H. polygyrus: B7-independence of the secondary type 2 response.

The gastrointestinal nematode parasite, Heligmosomoides polygyrus, has been used extensively in experimental studies of host immunity. The pronounced type 2 primary immune response to H. polygyrus is associated with elevated CD4+, TCR-alpha/beta + T cell IL-4 production and elevated serum IgE levels that are blocked by inhibiting CD28/ CTLA4-B7 interactions following in vivo administration of the chimeric fusion protein, CTLA4Ig. In the present study, we have examined the in vivo effects of blocking CTLA4Ig ligands on the secondary type 2 mucosal host protective immune response to this parasit

Trichomonas vaginalis Macrophage Migration Inhibitory Factor Mediates Parasite Survival during Nutrient Stress.

Trichomonas vaginalis is responsible for the most prevalent non-viral sexually transmitted disease worldwide, and yet the mechanisms used by this parasite to establish and maintain infection are poorly understood. We previously identified a T. vaginalis homologue (TvMIF) of a human cytokine, human macrophage migration inhibitory factor (huMIF). TvMIF mimics huMIF's role in increasing cell growth and inhibiting apoptosis in human host cells. To interrogate a role of TvMIF in parasite survival during infection, we asked whether overexpression of TvMIF (TvMIF-OE) confers an advantage to the paras

Immunogenic and cross-reactive carbohydrate determinants in ticks and other arthropods.

Ticks are parasites that feed on the blood of humans and animals, using it as their sole source of food. Due to their lifestyle, ticks at each life stage transmit various tick-borne pathogens while feeding on their hosts. Unlike mosquitoes - another important group of pathogen-transmitting arthropods - ticks can feed on the same host for prolonged periods, which underscores the importance of tick mechanisms that inhibit or reduce the host's defence mechanisms and, consequently, allow the transmission of pathogens. This unique tick-host-pathogen triangle requires the involvement of various tick

Recovering host DNA from their invertebrate parasites: a minimally invasive genetic sampling tool for the critically endangered flapper skate.

Obtaining host DNA from the blood meals of their invertebrate parasites (invertebrate-derived DNA, iDNA) has proven to be a powerful tool in terrestrial ecology for detecting vertebrates to answer questions relating to host community diversity. However, the use of iDNA in the marine environment is limited. Here, we assessed the efficacy of sampling parasites from the critically endangered flapper skate (Dipturus intermedius) to determine whether iDNA could (i) identify the host species and (ii) obtain host haplotype sequences. In total, 34 copepods (Trebius caudatus specimens, n = 30; Caligus

An Integrated Approach to Studying the Wheat-Diuraphis noxia Interaction for Identifying Aphid Biotypes and Assessing Plant Responses.

In the last four decades, Diuraphis noxia, commonly known as Russian wheat aphid (RWA), has been identified as one of the economically important pests, capable of causing huge wheat yield losses. In South Africa (SA) and North America (NA), extensive studies on RWA virulence on different wheat genotypes have classified the RWA into five SA- and eight NA-biotypes. However, standardized methods covering the wheat infestation processes from collecting RWA populations in the field, verifying the RWA biotypes, and using biotypes to infest wheat plants for biochemical, physiological, and molecular b

Parasite-Induced Changes in the Nervous System of the Shore Crab Hemigrapsus sanguineus.

Trematodes are a class of parasitic flatworms with complex life cycles involving multiple hosts. They can influence the physiology and behavior of their intermediate hosts to improve the efficiency of transmission to definitive hosts, including by increasing host mortality. However, the mechanisms of their effect on the crustacean nervous system leading to neuronal dysregulation and its consequences have not been sufficiently elucidated. This study investigated the effects of infection by metacercariae of the trematode Cercaria fluviocinguli on the serotonin (5-HT) immunoreactivity, inflammati

Selective Packaging of miR-22 and miR-29 Orthologues into Extracellular Vesicles of the Porcine Whipworm Trichuris Suis.

Helminths modulate host immunity, and in this respect, the porcine whipworm Trichuris suis has been explored clinically as an immunotherapy for inflammatory diseases. T. suis secretes a complex mixture of excretory/secretory products (ESP), including extracellular vesicles (EVs), nanosized membranous particles with bioactive cargoes of lipids, proteins and nucleic acids that are implicated in intercellular communication within organisms. Here, we characterise the EV protein and miRNA cargo of T. suis to identify potential roles in host-parasite communication. In contrast to EVs from clade III/

Darwin's solution: Honeybees survive mite vectors and viruses through natural selection.

Complex host-vector-virus interfaces can impose serious health challenges. Western honeybees have experienced high colony losses globally, mainly driven by the host-shifted, virus-vectoring ectoparasitic mites Varroa destructor and Tropilaelaps mercedesae. Host populations can survive mite infestations through natural selection, offering a long-term strategy for colony health. However, host-vector-virus coevolution requires local adaptations of this triad, which is poorly understood. We propose harnessing natural selection through a global approach focused on standardized monitoring of colony

TgSEC14-LTP1 is a vacuole-secreted SEC14/CRAL-TRIO-like protein required for host DAG flux and PC homeostasis in T. gondii.

