Targeting Ferroptosis by Natural Compounds in Nephrotoxicity: A Review.

Nephrotoxicity refers to the damage caused to the kidneys by drugs and toxic substances, leading to acute and chronic kidney injury. Ferroptosis is a type of iron-driven cell death where oxidative stress causes lipid peroxide accumulation in cells, which is critical in the nephrotoxicity pathogenesis. Several studies have investigated the protective effects of natural compounds against nephrotoxicity, including curcumin, quercetin, and baicalein. This review highlights that natural compounds reduce ferroptosis in nephrotoxicity by enhancing protective pathways such as GPX4, Nrf2/GPX4, and SIRT

The Key Glycolytic Enzyme Phosphofructokinase Is Involved in Resistance to Antiplasmodial Glycosides.

Plasmodium parasites rely heavily on glycolysis for ATP production and for precursors for essential anabolic pathways, such as the methylerythritol phosphate (MEP) pathway. Here, we show that mutations in the Plasmodium falciparum glycolytic enzyme, phosphofructokinase (PfPFK9), are associated with in vitro resistance to a primary sulfonamide glycoside (PS-3). Flux through the upper glycolysis pathway was significantly reduced in PS-3-resistant parasites, which was associated with reduced ATP levels but increased flux into the pentose phosphate pathway. PS-3 may directly or indirectly target e

Molecular pathways and targeted therapies in colorectal liver metastasis: from bench to bedside.

Colorectal liver metastasis (CRLM) is a major complication of colorectal cancer (CRC), significantly affecting prognosis and survival. Despite advancements in treatment, the management of CRLM remains challenging, primarily due to the complex molecular mechanisms involved. The key molecular pathways that contribute to CRLM development include the Wnt/β-catenin signaling, epidermal growth factor receptor (EGFR), and angiogenesis pathways. These pathways regulate critical processes such as tumor cell proliferation, invasion, and metastasis, and are central to the formation and progression of CRL

Signaling pathway regulators in preimplantation embryos.

Embryonic development during the preimplantation stages is highly sensitive and critically dependent on the reception of signaling cues. The precise coordination of diverse pathways and signaling factors is essential for successful embryonic progression. Even minor disruptions in these factors can result in physiological dysfunction, fetal malformations, or embryonic arrest. This issue is particularly evident in assisted reproductive technologies, such as in vitro fertilization, where embryonic arrest is frequently observed. A detailed understanding of these pathways enhances insight into the

Recent Advances in the Role of Non-coding RNAs in Fetal Alcohol Spectrum Disorders.

Despite numerous preclinical studies modeling fetal alcohol spectrum disorder (FASD)-associated neurodevelopmental deficits to date, a comprehensive molecular landscape dictating these deficits remains poorly understood. Non-coding RNAs constitute a substantial layer of epigenetic regulation of gene expression at the transcriptional, post-transcriptional, translational, and post-translational levels. Yet, little is known about the differential expression of non-coding RNAs in the context of prenatal alcohol exposure (PAE) that are mechanistically linked with FASD-related neurobehavior deficits

Regulation of Sterol Biosynthesis in the Human Fungal Pathogen Aspergillus fumigatus: Opportunities for Therapeutic Development.

Sterols are a major component of eukaryotic cell membranes. For human fungal infections caused by the filamentous fungus Aspergillus fumigatus, antifungal drugs that target sterol biosynthesis and/or function remain the standard of care. Yet, an understanding of A. fumigatus sterol biosynthesis regulatory mechanisms remains an under developed therapeutic target. The critical role of sterol biosynthesis regulation and its interactions with clinically relevant azole drugs is highlighted by the basic helix loop helix (bHLH) class of transcription factors known as Sterol Regulatory Element Binding

The role of innate immune receptors in the control of Brucella abortus infection: toll-like receptors and beyond.

Research into intracellular sensing of microbial products is an up and coming field in innate immunity. Toll-like receptors (TLRs) recognize Brucella spp. and bacterial components and initiate mononuclear phagocyte responses that influence both innate and adaptive immunity. Recent studies have revealed the intracellular signaling cascades involved in the TLR-initiated immune response to Brucella infection. TLR2, TLR4 and TLR9 have been implicated in host interactions with Brucella; however, TLR9 has the most prominent role. Further, the relationship between specific Brucella molecules and vari

Mechanism and regulation of programmed cell death in aging-related diseases.

Aging and its associated diseases are characterized by a complex interplay of cellular dysfunction, chronic inflammation, and tissue degeneration. Dysregulated cell death and the resulting inflammatory responses are pivotal drivers of this pathological synergy. Among various cell demise pathways, programmed cell death (PCD) mechanisms, including apoptosis, necroptosis, and pyroptosis, are critical for maintaining homeostasis but can drive pathology when aberrantly activated. Crucially, they leave specific and reliable biomarkers, allowing for their distinct assessment in complex aging tissues.
