A comparison of AmBisome to amphotericin B for treatment of systemic candidiasis in very low birth weight infants.

PURPOSE: Amphotericin B is considered the treatment of choice for systemic candidiasis, but adverse effects may limit its use. An alternative option for the treatment of candidiasis includes lipid preparations of amphotericin B. This study investigated the safety and efficacy of AmBisome, a lipid formulation of amphotericin B containing liposomal structures, for the treatment of systemic candidiasis in very low birth weight infants (VLBWI). MATERIALS AND METHODS: Data from 26 VLBWI treated with AmBisome in the study group (AmBisome group) from October 2003 to July 2006 were compared with data

Filamentous Fungi and the Biodeterioration of Organic Cultural Heritage Materials: A Systematic Review of Mechanisms, Risks, and Preventive Conservation Strategies.

Filamentous fungi are among the most significant biological agents responsible for the biodeterioration of organic cultural heritage materials preserved in archives, libraries, and museums. Cellulose-based substrates-such as paper, papyri, and plant-derived textiles-as well as protein-based materials, including parchment and leather, provide favourable conditions for fungal colonization due to their chemical composition and hygroscopic behaviour. Once activated, fungi contribute to deterioration through a combination of mechanical penetration and biochemical processes, including the secretion

DNA Aptamers Targeting BcSOD1: A Novel Strategy for Controlling Botrytis cinerea in Sustainable Agriculture.

Botrytis cinerea, the necrotrophic fungus responsible for grey mould disease, is a major threat to global crop production. Control strategies mainly rely on chemical fungicides, but resistance development limits their long-term effectiveness. This study introduces, for the first time in crop protection, the use of DNA aptamers as a novel and sustainable strategy. Aptamers are short, single-stranded DNA molecules that bind specific targets with high affinity, acting like 'chemical antibodies'. Using SELEX technology, two aptamers, SOD9.14F and SOD9.26F, were designed to target BcSOD1, a superox

Elucidating the virulence roles of secreted aspartyl proteinase (Sap 1972) in non-albicans Meyerozyma guilliermondii through construction of a structure-guided mutant.

Meyerozyma guilliermondii (Candida guilliermondii) is one of the non-albicans Candida species associated with invasive candidiasis. M. guilliermondii strain SO that was isolated from spoiled orange could express industrially important enzymes without hazardous methanol as inducer. Our prior studies confirmed the presence, expression, and activity of virulence-associated secreted aspartyl proteinases (MgSap). Given the time-consuming process to delete all MgSAP members from the fungal genome, this study aimed to elucidate the virulence roles of MgSap by constructing a structure-guided mutant. A

Rapid Cytological Diagnosis With Evaluation of Pre- and Post-Therapeutic Fungal Morphological Characteristics in Mucormycosis.

BACKGROUND: Mucormycosis necessitates rapid diagnosis and treatment. Microscopy and culture have been considered the gold standard for diagnosis but both take time of 3 - 5 days. KOH mount is another method for fungal identification that takes 1 - 2 h, but it has its own limitations. This study evaluated crush smear as a means of rapid cytological diagnosis. METHODS: Biopsy tissue (pre-treatment) from clinically suspicious mucormycosis patients (n = 52) was received in normal saline and crush/imprint smears were prepared; the remaining tissue was processed as routine biopsy specimen. After th

Comparison between Conventional Decalcification and a Microwave-Assisted Method in Bone Tissue Affected with Mycetoma.

Mycetoma is a lifelong granulomatous disease of subcutaneous tissues and bones. Histopathology is a substantiated indicative method based on the assumption of a definitive diagnosis of mycetoma. It requires efficient processing of tissues including bone decalcification. The decalcification process must ensure complete removal of calcium and also a proper preservation of tissue and microorganisms' staining ability. Objectives. To compare the conventional method used in decalcification with the microwave method using different decalcification solutions. Different characteristics were tested, inc

Elevated early callose deposition results in complete penetration resistance to powdery mildew in Arabidopsis.

A common response by plants to fungal attack is deposition of callose, a (1,3)-β-glucan polymer, in the form of cell wall thickenings called papillae, at site of wall penetration. While it has been generally believed that the papillae provide a structural barrier to slow fungal penetration, this idea has been challenged in recent studies of Arabidopsis (Arabidopsis thaliana), where fungal resistance was found to be independent of callose deposition. To the contrary, we show that callose can strongly support penetration resistance when deposited in elevated amounts at early time points of infec

Catalases play differentiated roles in the adaptation of a fungal entomopathogen to environmental stresses.

The catalase family of Beauveria bassiana (fungal entomopathogen) consists of catA (spore-specific), catB (secreted), catP (peroxisomal), catC (cytoplasmic) and catD (secreted peroxidase/catalase), which were distinguished in phylogeny and structure and functionally characterized by constructing single-gene disrupted and rescued mutants for enzymatic and multi-phenotypic analyses. Total catalase activity decreased 89% and 56% in ΔcatB and ΔcatP, corresponding to the losses of upper and lower active bands gel-profiled for all catalases respectively, but only 9-12% in other knockout mutants. Understanding metabolic interactions between bacteria and fungi for cross-kingdom consortia applications.

