Evaluation of key taste components in Huangjin green tea based on electronic tongue technology.

A random forest algorithm combined with correlation analysis, was employed to identify key taste compounds in Huangjin green tea by using an … e and strong correlations (P < 0.05) with the response of electronic tongue; specific for theanine, glutamic acid, lysine, gallocatechin, and catechin. A taste reconstruction experiment further confirmed that these 5 compounds significantly contributed to the overall taste profile, particularly enhancing the umami flavour.

Formation and reduction of furan in pumpkin puree by precursors, antioxidants, sterilization and reheating.

The formation and mitigation of furan in pumpkin puree (PP) were studied during the complete process of producing PP. The content of furan was determined using headspace solid-phase microextraction combined by GC-MS analysis. Various PP samples added with precursors (glucose, fructose, linoleic acid, linolenic acid, β-carotene, ascorbic acid, glutamic acid, alanine, and serine) showed increased furan formation (30.81 ∼ 94.45 µg/kg) compared with the control (30.81 µg/kg), with β-carotene resulting in the formation of the largest amount of furan. The effects of antioxidants, such as caffeic aci…

Altered amino acid levels in multiply affected sibships with seizures.

Evidence of genetic factors in seizure disorders by examination of plasma amino acid concentrations in multiply affected sibships was investigated. The strategy of multiply affected sibship ascertainment was used to reduce heterogeneity as one of several potential sources of variation in quantitative amino acid levels. Our results do not support previously reported increases in plasma taurine, aspartic acid, or glutamic acid in seizure patients. However, we do find that multiply affected sibships have significantly elevated plasma concentrations of arginine and asparagine, and significantly de…

FABP5 regulates ROS-NLRP3 inflammasome in glutamate-induced retinal excitotoxic glaucomatous model.

Fatty acid binding proteins (FABPs) are a class of small molecular mass intracellular lipid chaperone proteins that bind to hydrophobic ligands, such as long-chain fatty acids. FABP5 expression was significantly upregulated in the N-methyl-d-aspartic acid (NMDA) model, the microbead-induced chronic glaucoma model, and the DBA/2J mice. Previous studies have demonstrated that FABP5 can mediate mitochondrial dysfunction and oxidative stress in ischemic neurons, but the role of FABP5 in oxidative stress and cell death in retina NMDA injury models is unclear. In this study, we found that FABP5 is s

Excitatory Amino Acid Transporters in Physiology and Disorders of the Central Nervous System.

Excitatory amino acid transporters (EAATs) encompass a class of five transporters with distinct expression in neurons and glia of the central nervous system (CNS). EAATs are mainly recognized for their role in uptake of the amino acid glutamate, the major excitatory neurotransmitter. EAATs-mediated clearance of glutamate released by neurons is vital to maintain proper glutamatergic signalling and to prevent toxic accumulation of this amino acid in the extracellular space. In addition, some EAATs also act as chloride channels or mediate the uptake of cysteine, required to produce the reactive o

Tonic glutamatergic input in the rostral ventrolateral medulla is increased in rats with chronic heart failure.

Chronic heart failure (CHF) is characterized by increased sympathetic tone. The glutamatergic input in the rostral ventrolateral medulla (RVLM), which is a key region involved in sympathetic outflow, seems not to be involved in the generation of sympathetic tone in the normal state. The aim of this study was to determine the role of the RVLM glutamate receptors in the generation of sympathetic tone in CHF. CHF was produced by coronary artery ligation. Bilateral microinjection of the glutamate receptor antagonist kynurenic acid, the N-methyl-d-aspartate (NMDA) receptor antagonist D-2-amino-5-ph

Role of aminotransferases in glutamate metabolism of human erythrocytes.

Human erythrocytes require a continual supply of glutamate to support glutathione synthesis, but are unable to transport this amino acid across their cell membrane. Consequently, erythrocytes rely on de novo glutamate biosynthesis from α-ketoglutarate and glutamine to maintain intracellular levels of glutamate. Erythrocytic glutamate biosynthesis is catalyzed by three enzymes, alanine aminotransferase (ALT), aspartate aminotransferase (AST), and glutamine aminohydrolase (GA). Although the presence of these enzymes in RBCs has been well documented, the relative contributions of each pathway hav

Clinical Chemical Studies in Aleutian Disease of Mink.

Clinical chemical determinations were carried out on blood removed by cardiac puncture from 49 mink affected with Aleutian disease and 25 normal animals and the respective differences tested for statistical significance. Blood urea nitrogen, serum total protein and globulin, thymol turbidity, glutamic oxalacetic and glutamic pyruvic transaminases and amylase were definitely elevated in the affected animals whereas serum calcium, albumin and A/G ratio were depressed. No statistically significant difference was apparent between the two groups in the comparison of inorganic phosphorus, alkaline a

Effect of Pharmacological Modulation of Activity of Metabotropic Glutamate Receptors on Their Gene Expression after Excitotoxic Damage in Hippocampal Neurons.

