Neuropathic alterations of the myenteric plexus neurons following subacute intraperitoneal administration of salsolinol.

INTRODUCTION: Impairment of the enteric nervous system has been suggested to occur within the pathogenesis of neurodegenerative diseases. Thus, in the current study, we consider salsolinol (1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline, SAL) as a substance that can potentially induce myenteric neurodegen-eration. MATERIAL AND METHODS: Male Wistar rats were subjected to continuous intraperitoneal dosing of salsolinol (200 mg/kg in total) with osmotic mini-pumps for either two or four weeks. An equivalent group of rats served as the control. Jejunal myenteric neurons were subjected to i

Laxative effects and mechanism of action of Brazilian green propolis.

BACKGROUND: Brazilian green propolis is reported to have wide range of biological properties including antibacterial, anti-inflammatory, ant … ist) and clonidine (an α-2 adrenergic receptor agonist). We also investigated the effects of WEP on gastrointestinal transit and contractional tension of the ileum to uncover the mechanism of action of WEP. RESULTS: Treatment with WEP, but not with EEP, significantly increased the weight of stools (p<0.01 at 500 mg/kg). WEP treatment significantly restored stool frequenc …

Functional diagnostic work-up in patients with irritable bowel syndrome.

The first aim of the study was to find a cause of symptoms in patients suffering from "irritable bowel syndrome" using diagnostic tests aimed at functional disorders of lower gut. A second aim was to test, whether the presence of irritable bowel syndrome (or, synonymously, absence of classic organic disease) can be predicted by specific symptoms. 134 consecutive patients (50 +/- 16 SD years, range 17 to 88, 94 women) presenting in our gastroenterological outpatient department with abdominal pain and altered bowel habits were included. A conventional diagnostic work-up aimed at classic organic

The effects of PGF2 alpha, PGE2 and 16, 16 dimethyl PGE2 on gastric emptying and small intestinal transit in rat.

Prostaglandins are well known for their ability to stimulate contraction in gastrointestinal smooth muscle, yet very little information is available on how their activity affects propulsion in vivo. Thus, studies were undertaken to determine the effect of various prostaglandins on gastric emptying (GE) and small intestinal transit (SIT) in unanesthetized fasted rats. Rats were treated with intravenous, subcutaneous, or oral PGF2 alpha, PGE2, or 16, 16 dimethyl PGE2 at various doses, followed 1 (intravenous), 20 (subcutaneous) or 10 (oral) mins later by intragastric 51Cr oxide in black ink. ARE THERE ADVANTAGES IN DOUBLE TRANSIT RECONSTRUCTION AFTER TOTAL GASTRECTOMY IN PATIENTS WITH GASTRIC CANCER? A SYSTEMATIC REVIEW.

BACKGROUND: Curative treatment for gastric cancer involves tumor resection, followed by transit reconstruction, with Roux-en-Y being the main technique employed. To permit food transit to the duodenum, which is absent in Roux-en-Y, double transit reconstruction has been used, whose theoretical advantages seem to surpass the previous technique. AIMS: To compare the clinical evolution of gastric cancer patients who underwent total gastrectomy with Roux-en-Y and double tract reconstruction. METHODS: A systematic review was carried out on Web of Science, Scopus, EmbasE, SciELO, Virtual Health Li

Transcutaneous Electrical Acustimulation Improves Irritable Bowel Syndrome With Constipation by Accelerating Colon Transit and Reducing Rectal Sensation Using Autonomic Mechanisms.

INTRODUCTION: Slow colon transit and visceral hypersensitivity are recognized as major pathophysiological mechanisms in irritable bowel syndrome with constipation (IBS-C). However, there is a lack of therapies targeting both abdominal pain and colonic motility. This study was designed to investigate the long-term effects and possible mechanisms of transcutaneous electrical acustimulation (TEA) in patients with IBS-C. METHODS: Fifty-two patients with IBS-C were randomized into 2 groups: daily TEA for 4 weeks (n = 26) and daily sham-TEA for 4 weeks (n = 26). The number of complete spontaneous b

Baseline Predictors of Longitudinal Changes in Symptom Severity and Quality of Life in Patients With Suspected Gastroparesis.

BACKGROUND & AIMS: Whether gastric emptying tests predict longitudinal outcomes in patients with symptoms of gastroparesis is unclear. We aimed to determine whether baseline gastric emptying tests and gut motility parameters could impact longitudinal symptom(s) and quality of life (QOL) in a prospective, observational cohort study of patients with symptoms of gastroparesis. METHODS: One hundred fifty patients with gastroparesis symptoms underwent simultaneous scintigraphy (GES) and wireless motility capsule (WMC) measurement of gastric emptying and other motility parameters. Patient Assessmen

[Open surgical therapy of constipation].

This paper reviews diagnostic criteria, indications, and selection criteria for surgical therapy in chronic constipation. Special emphasis is laid on surgical therapy of pelvic outlet obstruction. Colonic resection may be considered as last surgical resort in those few cases where conventional therapeutic measures have been exhausted and an extensive diagnostic protocol based on clearly defined criteria has been followed. This may offer a justifiable therapeutic alternative only in cases where highly symptomatic and refractory colonic inertia (slow transit constipation) is confirmed, and assoc

Digestive performance and selective digesta retention in the long-nosed bandicoot, Perameles nasuta, a small omnivorous marsupial.