Bacterial-fungal interactions represent fundamental ecological associations that shape microbial community structure across diverse environments. While traditionally framed through the lens of antagonism, bacteria and fungi engage in sophisticated metabolic dialogs extending far beyond simple warfare. Both primary and specialized metabolites function as context-dependent signals, nutrient resources, and modulators of cellular processes that fundamentally influence the physiology, development, and evolutionary trajectory of both fungi and bacteria. Primary metabolites mediate mutualistic relati

Structural insight into the unexploited allosteric binding site of fructose 1, 6-bisphosphate aldolase from C. albicans with α-lipoic acid.

The rising incidence of drug-resistant fungal infections underscores the urgent need for innovative therapeutic strategies. However, developing selective treatments remains a formidable challenge due to the similarities between humans and fungal cells. Class II fructose 1,6-bisphosphate aldolase (FBA) represents an attractive pharmacological target for the development of antifungal agents due to its crucial role in microbial survival and its absence in human. In this work, we identified α-lipoic acid (ALA), a naturally occurring compound, as a novel inhibitor of C. albicans FBA (CaFBA). The co

Fungal Diversity and Interactions in the Nasal and Oral Cavities of Individuals with Allergic Rhinitis, Asthma and Healthy Controls.

Allergic rhinitis and asthma are common chronic airway diseases that present significant public health challenges. Previous research has shown how the nasal and oral mycobiomes influence the onset, progression and severity of these two conditions, but no study so far has directly compared those mycobiomes within the same cohort during health and disease. To address this gap, I analyzed next-generation fungal ITS sequence data from 349 participants, including individuals with allergic rhinitis, asthma, and healthy controls. The nasal and oral mycobiomes showed a great overlap in composition but

How host species and body part determine the microbial communities of five ambrosia beetle species.

The ambrosia beetles are farming insects that feed mainly on their cultivated fungi, which in some occasions are pathogens from forest and fruit trees. We used a culture-independent approach based on 16S and 18S rRNA gene metabarcoding analysis to investigate the diversity and composition of the bacterial and fungal communities associated with five ambrosia beetle species: four species native to America (Monarthrum dimidiatum, Dryocoetoides capucinus, Euwallacea discretus, Corthylus consimilis) and an introduced species (Xylosandrus morigerus). For the bacterial community, the beetle species h

Thermodynamics and kinetics of amphotericin B self-association in aqueous solution characterized in molecular detail.

Amphotericin B (AmB) is a potent but toxic drug commonly used to treat systemic mycoses. Its efficiency as a therapeutic agent depends on its ability to discriminate between mammalian and fungal cell membranes. The association of AmB monomers in an aqueous environment plays an important role in drug selectivity, as oligomers formed prior to membrane insertion - presumably dimers - are believed to act differently on fungal (ergosterol-rich) and mammalian (cholesterol-rich) membranes. In this work, we investigate the initial steps of AmB self-association by studying the structural, thermodynamic

Physical and biochemical interactions of soil fungi with asbestos fibers.

Iron, a structural component of most asbestos, is thought to play a crucial role in asbestos toxicity. Surface iron favors fiber uptake by mammalian cells and promotes the generation of free radicals with consequent DNA and lipid damage. In some cases, chemical removal of iron from asbestos fibers has reduced several cellular responses to asbestos. Since iron represents an essential element for soil microorganisms, many of them have developed mechanisms to scavenge this element from poorly soluble forms. We have investigated the ability of some soil fungi to produce iron chelators capable of e

The Laccase of Myrothecium roridum VKM F-3565: A New Look at Fungal Laccase Tolerance to Neutral and Alkaline Conditions.

Most of the currently known fungal laccases show their maximum activity under acidic environmental conditions. It is known that a decrease in the activity of a typical laccase at neutral or alkaline pH values is the result of an increase in the binding of the hydroxide anion to the T2/T3 copper center, which prevents the transfer of an electron from the T1 Cu to the trinuclear copper center. However, evolutionary pressure has resolved the existing limitations in the catalytic mechanism of laccase, allowing such enzymes to be functionally active under neutral/alkaline pH conditions, thereby giv

Climate warming, but not Spartina alterniflora invasion, enhances wetland soil HONO and NOx emissions.

Climate warming and invasive plant growth (plant invasion) may aggravate air pollution by affecting soil nitrogen (N) cycling and the emissions of reactive N gases, such as nitrous acid (HONO) and nitrogen oxides (NOx). However, little is known about the response of soil NOy (HONO + NOx) emissions and microbial functional genes to the interaction of climate warming and plant invasion. Here, we found that experimental warming (approximately 1.5 °C), but not Spartina alterniflora invasion, increased NOy emissions (0-140 ng N m-2 s-1) of treated wetland soils by 4-10 fold. Warming also decreased
