 for 16 h, then washed to induce a memory NK phenotype. K562-CD48-41BBL-mbIL-21 cells were generated by lentiviral transduction and used as feeder cells to expand and activate NK cells from PBMCs for 7 days. On day 7, an anti-CD3 monoclonal antibody (OKT3) and an anti-CD28 monoclonal antibody

Co-inhibition of Notch1 and ChK1 triggers genomic instability and melanoma cell death increasing the lifespan of mice bearing melanoma brain metastasis.

BACKGROUND: Melanoma brain metastases (MBM) are a leading cause of death in patients with advanced disease. MBM treatment relay on targeted and immunotherapy and on stereotactic radiosurgery as gold standard. Life expectancy has improved significantly with these therapies however, targeted therapy is short lived and only about half of the patients respond to immunotherapy, while radiation is limited by melanoma cells intrinsic resistance to DNA damage. New therapeutic approaches are therefore needed to treat MBM. Here we investigate a new role of Notch1 in genomic instability and demonstrate t

Co-encapsulated with shikonin/doxorubicin pH-sensitive liposomes: Preparation, characterization and its synergistic effect in drug-resistant breast cancer cells.

The present study was designed to prepare co-encapsulated with shikonin/doxorubicin pH-sensitive liposomes (SHK/DOX-pHSL) and investigate their synergistic anti-cancer effect in drug-resistant breast cancer cells. SHK/DOX-pHSL exhibited a mean diameter of around 145 nm, with a polydispersity index about 0.25. SHK/DOX-pHSL displayed high encapsulation efficiency, good colloidal stability, and sustained-release property. The cumulative release rates of DOX and SHK from SHK/DOX-pHSL were significantly higher in pH 5.0 than those in pH 7.4. In vitro the results showed that SHK/DOX-pHSL could inhib

Spontaneous Efficient Differentiation of Human Pluripotent Stem Cells (hPSC) Upon Co-culture of hPSCs with Human Neonatal Foreskin Fibroblasts in 3D.

Pluripotent stem cells (PSCs) form well-formed embryoid bodies (EBs) in 3D culture. These EBs are formed in culture media lacking leukemia inhibitory factor (LIF) or basic fibroblast growth factor (bFGF) in mouse and human PSCs, respectively. EBs are excellent technical tools for understanding developmental biology and inducing controlled differentiation in succeeding experimental steps. Technically speaking, EBs are spontaneously differentiated PSCs in 3D and exhibit all three lineages in a time-point/sequential manner. For example, ectoderm will form first, followed by mesoderm and endoderm.

Integrative analysis of molecular mechanisms in prostate cancer via single-cell RNA sequencing and weighted gene co-expression network analysis.

Despite extensive prior research on prostate cancer (PCa) transcriptomics, the molecular mechanisms underlying the disease's progression, particularly in the castration-resistant or metastatic stages, remain incompletely understood. The majority of recent research has concentrated on bulk RNA sequencing, which could mask the variation found in tumor microenvironments. This study aims to address this gap by integrating single-cell RNA sequencing (scRNA-seq) and bulk RNA sequencing with weighted gene co-expression network analysis (WGCNA) to investigate the molecular mechanisms of PCa at a highe

Co-treatments of vitamin D and mesenchymal stem cells effectively alleviate the diabetic kidney disease through attenuating the SIRT1-mediated pathways.

BACKGROUND: Diabetic kidney disease (DKD) is still as a common chronic micro-vascular complication of type 1 diabetic (T1DM). Although many studies had verified the therapeutic potential of mesenchymal stem cells (MSCs) on the clinical treatment of the related complications associated with T1DM, especially the DKD, by preserving the balances of immune micro-environment and so on. However, the immunomodulatory capabilities of MSCs are still controversial on the the efficacy of DKD treatment primarily due to the variations on the immune micro-environment, which could be modulated by a variety of

In Vitro CO-Releasing and Antioxidant Properties of Sulfonamide-Based CAI-CORMs in a H2O2-Stimulated Human Achilles Tendon-Derived Cell Model.

Tendinopathy is often described as a complex and multifactorial condition which affects tendons. Tendon disorders are marked by a reduction in mechanical function, accompanied by pain and swelling. At the molecular level, tendinopathy leads to oxidative stress-driven inflammation, increased cell death, disruption of extracellular matrix balance, abnormal growth of capillaries and arteries, and degeneration of collagen formation. Here, we report an innovative approach to modulate oxidative stress during tendinopathy based on sulfonamide-based Carbonic Anhydrase Inhibitors-carbon monoxide releas

Heme Modulates Bladder Contractility Through the HO-CO-sGC-cGMP Pathway: Insights into Sickle Cell Disease-Associated Bladder Dysfunction.

Intravascular hemolysis, a hallmark of sickle cell disease (SCD), leads to elevated plasma heme levels. Although heme is essential for physiological processes, its excess can be deleterious. Heme oxygenase (HO) degrades heme into carbon monoxide (CO), which activates the soluble guanylate cyclase (sGC)-cyclic guanosine monophosphate (cGMP) signaling cascade and can modulate smooth muscle tone. However, the direct effects of heme on bladder function remain unknown. This study investigated whether heme regulates detrusor smooth muscle contractility through the HO-CO-sGC-cGMP pathway. Detrusor st

Exploring the Co-delivery of Sorafenib and Venetoclax to Induce Ferroptosis and Apoptosis Mediated Cancer Cell Death using Surface Modified PLGA Nanoparticles.

Chemotherapy resistance continues to represent a profound impediment in oncologic therapy, and the co-delivery of chemotherapeutic agents with distinct mechanisms of action offers an effective approach. In the present investigation, we developed Poly(lactic-co-glycolic acid) (PLGA) nanoparticles (NPs) loaded with Sorafenib (SOR), a ferroptosis inducer, and Venetoclax (VTX), an apoptosis inducer for combined cell death. These PLGA(SOR + VTX) NPs were formulated via nanoprecipitation followed by successive coating with polydopamine (PDA) and bovine serum albumin (BSA). PDA was utilized to facili

Co-infusion of CD19-targeting and BCMA-targeting CAR-T cells for treatment-refractory systemic lupus erythematosus: a phase 1 trial.

Systemic lupus erythematosus (SLE) remains refractory to conventional immunosuppression in a subset of patients. In treatment-refractory SLE, we show that peripheral CD19⁺ B cells and bone marrow CD19⁻BCMA⁺ long-lived plasma cells are dominant autoantibody sources, motivating dual CD19 and BCMA targeting. Here we report results from a cohort of patients (14 female, one male) in an ongoing phase 1 dose-escalation trial of co-infused autologous anti-CD19 and anti-BCMA chimeric antigen receptor (CAR) T cells after fludarabine/cyclophosphamide lymphodepletion. The Immunophenotype and the Odontogenic Commitment of Dental Pulp Stem Cells Co-Cultured with Macrophages Under Inflammatory Conditions Is Modulated by Complex Magnetic Fields.

Dental inflammatory diseases remain a challenging clinical issue, whose causes and development are still not fully understood. During dental caries, bacteria penetrate the tooth pulp, causing pulpitis. To prevent pulp necrosis, it is crucial to promote tissue repair by recruiting immune cells, such as macrophages, able to secrete signal molecules for the pulp microenvironment and thus to recruit dental pulp stem cells (DPSCs) in the damaged site. To date, root canal therapy is the standard for dental caries, but alternative regenerative treatments are gaining attention. Complex Multifrequency
