Expansion Culture of Hair Follicle Stem Cells through Uniform Aggregation in Microwell Array Devices.

Hair regeneration using hair follicle stem cells (HFSCs) and dermal papilla cells is a promising approach for the treatment of alopecia. One of the challenges faced in this approach is the quantitative expansion of HFSCs while maintaining their hair induction capacity. In this study, HFSC expansion was achieved through the formation of uniform-diameter cell aggregates that were subsequently encapsulated in Matrigel. We designed a microwell array device, wherein mouse HFSCs were seeded, allowed to form loosely packed aggregates for an hour, and then embedded in Matrigel. Quantitative analysis r

Role of hormones in the induction of iron deficiency responses in Arabidopsis roots.

In "strategy I" plants, several alterations in root physiology and morphology are induced by Fe deficiency, although the mechanisms by which low Fe levels are translated into reactions aimed at alleviating Fe shortage are largely unknown. To prove whether changes in hormone concentration or sensitivity are involved in the adaptation to suboptimal Fe availability, we tested 45 mutants of Arabidopsis defective in hormone metabolism and/or root hair formation for their ability to increase Fe(III) chelate reductase activity and to initiate the formation and enlargement of root hairs. Activity stai

Cancer fatigue and hair loss experiences among American Indian men.

Background: Little is known about the cancer symptom experiences of Indigenous populations, particularly concerning fatigue and hair loss. Hair loss can have many detrimental effects given the significant cultural importance of hair in Indigenous communities. Gaining a better understanding of the unique perspectives of American Indian male cancer survivors as they manage these two distinct symptoms is essential for improving symptom management and quality of life measures in this underserved population. Methods: A multi-pronged study explored the barriers, facilitators, and cultural constructs

The role of vitamin D receptor signaling in hair follicle health and alopecia: Current understanding and therapeutic implications.

Vitamin D receptor (VDR) signaling plays a crucial role in hair follicle biology and represents a promising therapeutic target for various forms of alopecia. This review comprehensively examines the molecular mechanisms of VDR signaling in hair follicle development, cycling, and pathology. We discuss key molecular mechanisms of VDR-dependent gene regulation through chromatin remodeling, transcriptional regulation, and recruitment of coregulatory complexes, which collectively regulate hair follicle homeostasis. Recent advances in understanding VDR genetic polymorphisms and their impact on treat

Determination of mercury and methylmercury in hair of the Czech children's population.

The developed method for mercury speciation analysis has been validated and used for the biomonitoring study of mercury species in human hair. Statistical evaluation proved the reliability of simplified determination of inorganic mercury (difference between total mercury and methylmercury). The results of the validation showed that the method is very well suitable for the determination of both species of mercury in hair for biomonitoring purposes. Non-exposed schoolchildren from three areas in the western and central part of the Czech Republic were chosen as the target group. Tenth of a microg

Interpretation of Cannabis Findings in the Hair of Very Young Children: Mission Impossible.

BACKGROUND: Hair has been suggested since the middle of the 90's to be a suitable matrix to document repetitive exposure to cannabis. Because it is possible to detect Δ9-tetrahydrocannabinol (THC), cannabinol (CBN) and cannabidiol (CBD) in cannabis smoke, the identification of the metabolite, 11-nor-Δ9-tetrahydrocannabinol carboxylic acid (THC-COOH) has been considered to allow the discrimination of active use. OBJECTIVE: Although the identification of an active compound in a child's hair shows contamination of the local environment, it is a challenge to discriminate between hair incorporatio

Identification of N-piperidinyl etonitazene in alternative keratinous matrices from a decomposing cadaver.

This short report describes research on N-piperidinyl etonitazene, also known as etonitazepipne, in keratinous matrices (hair and nails) after death related to a suspected opioid overdose. Etonitazepipne belongs to the family of benzimidazole opioids, a class of new synthetic opioids that has penetrated the illicit drug market. Analysis in the case under study showed the presence of etonitazepipne in both hair and nails, confirming that the substance accumulates in the body with repeated intake.

Sublingual minoxidil increases fibre diameter in male androgenetic alopecia: a proxy for reversal of hair follicle miniaturization.

BACKGROUND: The histological hallmark of male androgenetic alopecia (MAGA) is the transformation of terminal follicles into miniaturized secondary vellus follicles. As the volume of the dermal papilla determines the size of the hair bulb and hair fibre diameter, any treatment-induced increase in fibre diameter could be used as a proxy for reversal of hair follicle miniaturization. Although clinical trials with minoxidil topical solution in MAGA do not demonstrate increased fibre diameter, vellus-to-terminal reconversion is shown in a humanized mouse model treated with minoxidil. OBJECTIVES: T

The follicular microbiome in hair-follicle-associated inflammatory diseases: A systematic review.

BACKGROUND: Hair follicles represent distinct cutaneous microenvironments that host microbial communities. Dysbiosis has been increasingly associated with inflammatory skin and hair disorders, but available studies were limited in scope and sample size, limiting confidence in the conclusions that can be drawn. OBJECTIVES: To assess the bacterial microbiome of hair follicles across inflammatory hair-follicle-associated skin and hair diseases and identify recurrent alterations. METHODS: We searched six databases, including MEDLINE and Embase, for studies published from 2007 to August 2024. Stu

Puberty in a case with novel 17-hydroxylase mutation and the putative role of estrogen in development of pubic hair.

OBJECTIVE: 17-Hydroxylase/17,20-lyase deficiency (17OHD) results from mutations in the CYP17A1 gene, leading to failure to synthesize cortisol, adrenal androgens, and gonadal steroids. Adrenarche is a consequence of the increased production of adrenal androgens. Here, we report a case carrying novel R239Q mutation causing complete functional loss of CYP17A1, and thus absence of adrenal and gonadal sex hormone production. The patient has had unexpected pubic hair development and insufficient breast development with estrogen replacement therapy. Possible mechanisms leading to pubic hair developm

Total mercury levels in hair, toenail, and urine among women free from occupational exposure and their relations to renal tubular function.

