Experimental intranasal infection reveals broad tissue tropism of bovine coronavirus.

Bovine coronavirus (BCoV) causes neonatal calf diarrhea, winter dysentery, and respiratory disease worldwide. However, its primary replication site remains uncertain, and systemic studies on tissue tropism are limited. We, therefore, intranasally inoculated colostrum-fed Holstein calves (6-13-day-old) with a BCoV isolate derived from diarrheic calf, and monitored clinical signs, gross and histopathological lesions, antigen distribution, and viral replication in multiple tissues. All calves were seropositive before challenge. Infected animals developed diarrhea without respiratory manifestation…

Starting and Increasing Feeds, Milk Tolerance and Monitoring of Gut Health in Significantly Preterm Infants.

Approaches to enteral feeding significantly preterm infants' impact short-term outcomes including survival, late-onset sepsis (LOS), and necrotizing enterocolitis (NEC), and neurodevelopmental and later health outcomes. Clinical practice and trial data are dominated by short-term outcomes (NEC and LOS) with limited longer-term outcomes. Strategies maximizing early maternal breast milk (MOM) exposure and duration of MOM use are key given global health benefits of MOM, but few feeding trials use these as outcomes. Current data support colostrum receipt, early introduction, and progression of vol…

A randomised controlled trial of the immunogenicity and safety of a formaldehyde-inactivated Coxiella burnetii vaccine in 8-week-old goats.

Coxiella burnetii causes Q fever in individuals exposed to infected ruminants. Vaccination in 3-4-month-old goats, has been reported to result in significantly greater reduction in C. burnetii shedding compared to goats vaccinated one month before breeding, the most commonly used strategy of controlling Q fever on infected intensively-managed herds. It is possible that an even greater reduction in the number of animals shedding C. burnetii could be achieved if vaccination were administered shortly after protection from colostrum antibodies wanes and animals become susceptible to infection with…

Growth, health, and blood glucose concentrations of calves fed high-glucose or high-fat milk replacers.

The influence of age, carbohydrate-fat ratios of milk replacers, and development of ruminal function on growth, health, and blood glucose concentrations were evaluated in calves. Colostrum-fed, 3-day-old Holstein bull calves were fed to 12 wk on one of three dietary treatments: 1) a high carbohydrate, low fat (60.5% glucose, 9.5% lactose, and 3% lard) milk replacer; 2) a low carbohydrate, high fat (23% glucose, 12.5% lactose, and 30% lard) milk replacer; and 3) weaning at 6 wk of age from high-fat replacer to a standard calf starter. The high fat milk replacer was superior to low fat milk repl…

Milk Odd- and Branched-Chain Fatty Acids as Biomarkers of Rumen Fermentation.

Cow's milk and dairy products are the primary sources of OBCFAs, which have beneficial health properties. The goal of this study was to iden … entially useful noninvasive indicators of rumen fermentation. The concentration of BCFA is lower in colostrum and transitional milk than in full lactation milk. The proportions of total OBCFAs are higher in first- and second-parity cows. The most effective predictors of the biohydrogenation of fatty acids in the rumen are likely C18:2 cis-9, trans-11, iso-C …

Administration of granulocyte-colony stimulating factor (G-CSF) to pigs results in a longer mean survival time after exposure to Streptococcus suis.

The use of immunomodulators is a promising alternative to the use of antibiotics for therapeutic, prophylactic, and metaphylactic use to prevent and combat infectious disease. Previously we demonstrated a replication-defective adenovirus vector that expresses porcine granulocyte colony-stimulating factor (G-CSF) elicited a sustained neutrophilia, lasting nearly 3 weeks, which may be beneficial to prevent bacterial diseases during times of peak incidence. In a pilot study using the vectored G-CSF with a Caesarian-derived, colostrum-deprived (CDCD) pig model of Streptococcus suis disease, only 1…

Dual binding mode of antithyroid drug methimazole to mammalian heme peroxidases - structural determination of the lactoperoxidase-methimazole complex at 1.97 Å resolution.

Lactoperoxidase (LPO, EC 1.11.1.7) is a member of the mammalian heme peroxidase family which also includes thyroid peroxidase (TPO). These two enzymes have a sequence homology of 76%. The structure of LPO is known but not that of TPO. In order to determine the mode of binding of antithyroid drugs to thyroid peroxidase, we have determined the crystal structure of LPO complexed with an antithyroid drug, methimazole (MMZ) at 1.97 Å resolution. LPO was isolated from caprine colostrum, purified to homogeneity and crystallized with 20% poly(ethylene glycol)-3350. Crystals of LPO were soaked in a res…

Pathogenesis and epidemiology of brucellosis in yellowstone bison: serologic and culture results from adult females and their progeny.

Our objective in this prospective study was to determine the natural course of Brucella abortus infection in cohorts of seropositive and ser … ctive against infection, even for calves that passively received antibody from an infected mother's colostrum. Antibody levels stayed remarkably constant, with only a slow decline over time. We found only two seroconversions from a weak positive status to negative. Natural polyreactive secretory immunoglobulin A autoantibodies as a possible barrier to infection in humans.

Secretory immunoglobulin A (S-IgA) was investigated in human secretions for the presence of natural antibodies (Abs) acting as the first "immune barrier" to infection before induction or boosting of specific responses. These molecules could be the secretory counterpart of the natural Abs in serum that were previously shown by our laboratory to be polyreactive to autoantigens. Significant levels of S-IgA Abs to human actin, myosin, tubulin, and spectrin were detected in 10 saliva and 8 colostrum samples from normal subjects. Computer-assisted analysis of immunoblots of extracts from human muscl…

Immunofluorescence studies on the pathogenesis of hemagglutinating encephalomyelitis virus infection in pigs after oronasal inoculation.

Hemagglutinating encephalomyelitis virus (HEV; also designated vomiting and wasting disease virus) was inoculated oronasally in 14 colostrum-deprived pigs at the day of birth. Anorexia and vomition were seen after 4 days. Pigs were killed at different times after inoculation, and the results of the examination by immunofluorescent antibody technique revealed that the epithelial cells of nasal mucosa, tonsils, lungs, and small intestine served as sites of primary viral replication. After the local replication near the sites of entry, the virus spread via peripheral nervous system to the CNS. Du…
