Thyroid Function Affects the Risk of Stroke via Atrial Fibrillation: A Mendelian Randomization Study.

CONTEXT: Observational studies suggest that variations in normal range thyroid function are associated with cardiovascular diseases. However, it remains to be determined whether these associations are causal or not. OBJECTIVE: To test whether genetically determined variation in normal range thyroid function is causally associated with the risk of stroke and coronary artery disease (CAD) and investigate via which pathways these relations may be mediated. DESIGN, SETTING, AND PARTICIPANTS: Mendelian randomization analyses for stroke and CAD using genetic instruments associated with normal rang

The Influence of Tobacco Smoke/Nicotine on CYP2A Expression in Human and African Green Monkey Lungs.

CYP2A enzymes metabolically inactivate nicotine and activate tobacco-derived procarcinogens [e.g., 4-[methylnitrosamino]-1-(3-pyridyl)-1-butanone]. Smoking decreases nicotine clearance, and chronic nicotine reduces hepatic CYP2A activity. However, little is known about the impact of smoking or nicotine on the expression of CYP2A in the lung. We investigated 1) the levels of human lung CYP2A mRNA in smokers versus nonsmokers and 2) the impact of daily nicotine treatment on lung CYP2A protein levels in African green monkeys (AGMs). Lung CYP2A13, CYP2A6, and CYP2A7 (and CYP1A2) mRNA levels in smo

The genetic architecture of sporadic and multiple consecutive miscarriage.

Miscarriage is a common, complex trait affecting ~15% of clinically confirmed pregnancies. Here we present the results of large-scale genetic association analyses with 69,054 cases from five different ancestries for sporadic miscarriage, 750 cases of European ancestry for multiple (≥3) consecutive miscarriage, and up to 359,469 female controls. We identify one genome-wide significant association (rs146350366, minor allele frequency (MAF) 1.2%, P = 3.2 × 10-8, odds ratio (OR) = 1.4) for sporadic miscarriage in our European ancestry meta-analysis and three genome-wide significant associations fo

Diet modulates brain network stability, a biomarker for brain aging, in young adults.

Epidemiological studies suggest that insulin resistance accelerates progression of age-based cognitive impairment, which neuroimaging has linked to brain glucose hypometabolism. As cellular inputs, ketones increase Gibbs free energy change for ATP by 27% compared to glucose. Here we test whether dietary changes are capable of modulating sustained functional communication between brain regions (network stability) by changing their predominant dietary fuel from glucose to ketones. We first established network stability as a biomarker for brain aging using two large-scale (n = 292, ages 20 to 85

Recurrent moderate-risk mutations in Finnish breast and ovarian cancer patients.

Mutations in BRCA1 and BRCA2 genes predispose to breast and ovarian cancer (BC/OC) with a high lifetime risk, whereas mutations in PALB2, CHEK2, ATM, FANCM, RAD51C and RAD51D genes cause a moderately elevated risk. In the Finnish population, recurrent mutations have been identified in all of these genes, the latest being CHEK2 c.319+2T>A and c.444+1G>A. By genotyping 3,156 cases and 2,089 controls, we estimated the frequencies of CHEK2 c.319+2T>A and c.444+1G>A in Finnish BC patients. CHEK2 c.319+2T>A was detected in 0.7% of the patients, and it was associated with a high risk of BC in the uns

A practical approach to illustrate the importance of the bodily energy and heat balances and of the associated regulatory loops to healthcare students.

BACKGROUND: Training medical students is a demanding task, and some contents remain difficult to teach and understand. This also applies to topics that require increasing attention. For instance, obesity rates keep rising, while almost each year turns out to be hotter than the last. Consequently, it is inevitable that current healthcare students will one day be confronted with these challenges. But the underlying physiological principles are concepts that prove difficult to understand. To remedy this situation and improve students' awareness and understanding of impacted patients, we have deve

Research Trends in Neutrophil Extracellular Traps and Ischemia-Reperfusion Injury: A Dual-Database Bibliometric Analysis from 2013 to 2025.

INTRODUCTION: Neutrophil extracellular traps (NETs), defined as DNA-protein complexes released upon neutrophil activation, have been increasingly recognized as pivotal mediators in the initiation and perpetuation of ischemia-reperfusion injury (IRI) in recent scholarly investigations. Despite the increasing number of related studies, there is no bibliometric analysis in this field. This study aims to analyze the development trend, research status, and hotspots of NETs and IRI, thereby providing a comprehensive and intuitive academic reference for investigators in this field. METHODS: The Web

Unveiling underutilized public data on Canadian industrial pollutant transfers and disposals.

