The placenta ages as pregnancy advances, yet its continued function is required for a successful pregnancy outcome. Placental aging is a physiological phenomenon; however, there are some placentas that show signs of aging earlier than others. Premature placental senescence and aging are implicated in a number of adverse pregnancy outcomes, including fetal growth restriction, preeclampsia, spontaneous preterm birth, and intrauterine fetal death. Here we discuss cellular senescence, a state of terminal proliferation arrest, and how senescence is regulated. We also explore the role of physiologic…

This review delves into the molecular mechanisms that initiate cellular senescence within metabolic tissues and examines how the ensuing SASP fosters an inflammatory microenvironment, linking senescence to disorders such as insulin resistance, metabolic dysfunction-associated steatotic liver disease (MASLD), and type 2 diabetes. Cellular senescence is a common outcome of cytostatic and cytotoxic cancer therapies. While in some contexts therapy-induced senescence can drive tumor growth suppression and anti-tumor immunity through the senescence-associated secretory phenotype (SASP), in others it can promote metastatic progression and immune suppression. A systematic interrogation of the impact of different classes of therapy on heterogeneous senescent phenotypes is required to effectively apply these drugs in specific cancer contexts and to rationally design combination treatments. Using genetically-defined human and mu…

Defects in decidual response are associated with adverse pregnancy outcomes which includes recurrent pregnancy loss (RPL). It is reported that cellular senescence happens during decidualization and pro-senescent decidual response in the luteal phase endometrium is related to RPL. However, the underlying mechanisms of how excessive decidual senescence takes place in RPL decidua cells remain largely unexplored. The senescent phenotype of RPL decidua and tumor necrosis factor receptor 1(TNFR1) expression were analyzed by using our previously published single-cell sequencing dataset of decidua cel…

In patients with chronic obstructive pulmonary disease (COPD), lung-tissue regenerative mechanisms are thought to be exhausted, to which cellular senescence may contribute. Lung-derived mesenchymal stem/stromal cells (LMSCs) constitute a potent supportive cell type able to self-renew and promote alveolar regeneration. We hypothesized that LMSCs are less sensitive to senescence induction in COPD than other supportive cells, for example, lung fibroblasts (LFs), and therefore more promising in regenerative strategies. We compared senescence markers in LMSCs and LFs from the same subjects with/wit…

Background Diabetic retinopathy (DR) is a leading cause of blindness worldwide and traditionally considered a microvascular complication. However, accumulating evidence indicates that retinal neurodegeneration is also crucial in DR pathogenesis. Retinal ganglion cells (RGCs), the output neurons of the retina, are particularly vulnerable to diabetic stress. Cellular senescence has been implicated in diabetes-related tissue damage, but its contribution to RGC degeneration remains unclear. We hypothesized that diabetes contributes to retinal neurodegeneration by inducing senescence in RGCs. Metho…

Over the past years, through in vitro studies and unique animal models, biologists and clinicians have demonstrated that cellular senescence is at the root of numerous age-related chronic diseases including osteoarthritis and osteoporosis. This non-proliferative cellular syndrome can modify other surrounding tissue-resident cells through the establishment of a deleterious catabolic and inflammatory microenvironment. Targeting these deleterious cells through local or systemic seno-therapeutic agent delivery in pre-clinical models improves dramatically clinical signs and extends health span. In …

Aging is characterized by progressive changes in the physiology of brain cells, which may contribute to cognitive decline, ultimately leading to dementia and impaired quality of life. The increase in senescent cells, including glial cells in the brain, is a general feature of normal aging and has been associated with age-related pathologies. Although recent evidence suggests that astrocytes undergo senescence in these conditions, little is known about the molecular, and cellular mechanisms underlying this event. This mini review, prepared as part of the special issue Neurochemistry in Latin Am

BACKGROUND: The testis is a male-specific organ that plays a central role in spermatogenesis and androgen secretion. Testicular ageing is a hallmark of male senescence. However, the underlying mechanisms, as well as strategies to prevent, delay, or reverse this process, remain incompletely understood.
DISCUSSION: As a critical component of male ageing, testicular ageing has attracted broad scientific attention, with ongoing efforts to identify effective strategies to delay or prevent its onset, though therapeutic efficacy remains limited. In this review, we summarise the biological and clinic

Alzheimer's disease (AD) is a progressive and irreversible neurodegenerative disease, which represents the most prevalent dementia worldwide. Although amyloid-β (Aβ) and tau pathology have been the classic focus of treatment, accumulating evidence indicates that ageing-associated cellular senescence plays a central role in AD pathogenesis. Senescent neurons, astrocytes, microglia and endothelial cells accumulate in the ageing and Alzheimer's brain and adopt a senescence-associated secretory phenotype characterized by sustained release of pro-inflammatory and neurotoxic factors. This chronic in…

