A new nanohydrogel drug delivery platform based on Laponite nanodiscs, polyacrylate, and sodium phosphate salts is described. The hybrid nanohydrogel is tailored to obtain soft and flexible nanohydrogels with G' around 3 kPa, which has been proposed as the ideal stiffness for drug delivery applications. In vitro studies demonstrate that the new nanohydrogels are biocompatible, biodegradable, nonswellable, pH-responsive, and noncytotoxic and are able to deliver antineoplastic drugs into cancer cells. The IC50 of nanohydrogels containing cisplatin, 4-fluorouracil, and cyclophosphamide is signifi

BACKGROUND: The increasing prevalence of antimicrobial-resistant bacteria, such as Escherichia coli, Klebsiella pneumoniae, and non-typhoidal Salmonella, poses a significant healthcare problem, leading to increased mortality. Liposomal nanocarriers have already shown their potential in overcoming resistance and improving antibiotic delivery.
METHODS: In this study, liposome-based nanocarriers encapsulating conventional antibiotics were developed, characterised and evaluated for their antibacterial efficacy and interactions with bacteria. The liposomal nanobiotics were prepared using the thin-

The development of nanocarrier systems for antiviral delivery presents a promising approach to overcoming the physicochemical and therapeutic limitations of conventional drugs. In this study, we optimized a thin-film hydration method to produce stable niosomes, capable of encapsulating three antiviral agents: mycophenolic acid, ribavirin, and curcumin. Physicochemical characterization revealed niosome sizes (80-200 nm), monodisperse distribution (PdI < 0.3), and negative surface charge, indicating good colloidal stability and biocompatibility. Encapsulation efficiency ranged from 35 % to 78 %,

Lactobacilli are sensitive to heat, which limits their application as probiotics in livestock production. Lactobacillus rhamnosus LB1 was previously shown to reduce enterotoxigenic Escherichia coli (ETEC) and Salmonella infections in pigs. To investigate its potential in the application, the bacterium was microencapsulated and examined for its survival from feed pelleting and long-term storage as well as its function in modulating pig intestinal microbiota. The in vitro studies showed that freshly microencapsulated Lactobacillus rhamnosus LB1 had viable counts of 9.03 ± 0.049 log10 colony-form

AIM: Coral reefs support more than one-third of the global marine biodiversity, but global stressors have negatively impacted their health by disrupting fragile microbial partnerships, leading to widespread mass bleaching events. To address this issue, the use of beneficial microorganisms for coral (BMCs) has been shown to enhance coral resilience. In this study, we evaluated the potential of alginate beads as a delivery method for BMCs underwater.
METHODS AND RESULTS: BMCs were encapsulated in alginate beads and incubated in artificial seawater at 26°C and 32°C. Microbial viability within th

The healing process of open wounds is a competition between cells and bacteria. Therefore, a strategy that can quickly remove bacteria and promote cell proliferation to accelerate wound healing is urgently needed. Inspired by photoelectric synergy tactics, we improved both the optical and electrical response of zinc oxide (ZnO) through the modification of polydopamine (PDA) and reduced graphene oxide (rGO), thus obtaining a ZnO composite named PDA-rGO-ZnO (PrZ). Combined with the photoelectric double stimulation, the sterilization target could be completed from multiple physical levels simulta

Encapsulation of cells in biocompatible polymer matrices represents a powerful tool for cell-based therapies and therapeutic delivery systems. This technology has successfully been used to deliver pancreatic islets to humans for the treatment of Type 1 diabetes. However, the clinical impact of this technology may be improved by reducing the inflammatory response brought on after implantation of capsules in vivo. Within this study a biocompatible polymeric delivery system combining alginate and photo-crosslinked methacrylated glycol chitosan (MGC) was developed. This approach involved encapsula

Cyclodextrin encapsulated beta-estradiol and progesterone were used for enhancement of gene delivery using the breast cancer cell line Sk-Br-3. A non-toxic concentration of cyclodextrin encapsulated sex steroids (50 microM) added to lipid or liposomal transfection led to a 12-fold increase of reporter gene expression (luciferase) with progesterone and an 8-fold increase with estradiol using Lipofectamine Plus mediated transfection. Using the lipid formulation Fugene-6 the results were a 5.5-fold and a 4.5-fold increase respectively. This enhancement could only be observed if the sex steroids w

Multidrug-resistant tumor cells pose significant challenges in cancer treatment. Alternative strategies such as targeted gene silencing and the use of compounds with minimal cytotoxicity toward normal cells are therefore of great interest. Antimicrobial peptides (AMPs) have demonstrated anticancer potential due to their physicochemical properties. In lung cancer, overexpression of AKT serine/threonine kinase 1 (AKT1) promotes abnormal tumor growth and progression. In this study, we synthesized chitosan-based nanoparticles (CSNPs) co-loaded with Pseudomonas aeruginosa peptide from strain P3 (PA

