Anti-cancer Drugs in Endometriosis Management: Mechanisms and Therapeutic Potential.

INTRODUCTION: Endometriosis is a widespread estrogen-driven condition causing pelvic pain and infertility in women. This disease shares five features with cancer: Intrinsic growth signals, insensitivity to antiproliferative signals, impaired apoptosis, induction of angiogenesis, and heightened tissue invasion, suggesting common therapeutic targets for both conditions. This article reviews studies investigating the anti-cancer drugs' protective effects and mechanisms in endometriosis treatment, providing essential insights into their efficacy and the relevant pathways in managing the disease.

Plant molecular farming: a promising frontier for orphan drug production.

Orphan diseases comprise a range of disorders that individually affect a small percentage of people, but collectively impact millions of people worldwide. Patients with this disorder often face significant challenges in diagnosis, treatment, and access to care due to their rare nature and limited understanding and treatment options. In recent years, significant advancements have been made in the global healthcare in addressing the accessibility of essential treatments and medicines, but still challenges persist particularly related to orphan drugs (to treat rare diseases) in the developing wor

Assessment of Prescribing Pattern and Safety Profile of Drugs Used in Intranasal Route in Paediatric Age Group of Patients in a Tertiary Care Hospital.

Background Recently there has been an increased preference for intranasal delivery of drugs due to highly vascular nasal mucosa, bypassing first pass metabolism and the blood brain barrier (BBB) lead in quick drug absorption to the systemic circulation and direct access to brain from olfactory region. For pediatric patients this route offers significant benefits over injections or oral routes, like increased compliance, easy administration, and minimal side effects. Objective Assessment of prescription pattern of drugs and safety profile of drugs used by intranasal route in paediatric age grou

Anti-VEGFR therapy is one of the healing inhibitors of antiresorptive-related osteonecrosis of the jaw.

INTRODUCTION: Antiresorptive-related osteonecrosis of the jaw (ARONJ) is a rare but serious adverse event associated with bone-modifying agents (BMAs) and affects patients in the terminal stages of cancer. Molecular targeting drugs (MTDs), anti-vascular endothelial growth factor receptor (VEGFR), and anti-epidermal growth factor receptor (EGFR) drugs are essential in various cancer treatments, although MTDs are risk factors for ARONJ. However, the mechanism through which MTDs affect treatment prognosis of ARONJ remains unclear. Therefore, we investigated the potential inhibitory factors for he

Carbapenems and Rifampin Exhibit Synergy against Mycobacterium tuberculosis and Mycobacterium abscessus.

An effective regimen for treatment of tuberculosis (TB) is comprised of multiple drugs that inhibit a range of essential cellular activities in Mycobacterium tuberculosis. The effectiveness of a regimen is further enhanced if constituent drugs act with synergy. Here, we report that faropenem (a penem) or biapenem, doripenem, or meropenem (carbapenems), which belong to the β-lactam class of antibiotics, and rifampin, one of the drugs that forms the backbone of TB treatment, act with synergy when combined. One of the reasons (carba)penems are seldom used for treatment of TB is the high dosage le

The effect of acidic and basic additives on the enantioseparation of basic drugs using polysaccharide-based chiral stationary phases.

The enantioseparation of nine commercially available basic drugs was achieved on polysaccharide-based chiral stationary phases with the acidic additive ethanesulfonic acid and the basic additive butylamine. Seven different commercially available CSPs were used for the study (AD, AS, OD, OJ, OG, OB, and OC). Mobile phase additives have been proven to be essential in obtaining satisfactory enantio-resolution in terms of both efficiency and selectivity. Significantly improved selectivities were obtained for the basic probe drugs with the acidic additive, ethanesulfonic acid, rather than the basic

Drug Target Elucidation Through Isolation and Analysis of Drug-Resistant Mutants in Cryptococcus neoformans.

Drug target identification is an essential component to antifungal drug development. Many methods, including large chemical library screening, natural product screening, and drug repurposing efforts, can identify compounds with favorable in vitro antifungal activity. However, these approaches will often identify compounds with no known mechanism of action. Herein, we describe a method utilizing the human fungal pathogen Cryptococcus neoformans to identify antifungal drug targets through the isolation of spontaneous resistant mutants, antifungal testing, whole-genome sequencing, and variant ana

The elements of life and medicines.

Which elements are essential for human life? Here we make an element-by-element journey through the periodic table and attempt to assess whether elements are essential or not, and if they are, whether there is a relevant code for them in the human genome. There are many difficulties such as the human biochemistry of several so-called essential elements is not well understood, and it is not clear how we should classify elements that are involved in the destruction of invading microorganisms, or elements which are essential for microorganisms with which we live in symbiosis. In general, genes do

[Tin compounds in pharmacy and nutrition].