In this issue of Cancer Discovery, Chen and colleagues leverage single-cell RNA sequencing of neutrophils in the tumor microenvironment of colorectal cancer liver metastases to characterize neutrophil heterogeneity within the metastatic immune landscape. They show that the glucose-deprived metastatic microenvironment drives BHLHE40 upregulation in neutrophils, inducing a senescent tumor-associated neutrophil (TAN) population, TAN1, that actively promotes angiogenesis and excludes CD8+ T cells from the tumor, thereby supporting tumor growth and survival. With the acceleration of global population aging, the pathological accumulation of senescent cells (SnCs) has been confirmed as the core biological mechanism driving multiple aging-related diseases. This article aims to systematically review the formation mechanism of SnCs and their pathological roles in various tissue lesions, and to specifically evaluate the progress of three cutting-edge intervention strategies (Senolytics, Immuno-senolytics and Senomorphics). Senolytics selectively induces programmed apoptosis in SnCs by antagonizing senescent cell anti-apoptotic pathways. Immuno-senolytic

Cellular senescence is a fundamental biological process contributing to aging, often accompanied by extensive chromatin remodeling. Dynamic alterations of three-dimensional (3D) genomic spatial structure, driven by chromatin reorganization, play a critical role in cell fate determination, but their relevance in therapy-induced senescence (TIS) remains underexplored. Here, we perform an integrative multi-omics analysis of Hi-C, ATAC-seq, CUT&RUN, and RNA-seq in primary human fibroblasts undergoing TIS induced by ionizing radiation (RAD) or bleomycin (BLEO). We show that TIS leads to global chro…

Aging is multi-causal, yet its molecular hallmarks may converge on a few upstream integrating nodes. We advance the hypothesis that chronic, largely mutation-independent overactivation of the DNA damage response (DDR) is one such node, dysregulating the cell's intact guardian pathways-the tumor-suppressor networks and their negative regulators-to induce senescence, deplete stem-cell pools, and drive inflammaging. Unlike cancer, where mutations inactivate these guardians, aging more often reflects functional dysregulation of wild-type pathways, either chronically overactivated (p53, p16INK4a) o

Post-ovulatory aging (POA) reduces oocyte quality, yet effective interventions remain limited. Here we demonstrate that early intervention with vitamin K2 (menaquinone-4 isoform, MK-4) alleviates POA by inhibiting iron-disorder-induced oxidative stress. Western blot revealed that the expressions of ferroptosis suppressor protein 1 (FSP1) and vitamin K epoxide reductase-like protein (VKORC1L1) were significantly decreased in mouse POA oocytes; however, MK-4 attenuated these changes. Further, MK-4 inhibited ferritinophagy by downregulating NCOA4 and LC3II, markedly restored ferritin level, reduc

Autophagy was originally identified as a survival mechanism to allow cells to survive under nutrient-deprived or stressful conditions whereas cellular senescence was considered a tumor-suppressive mechanism. Both processes can be induced by similar stimuli and can influence each other. There have been continued debates about whether they are causally linked, whether autophagy promotes or prevents senescence or if they are independent of each other. Protein kinases play integral roles in cell fate decision and have a major influence on both autophagy and senescence. While mechanistic target of …

Ding et al. mapped over 7,000 plasma proteins to more than 40 cell types and developed machine learning aging clocks across 60,000 individuals, demonstrating that cell-type-specific biological aging is heterogeneous, measurable from blood alone, and powerfully predictive of neurodegenerative disease, cancer, and mortality up to 15 years before clinical onset.1.

Repurposing previously approved drugs may fast track the route to the clinic for potential senotherapeutics and improves the inefficiency of the clinical drug development pipeline. We performed a repurposing screen of 240 clinically approved molecules in human primary dermal fibroblasts for their effects on CDKN2A expression. Molecules demonstrating effects on CDKN2A expression underwent secondary screening for senescence-associated beta galactosidase (SAB) activity, based on effect size, direction, and/or molecule identity. Selected molecules then underwent a more detailed assessment of senes

Multiple changes occur in hormonal regulation with aging and across various endocrine organs. These changes are associated with multiple age-related disorders and diseases. A better understanding of responsible underling biological mechanisms could help in the management of multiple endocrine disorders over and above hormone replacement therapy (HRT). Cellular senescence is involved in multiple biological aging processes and pathologies common in elderly individuals. Cellular senescence, which occurs in many older individuals but also across the lifespan in association with tissue damage, acut…