To investigate the relations among total mercury levels in hair, toenail, and urine, together with potential effects of methylmercury intake on renal tubular function, we determined their levels, and urinary N-acetyl-beta-D-glucosaminidase activity (NAG) and alpha1-microglobulin (AMG) in 59 women free from occupational exposures, and estimated daily mercury intakes from fish and other seafood using a food frequency questionnaire. Mercury levels (mean+/-SD) in the women were 1.51+/-0.91 microg/g in hair, 0.59+/-0.32 microg/g in toenail, and 0.86+/-0.66 microg/g creatinine in urine; and, there w

Afferent innervation of outer and inner hair cells is normal in neonatally de-efferented cats.

It has been hypothesized that normal pruning of exuberant branching of afferent neurons in the developing cochlea is caused by the arrival of the olivocochlear efferent neurons and the resulting competition for synaptic sites on hair cells. This hypothesis was supported by a report that afferent innervation density on mature outer hair cells (OHCs) is elevated in animals deefferented at birth, before the olivocochlear system reaches the outer hair cell area (Pujol and Carlier [1982] Dev. Brain Res. 3:151-154). In the current study, this claim was evaluated quantitatively at the electron micros

Thrombocytosis in a patient with NUDT15 homozygous variant: clues in clinical pharmacogenetics.

A female patient diagnosed with an undifferentiated connective tissue disorder (UCTD) experienced progressive symptoms including hair loss, vomiting, diarrhoea and abdominal pain accompanied by haematological changes of anaemia, absolute neutropenia with thrombocytosis following 7 weeks of azathioprine therapy. Consequently, azathioprine was discontinued, resulted in hematologic recovery and hair regrowth. Genetic analysis revealed the presence of an NUDT15 polymorphism, indicating impaired thiopurine metabolism. Hair loss was the first symptom, above all others, indicating an early onset of a

The Immunogenetics of Alopecia areata.

Alopecia areata (AA) is an autoimmune disease that targets the hair follicles (HF) and results in non-scarring hair loss. AA results from the collapse of the HF's immune privilege due to a combination of environmental and genetic factors that either change the local HF dynamics or dysregulate the central immune tolerance. Multiple genetic studies have attempted to identify AA susceptibility genes through candidate gene approaches and genome-wide analysis. These studies were able to show an association between AA and multiple immune-related genes such as those encoding cytokines, chemokines, mo

Prehistorical Pediculus humanus capitis infestation: quantitative data and low vacuum scanning microscopy.

A pre-Columbian Peruvian scalp was examined decades ago by a researcher from the Oswaldo Cruz Foundation. Professor Olympio da Fonseca Filho described nits and adult lice attached to hair shafts and commented about the origin of head lice infestations on mankind. This same scalp was sent to our laboratory and is the subject of the present paper. Analysis showed a massive infestation with nine eggs/cm2 and an impressive number of very well preserved adult lice. The infestation age was roughly estimated as nine months before death based on the distance of nits from the hair root and the medium r

Trichothiodystrophy.

We describe a case of classic trichothiodystrophy occurring as an isolated disorder. This is the second reported case of trichothiodystrophy unassociated with systemic disorders, in contradistinction to other cases where there have been profound associated neuro-ectodermal abnormalities. The hair has shown the pathognomonic polarizing light-microscopic findings and the sulphur content has been confirmed to be reduced to approximately 50% by energy dispersive X-ray micro-analysis. Formal light-microscopic studies of hair cross-sectional shape using a Hardy microtome have confirmed the previous

Yellow mutant albinism: cytochemical, ultrastructural, and genetic characterization suggesting multiple allelism.

This report describes three sisters, including monozygotic (MZ) twins, with clinical, ultrastructural, and histochemical features typical of yellow mutant albinism; This form of albinism is clinically similar to the tyrosinase-positive type, but hair bulbs showed (1) organelles similar to red hair pheomelanosomes and (2) absence of tyrosinase activity. Classical tyrosinase-negative albinism was found in a maternal cousin of the probands. Pedigree analysis of this family suggests multiple alleles occupying a single locus.

Prevalence and Predictors of Hair Shedding among GLP-1 Receptor Agonist Users: A Cross-Sectional Study from Saudi Arabia.

INTRODUCTION: Although glucagon-like peptide-1 receptor agonists (GLP-1 RAs) demonstrate remarkable efficacy for weight management, emerging pharmacovigilance data suggest an increased hair loss risk, particularly with newer agents. This study investigated the prevalence, risk factors, and clinical impact of hair shedding in GLP-1 RA users in Saudi Arabia. METHODS: A cross-sectional survey was conducted between September 2024 and January 2025 of current and former GLP-1 RA users. Participants with preexisting hair loss were excluded from the study. Hair shedding prevalence, associated factors

Potential of Hair Follicle Stem Cell-Derived Exosomes Carrying miR-181a-5p to Promote the Telogen-to-Anagen Stage.

BACKGROUND: Hair loss is a common clinical and aesthetic concern, and existing treatments remain limited by side effects, invasiveness, or unsatisfactory long-term efficacy. Stem cell-derived exosomes have emerged as a promising cell-free strategy for tissue regeneration. However, the hair-regenerative potential of hair follicle stem cell-derived exosomes (HFSC-Exos), particularly when enriched with miR-181a-5p, remains insufficiently explored. METHODS: Commercially available rat hair follicle stem cells (HFSCs) were cultured, and exosomes were isolated from cells at passages 3-4 after transf