Intracellular apicomplexan parasites depend on host-derived lipids to expand the parasitophorous vacuole (PV) and replicate. Toxoplasma gondii secretes numerous dense granule proteins (GRAs) for PV remodeling, but PV lumen factors that selectively bind host lipids and shape host-to-parasite lipid flux remain poorly defined. Here, we identify TgSEC14-LTP1, a previously uncharacterized GRA with a divergent SEC14/CRAL-TRIO domain. Structural and genetic analyses show that TgSEC14-LTP1 is secreted into the PV lumen and is essential for parasite replication. Lipidomic profiling, protein-lipid pulld

The Role of Innate Lymphoid Cells in the Pathogenesis of Hepatic Echinococcosis: A Literature Review.

Hepatic echinococcosis (HE), a major zoonotic disease, remains endemic in pastoral regions of Western China, where its incidence is closely linked to geographical, livestock and sanitary conditions. The disease presents a significant clinical challenge due to the complexity of host-parasite interactions and a high recurrence rate. To date, the underlying pathogenesis and effective prevention and control strategies remain inadequately studied. Particularly, it is still unclear how innate lymphoid cells (ILCs) affect disease progression and therapeutic intervention. This review focuses on ILCs a…

Testing the "niche centroid hypothesis" in flea-mammal networks from the palearctic: species' centrality across their environmental niches.

A species' centrality in a network is expected to decrease with distance from the centroid of its environmental niche. We investigated the variation of a species' centrality in flea-mammal networks across the space of its niche for 30 flea and 21 small mammal species occurring in 10-46 networks from different localities in the Palearctic. We characterized the environmental niche of a flea or a host species using the ellipsoid envelope model, based on a species' occurrence points and climatic, soil, and vegetation data. We found significant relationships between centrality in a network and the

The stage-specific regulation and role of root-knot nematode SWEET genes.

The root-knot nematode Meloidogyne incognita is a globally significant plant parasite that causes substantial crop losses. While pre-parasitic juveniles rely on innate energy reserves, later life stages acquire nutrients from host plants through specialized feeding structures. SWEET (Sugars Will Eventually be Exported Transporter) genes exhibit a conserved sugar transporting ability across all kingdoms of life, yet their function in plant-parasitic nematodes remains underexplored. Here, we functionally characterise the SWEET gene family in M. incognita, revealing their critical and stage-speci

From biosynthesis to scavenging: evolutionary rewiring of PUFA and oxylipin biosynthesis in parasitic nematodes.

Polyunsaturated fatty acids (PUFAs) and their oxygenated metabolites (oxylipins) are mediators in regulating membrane function, larval development, and host immune response. In this review, we present that free-living nematodes maintain a relatively complete repertoire of enzymes for PUFA and oxylipin biosynthesis, while parasitic lineages exhibit extensive gene loss and plant-parasitic nematodes show expansion in some enzymes. Meanwhile, parasitic nematodes compensate by scavenging lipids from hosts through the recruitment of various lipid transport proteins. Thus, parasites have lost some en

Beech Leaf Disease: A New Threat to Beech Forest Ecosystems.

Beech leaf disease (BLD) has emerged as a significant threat to beech forests in North America, causing the widespread decline of American beech while posing a potential threat to other beech species worldwide. BLD has drawn considerable attention due to its rapid spread and the visible decline it causes in beech trees. Since its first detection, the disease has rapidly expanded its geographic range, underscoring the urgent need for a comprehensive understanding of its etiology and epidemiology. This disease is characterized by interveinal dark banding of the leaf, bud abortion, and progressiv

No Simple Fluke: Schistosomiasis as an Emerging Regulator of Haematopoiesis.

Schistosomiasis, or bilharzia, is a highly prevalent water-borne helminth infection that accounts for an estimated 1.5-1.7 billion disability-adjusted life years lost annually. Pathology driven by adult worms and eggs is accompanied by robust immunomodulation in the human host. Protective responses may promote worm elimination, limit bystander tissue injury caused by parasite migration and contain eggs within granulomatous lesions. However, evidence also suggests that schistosomes can actively or indirectly influence haematopoiesis, thereby reshaping downstream mature immune cell responses. Th

Parasite infection intensity and infection site, rather than host species, shape the histopathological effects of trematode infections in anuran tadpoles.

Anuran amphibians play essential ecological roles and are considered important bioindicators. This is especially true during the larval stage, when tadpoles exhibit high sensitivity to parasitic infections, including trematode metacercariae. These infections can profoundly affect the physiology and morphology of juvenile hosts, potentially compromising tissues involved in osmoregulation, locomotion, and development. We analysed naturally infected tadpoles, performing parasitological screening and histopathological evaluation. Through field collection, euthanasia, histological processing, and m

Unravelling malaria latency: parasite intrinsic and environmental factors influencing dormant liver stages.

Hypnozoites - dormant Plasmodium parasites in the liver - can cause relapse infections and form a major obstacle to malaria eradication. The mechanisms controlling dormancy remain poorly understood, but hypnozoite formation and reactivation is likely regulated by a combination of parasite intrinsic factors and external stimuli. We reviewed current knowledge of Plasmodium dormancy and drew parallels with dormancy in other parasites and life-cycle stages. Epigenetic, post-transcriptional, or post-translational regulation probably jointly control hypnozoite dormancy at the intrinsic level. Additi

Protein families secreted by nematodes to modulate host immunity.

Parasitic nematodes release a wide variety of immunomodulatory proteins, which allow them to escape the host's immune-mediated killing or ejection mechanisms. This immunomodulation is mediated by nematode excretory/secretory (E/S) products, which contain multiple families of immunomodulatory proteins. Many of these families are conserved across different parasitic nematodes, while others are apparently unique to specific species. While some E/S products interact with host proteins, others have evolved to target host lipids, glycans, and metabolites. In this review, we will focus on three famil