Bandicoots are opportunistic omnivores that feed on invertebrates, fungi and both epigeal and hypogeal plant parts. We examined the performance of the digestive tract of the long-nosed bandicoot (Perameles nasuta) in terms of intake and total digestibility, patterns of excretion of inert digesta markers, and likely sites of digesta retention, on two diets designed to mimic part of their natural plant and insect diets. On the insect diet (mealworm larvae), bandicoots virtually maintained body mass at a digestible energy intake of 511 kJ.kg-0.75.day-1 and were in strongly positive nitrogen balan

Quantitative measurement of feline colonic transit.

Colonic transit scintigraphy, a method for quantitatively evaluating the movement of the fecal stream in vivo, was employed to evaluate colonic transit in the cat. Scintigraphy was performed in duplicate in five cats and repeated four times in one cat. After instillation of an 111In marker into the cecum through a surgically implanted silicone cecostomy tube, colonic movement of the instillate was quantitated for 24 h using gamma scintigraphy. Antegrade and retrograde motion of radionuclide was observed. The cecum and ascending colon emptied rapidly, with a half-emptying time of 1.68 +/- 0.56

[Stenosis endoscope. A new instrument for the diagnosis and therapy of stenoses in the gastrointestinal tract].

A new type of endoscope combines the advantages of the narrow-calibre paediatric endoscopes and the stable-shaft regular endoscopes. This is achieved by reduction of the distal diameter from 13 mm to 9,5 mm with a conical transition between both parts. These characteristics make the instrument particularly suitable for the diagnosis of gastrointestinal stenoses (stenosis endoscope). At the same time the stenosis endoscope dilates as well as a metal olive or a rubber bougie. Thus dilatation of gastrointestinal stenoses is now possible beyond the reach of instruments used as bougie or for dilata

Tissue content of mucins in the colonic mucosa with and without fecal stream: a clinical study.

BACKGROUND: Alterations in mucin expression in the intestinal mucosa are among the main histological findings in inflammatory colonic diseases. No previous study has quantified the tissue expression of neutral and acidic mucins, including sulfomucins and sialomucins, in patients with diversion colitis. AIMS: To quantify the tissue content of neutral and acidic mucins (sulfomucins and sialomucins) in colonic mucosa with and without intestinal transit. METHODS: Forty patients undergoing elective colonoscopy at the Hospital de Clínicas - Unicamp were included. Group 1 comprised 20 patients with

The "oral-gut axis" transmission of microorganisms in colorectal cancer: Insights from Peptostreptococcus' perspective.

Colorectal cancer (CRC) is linked to gut microecological imbalances, and increasing evidence suggests that oral-gut microecosystem aberrant interactions also significantly contribute to CRC pathogenesis. The emerging "oral-gut axis" offers novel perspectives into the cross-organ microbial regulation. Notably, the oral bacterium Peptostreptococcus exhibits spatiotemporal specificity during CRC progression and may modulate gut microecology and CRC via this axis. This review examines the oral-gut microecological interplay and teases apart the multidimensional associations between Peptostreptococc

Gut Transit Defects in Drosophila Models of Monogenic Parkinson's Disease Using a Constipation Assay.

Parkinson's disease (PD) is a common neurodegenerative motor disorder and is frequently accompanied by several non-motor symptoms. Among the latter, gastrointestinal problems, including constipation, often emerge years before the manifestation of locomotion difficulties and before a diagnosis of PD is established. A recent study proposed a method to measure constipation in a PD fly model that expresses wild-type human alpha-synuclein. In this study, we examined the feasibility of this method and tested whether other genetic fly PD models equally result in constipation. Available fly lines were

Robotic Heller Myotomy for Advancements in Surgical Management of Achalasia.

Achalasia is an esophageal motility disorder. It occurs due to the destruction of nerves in the lower esophageal sphincter (LES), which leads to the failure of the LES to relax. Patients typically complain of dysphagia, chest pain, and regurgitation. They often report drinking liquids with solids intake to help propel food boluses into the stomach. The diagnosis of achalasia is typically confirmed with an esophagogram and a motility study (esophageal manometry). An esophagogram classically shows the bird beak sign with tapering in the distal esophagus. The treatment for achalasia includes both

Butterfly Wings Effect on High Resolution Manometry in A Patient with Esophageal Achalasia.

A 57-year-old man presented with dysphagia in solids and liquids deteriorating in the last months and weight loss of 3 kg. A thoracic CT revealed a limit dilatation of the lower esophagus with food residue. An upper endoscopy was performed revealing bubble content and a contraction of the Lower Esophageal Sphincter (LES). A barium esophagogram demonstrated deceleration of esophageal emptying and a bird beak sign indicative of esophageal achalasia (Figure A). High resolution esophageal manometry was performed to evaluate the subtype of achalasia. The catheter could not be intubated into the sto

[Functional abdominal symptoms in children: the role of the abdominal X-ray and measurement of colon transit time].

Functional constipation and functional non-retentive faecal incontinence are common problems in childhood; these functional defaecation disorders are diagnosed according to the Rome IV criteria. Here we describe the role of an abdominal X-ray in diagnosing functional defaecation disorders, and address the limited additional value of a plain abdominal X-ray. In limited cases it may be helpful to determine colonic transit time using an abdominal X-ray to differentiate between functional constipation and functional non-retentive faecal incontinence. The diagnostic value of abdominal X-rays in chi