The occurrence of tin in plants, animals and humans is discussed, in relation to its abundance in the lithosphere and hydrosphere and the range of the different tin(II) and tin(IV) complexes formed. A reasoned consideration of its essentiality for living species is provided. It is concluded that tin is beneficial, even if not yet proved to be an essential element. After reference to the chemistry of tin compounds, there is a detailed discussion of their toxicity in animals and humans. Feasible routes for tin intake and uptake into humans are described. The past and current use of tin pharmaceu

Effect of long-term celiprolol therapy on haemostasis in essential hypertension.

Undesirable changes of haemostasis induced by some anti-hypertensive drugs can encourage the acceleration of atherogenesis. Therefore, the changes of haemostasis parameters in 22 patients with essential hypertension under long-term celiprolol therapy (> 2 months) were of interest. In the placebo group of 15 essentially hypertensive patients there were no significant changes in platelet activity. On the other hand, the therapeutic dose of celiprolol was shown to reduce total platelet aggregation, without any harmful effects on fibrinolytic activity and coagulation inhibitors such as protein C a

Averting transmission: A pivotal target to manage amoebiasis.

Amoebiasis is a parasitic infectious disease caused by the enteric protozoan Entamoeba histolytica, a leading basis of deaths accounted to parasites, succeeding malaria and schistosomiasis. Conventional treatment methodologies used to deal with amoebiasis mainly rely on the administration of anti-amoebic compounds and vaccines but are often linked with substantial side-effects on the patient. Besides, cases of development of drug resistance in protozoans have been recorded, contributing further to the reduction in the efficiency of the treatment. Loopholes in the efficacious management of the

Clinically significant drug interactions of antiretrovirals.

Evaluating of the clinical significance of drug interactions is one of the domains of clinical pharmacist expertise. Drug interactions are discussed when two or more drugs co-administer and the effect of one of them changes. Drug interactions can be a serious cause of toxicity and side effects. They can also lead to the failure of the treatment. On the other hand, they can also play a positive role, e. g. in the treatment of people living with HIV. The combination of some antiretrovirals with boosters is based on the principle of drug interaction. The article presents pharmacokinetic drug inte

[Euthanasia of pregnant animals].

For euthanasia of pregnant animals, a rapid and painless death is essential. The technique of euthanasia should minimize stress and anxiety experienced by the animal so far as possible. The death is induced by an anoxia in the central nervous system or by a pharmacologic inhibition of essential neuronal functions. Pentobarbital is the best suited drug for euthanasia of animals and especially of pregnant animals. Combinations with muscle relaxing agents should not be used because of possible apnoe without unconsciousness.

Green synthesis of nano-liposomes containing Bunium persicum and Trachyspermum ammi essential oils against Trichomonasvaginalis.

BACKGROUND: Trichomonas vaginalis, a parasitic flagellated protozoan, is one of the main non-viral sexually transmitted diseases worldwide. Treatment options for trichomoniasis are limited to nitroimidazole compounds. However, resistance to these drugs has been reported, which requires the development of new anti-Trichomonas agents that confer suitable efficacy and less toxicity. METHODS: In the present work, we assessed the effectiveness of the liposomal system containing essential oils of Bunium persicum and Trachyspermum ammi against T. vaginalis in vitro. The chemical composition of B. pe

A microbe-drug association prediction model based on graph attention networks and rotation forest.

BACKGROUND: In recent years, with the diversification and expansion of drug research in the medical field, the widespread use of drugs, particularly antibiotics, has led to increased microbial resistance. Consequently, exploring potential associations between drugs and microbes has become critically important. However, traditional biological experiments are extremely expensive and time-consuming. Therefore, developing more effective computational models for predicting potential associations between microbes and drugs is both essential and challenging. RESULTS: We proposed GATROF, a hybrid het

Ultrasonic head-mounted device spatiotemporal opening blood-brain barrier enhances the brain permeation of drugs for treatment of radiation-induced brain injury.

The field of physical therapy is advancing and using focused ultrasound to deliver drugs into the brain gains growing interest. However, the blood-brain barrier makes it difficult for drugs to enter. Finding safe and efficient physical therapy strategies to complement drug treatments is essential. Here, the rule and molecular mechanisms of spatiotemporal opening blood-brain barrier of ultrasound were explored using a Bluetooth-controlled ultrasonic head-mounted device which was used to enhance the brain permeation of drugs for the treatment of radiation-induced brain injury. Analysis of the interaction of antimalarial agents with Plasmodium falciparum glutathione reductase through molecular mechanical calculations.

CONTEXT: Malaria remains a significant global health challenge with emerging resistance to current treatments. Plasmodium falciparum glutathione reductase (PfGR) plays a critical role in the defense mechanisms of malaria parasites against oxidative stress. In this study, we investigate the potential of targeting PfGR with conventional antimalarials and dual drugs combining aminoquinoline derivatives with GR inhibitors, which reveal promising interactions between PfGR and studied drugs. The naphthoquinone Atovaquone demonstrated particularly high affinity and potential dual-mode binding with th