Over 50 nations worldwide have Pollutant Release and Transfer Registers (PRTRs), including Canada's National Pollutant Release Inventory (NPRI), which comprise large public datasets of chemical releases to air, water, and land and also transfers to various on and off-site waste management practices. These inventories aim to support a myriad of audiences in pollution-related decision-making. While the Organisation for Economic Cooperation and Development (OECD) framed a role for PRTRs as indicators for Sustainable Development Goal (SDG) 12 - the sound management of chemicals and wastes, researc

Competing risk analysis for predicting distant metastasis in patients with upper urinary tract urothelial carcinoma following radical nephroureterectomy: based on a large dataset from a national medical center.

BACKGROUND: The use of traditional Cox regression models to identify risk factors for distant metastasis (DM) after RNU (Radical nephroureterectomy) in UTUC (upper urinary tract urothelial carcinoma) patients may introduce bias. This study aims to utilize a large UTUC dataset from our center and apply the Fine-Gray model to determine predictive factors for DM. Additionally, we manage to construct a prediction model that can accurately estimate the likelihood of DM following RNU. METHODS: A retrospective analysis was conducted on the clinical and pathological data of 2,546 patients with UTUC f

Precision, prognosis, and clinical performance of rounded and trabecular segmentation of cine cardiovascular magnetic resonance.

BACKGROUND: Measurements of cardiac size and function drive clinical decisions. Left ventricular (LV) metrics can be derived from cardiac MR images by delineating the blood pool and myocardium, by either drawing a rounded contour to approximate the compacted myocardial border, or by delineating the papillary muscles and trabeculae (trabecular segmentation). There is no consensus as to which is best, particularly in the emergent AI era. We developed machine-learning (ML) approaches for both and compared them for clinically important metrics (error rate, precision, and prognosis). METHODS: Sepa

PodoSighter: A Cloud-Based Tool for Label-Free Podocyte Detection in Kidney Whole-Slide Images.

BACKGROUND: Podocyte depletion precedes progressive glomerular damage in several kidney diseases. However, the current standard of visual detection and quantification of podocyte nuclei from brightfield microscopy images is laborious and imprecise. METHODS: We have developed PodoSighter, an online cloud-based tool, to automatically identify and quantify podocyte nuclei from giga-pixel brightfield whole-slide images (WSIs) using deep learning. Ground-truth to train the tool used immunohistochemically or immunofluorescence-labeled images from a multi-institutional cohort of 122 histologic secti

COVIDiagnosis-Net: Deep Bayes-SqueezeNet based diagnosis of the coronavirus disease 2019 (COVID-19) from X-ray images.

The Coronavirus Disease 2019 (COVID-19) outbreak has a tremendous impact on global health and the daily life of people still living in more than two hundred countries. The crucial action to gain the force in the fight of COVID-19 is to have powerful monitoring of the site forming infected patients. Most of the initial tests rely on detecting the genetic material of the coronavirus, and they have a poor detection rate with the time-consuming operation. In the ongoing process, radiological imaging is also preferred where chest X-rays are highlighted in the diagnosis. Early studies express the pa

ACE2 and TMPRSS2 expression by clinical, HLA, immune, and microbial correlates across 34 human cancers and matched normal tissues: implications for SARS-CoV-2 COVID-19.

BACKGROUND: Pandemic COVID-19 by severe acute respiratory syndrome (SARS) coronavirus 2 (SARS-CoV-2) infection is facilitated by the ACE2 receptor and protease TMPRSS2. Modestly sized case series have described clinical factors associated with COVID-19, while ACE2 and TMPRSS2 expression analyses have been described in some cell types. Patients with cancer may have worse outcomes to COVID-19. METHODS: We performed an integrated study of ACE2 and TMPRSS2 gene expression across and within organ systems, by normal versus tumor, across several existing databases (The Cancer Genome Atlas, Census of

Identification of gene signature for treatment response to guide precision oncology in clear-cell renal cell carcinoma.

Clear-cell renal cell carcinoma (ccRCC) is a common therapy resistant disease with aberrant angiogenic and immunosuppressive features. Patients with metastatic disease are treated with targeted therapies based on clinical features: low-risk patients are usually treated with anti-angiogenic drugs and intermediate/high-risk patients with immune therapy. However, there are no biomarkers available to guide treatment choice for these patients. A recently published phase II clinical trial observed a correlation between ccRCC patients' clustering and their response to targeted therapy. However, the c