Glioblastoma is the most common primary malignant brain tumor with a 5-year survival rate < 5%. The standard of care involves surgical resection followed by treatment with the alkylating agent temozolomide (TMZ). GBM cells that evade surgery eventually become resistant to TMZ and lead to recurrence of tumors in patients. With only four drugs currently FDA-approved for GBM treatment, there is a need for a clinically relevant model capable of accelerating the identification of new therapies. Microgels are microscale (~10-1000 μm) hydrogel particles that can be used to encapsulate cells in a tail

Targeted delivery systems offer a promising approach for selectively modulating cellular processes; yet the intracellular consequences of targeted nutrient delivery to trophoblast cells remain poorly defined. Here, we investigated a previously validated placenta-targeting peptide conjugated to liposomes encapsulating stable isotope-labelled L-arginine and L-lysine to examine cellular uptake and downstream molecular responses in a trophoblast-like cell model. Peptide-dependent uptake of fluorescently labelled liposomes was confirmed in BeWo cells, demonstrating selective internalisation compare

Recapitulation of the microstructural organization of cellular and extracellular components found in natural tissues is an important but challenging feat for tissue engineering, which demands innovation across both process and material fronts. In this work, a highly versatile ultrasound-assisted biofabrication (UAB) approach is demonstrated that utilizes radiation forces generated by superimposing ultrasonic bulk acoustic waves to rapidly organize arrays of cells and other biomaterial additives within single and multilayered hydrogel constructs. UAB is used in conjunction with a novel hybrid b

Huntington's disease is an inherited neurodegenerative disorder caused by a CAG repeat expansion in the huntingtin gene. The peripheral innate immune system contributes to Huntington's disease pathogenesis and has been targeted successfully to modulate disease progression, but mechanistic understanding relating this to mutant huntingtin expression in immune cells has been lacking. Here we demonstrate that human Huntington's disease myeloid cells produce excessive inflammatory cytokines as a result of the cell-intrinsic effects of mutant huntingtin expression. A direct effect of mutant huntingt

Polymersomes are synthetic vesicles capable of encapsulating both hydrophilic and hydrophobic cargo, making them attractive candidates for drug delivery applications. The inherent stability of polymer vesicles, such as poly-(butadiene)33-b-poly-(ethylene oxide)20 polymersomes, is advantageous for cargo retention, but can also make controlled cargo release a challenge. In this work, nano-polymersomes containing membrane-incorporated gold nanoparticles (AuNPs) were investigated as a pulsed laser-responsive delivery platform for intracellular cargo release. Hydrophobic AuNPs embedded within the m

Probiotics as therapeutic microorganisms have been widely used for gastrointestinal disorders and wound therapy. However, poor viability in wound and gastrointestinal environments limits their efficacy. Polysaccharide-based hydrogels are suitable delivery systems for probiotics to overcome these challenges. These hydrogels by benefiting from their biocompatibility, biodegradability, and non-toxicity can create suitable microenvironments for probiotics. Unlike passive traditional carriers (such as capsules or inert nanoparticles), these "live hydrogels" are dynamic carriers encapsulating viable

Dogs are the main source of animal and human cystic echinococcosis caused by the Cestode parasite Echinococcus granulosus. Dog vaccination seems to be a good strategy to control this parasitic disease. Here we present the development of a polymeric nanoparticle-based oral vaccine for dogs against Echinococcus granulosus delivered in enteric-coated capsules. To achieve our target, we encapsulated two recombinant antigens into biodegradable polymeric nanoparticles in the presence of Monophosphoryl lipid A as an adjuvant to ensure efficient delivery and activation of a protective mucosal immune r

The infrequent exposure of pathologists to soft tissue spindle cell neoplasms coupled with overlapping histologic patterns can often make diagnosis challenging. We reviewed all nonodontogenic spindle cell neoplasms seen between 1982 and 2002 (86,162 total accessions). Diagnoses were reclassified according to current standards supplemented with immunohistochemistry. Of the 307 neoplasms reviewed (0.36% of total accessions), neural tumors were the most common benign entities, accounting for 21% of total cases. Kaposi's sarcoma was the most common malignancy, accounting for 67% of all cases. Diag

A patient-friendly and efficient treatment method for patients with spinocerebellar ataxia type 3 (SCA3) was provided through a nose-to-brain liposomal system. Initially, PGK1 was overexpressed in HEK 293-84Q-GFP diseased cells (HEK 293-84Q-GFP-PGK1 cells) to confirm its effect on the diseased protein polyQ. A decrease in polyQ expression was demonstrated in HEK 293-84Q-GFP-PGK1 cells compared to HEK 293-84Q-GFP parental cells. Subsequently, PGK1 was encapsulated in a liposomal system to evaluate its therapeutic efficiency in SCA3. The optimized liposomes exhibited a significantly enhanced pos