Microinjection of kainic acid into rat hippocampus causes excitotoxic neuronal damage predominantly in the CA3 and CA1 fields. These lesions can be significantly reduced by simultaneous administration of MPEP, a negative allosteric modulator of type 5 metabotropic glutamate receptors, and LY354740, an agonist of type 2 metabotropic glutamate receptors. The decrease in neuronal death in the hippocampus during pharmacological modulation was paralleled by adaptive changes in gene expression. In the hippocampus, gene expression of type 5 postsynaptic metabotropic glutamate receptor was close to th

Proline oxidase controls proline, glutamate, and glutamine cellular concentrations in a U87 glioblastoma cell line.

L-Proline is a multifunctional amino acid that plays an essential role in primary metabolism and physiological functions. Proline is oxidized to glutamate in the mitochondria and the FAD-containing enzyme proline oxidase (PO) catalyzes the first step in L-proline degradation pathway. Alterations in proline metabolism have been described in various human diseases, such as hyperprolinemia type I, velo-cardio-facial syndrome/Di George syndrome, schizophrenia and cancer. In particular, the mutation giving rise to the substitution Leu441Pro was identified in patients suffering of schizophrenia and

Amino acid release at the spinomedullary junction after inflammation of the TMJ region in male and female rats.

Temporomandibular joint (TMJ) disorders are painful conditions that are more prevalent in women than men. This study tested the hypothesis that acute inflammation of the TMJ region evoked sex-related changes in amino acid transmitter concentrations at the trigeminal subnucleus/upper cervical cord (Vc/C2) junction, the major terminal zone for TMJ sensory afferents. Microdialysis samples were collected in male, intact and ovariectomized (OvX) female rats after injection of mustard oil into the TMJ region (TMJ-MO) under barbiturate anesthesia. Males displayed increases in glutamate, aspartate and

Non-excitatory Amino Acids Worsen Synaptic Transmission under Ischemia-like Conditions: Hypoxia and Hypoglycemia.

INTRODUCTION: Stroke is the leading cause of adult disability and the second leading cause of death worldwide. Understanding mechanisms of neuronal damage and developing new treatments to stop its progression are key goals in brain ischemia research. We previously showed that a mixture of non-excitatory amino acids at plasma concentrations (PlasmaAA: L-alanine, glycine, L-glutamine, L-histidine, L-serine, taurine, L-threonine) worsens the deleterious effects on synaptic transmission caused by hypoxia. METHOD: We hypothesized that this amino acid combination could be especially harmful in the

Renal ammoniagenesis and acid excretion in the dogfish, Squalus acanthias.

Renal ammoniagenesis and acid excretion were investigated in normal dogfish (Squalus acanthias) and dogfish made acidotic by HCl injection (0.65 meq X kg-1). After acid loading, renal ammonia excretion doubled, rising from 0.11 to 0.25 mueq X h-1, and titratable acid output increased from 28.6 to 44.9 mueq X h-1. Trimethylamine excretion averaged 23.2 mueq X h-1 and did not change in response to the acidosis. During the first 24 h postinjection, the increase in renal acid excretion accounted for the elimination of 15% of the acid load. In vitro studies with kidney slices demonstrated that the

Biogenic carbon dots derived from the microwave carbonization of amino acid mixture: cellular biocompatible, biomolecular probes, and live cell imaging agents.

Developing highly fluorescent biogenic carbon dots (CDs) as molecular probes and cellular imaging agents with excellent cellular biocompatibility and water dispersibility can advance the scope of CDs in various biomedical applications. The present work aims to produce CDs from biological precursors such as natural amino acids. Due to their molecular recognition properties, the resultant CDs exhibit biocompatibility, polar functional groups, and selective molecule sensing. Microwave (MW)-assisted carbonization of L-tryptophan, L-glutamic acid, and their mixture led to the formation of CDs. Howe…

Proposing Bromo-Epi-Androsterone (BEA) for Stiff Person Syndrome (SPS).

SPS is characterized by progressive spasmodic muscular rigidity. SPS is thought to be an autoimmune disease with a prominent feature of antibodies against glutamic acid decarboxylase (GAD). GAD is responsible for the enzymatic conversion of glutamic acid (glutamate) into the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). Reduced GABA activity leads to increased excitability in the central nervous system, resulting in muscle rigidity and spasms characteristic of SPS. While SPS is rare, anti-GAD antibodies seen in SPS are also seen in the much more common autoimmune disease, type 1 …
