Effect of Helicobacter pylori-mediated inflammation on nonsteroidal anti-inflammatory drugs-induced gastric mucosal injury.

Helicobacter pylori (H. pylori) infection and nonsteroidal anti-inflammatory drugs (NSAIDs) are two major causes of gastric ulceration, but relation between H. pylori infection and use of these drugs in gastric mucosal injury is controversial. Neutrophils have been implicated in the pathogenesis of gastric mucosal damage induced by H. pylori or NSAIDs. H. pylori itself and H. pylori extract induce neutrophil activation, such as superoxide production, expression of adhesion molecule (CD11b/CD18) and transendothelial migration, capillary plugging. Several clinical and experimental studies demons

Immunostimulatory DNA mediates inhibition of eosinophilic inflammation and airway hyperreactivity independent of natural killer cells in vivo.

BACKGROUND: Immunostimulatory DNA sequences (ISS) inhibit eosinophilic inflammation and airway hyperreactivity in mouse models of asthma. In vitro ISS activate natural killer (NK) cells to secrete IFN-gamma, and this cytokine is hypothesized to contribute to the antiallergic effect of ISS in vivo. OBJECTIVE: We investigated whether ISS activation of NK cells is important in mediating the reduction in airway hyperreactivity and the antieosinophilic effect of ISS in vivo. METHODS: We assessed whether ISS modulated the development of eosinophilic airway inflammation and airway hyperreactivity t

Early-onset multifocal inflammation in the transforming growth factor beta 1-null mouse is lymphocyte mediated.

Transforming growth factor beta 1 (TGF beta 1)-null mice die fro complications due to an early-onset multifocal inflammatory disorder. We show here that cardiac cells are hyperproliferative and that intercellular adhesion molecule 1 (ICAM-1) is elevated. To determine which phenotypes are primarily caused by a deficiency in TGF beta 1 from those that are secondary to inflammation, we applied immunosuppressive therapy and genetic combination with the severe combined immunodeficiency (SCID) mutation to inhibit the inflammatory response. Treatment with antibodies to the leukocyte function-associat

Listeriolysin negative mutants of Listeria monocytogenes specifically stimulate T-lymphocytes mediating protection and granulomatous inflammation.

The ability of several listeriolysin O negative mutants of the virulent EGD strain of Listeria monocytogenes to activate specific T cell responses in vitro and in vivo was determined. A T cell line and a derived clone specific for Listeria monocytogenes, strain EGD, which are able to adoptively transfer protection and granuloma formation were examined. Specificity testing showed no difference between listeriolysin positive and negative strains to induce proliferation of the T cell lines and clones. Similar results were obtained when we examined T cell mediated granuloma formation in the livers

Dual Function of Ccr5 during Langat Virus Encephalitis: Reduction in Neutrophil-Mediated Central Nervous System Inflammation and Increase in T Cell-Mediated Viral Clearance.

Tick-borne encephalitis virus (TBEV) is a vector-transmitted flavivirus that causes potentially fatal neurologic infection. There are thousands of cases reported annually, and despite the availability of an effective vaccine, the incidence of TBEV is increasing worldwide. Importantly, up to 30% of affected individuals develop long-term neurologic sequelae. We investigated the role of chemokine receptor Ccr5 in a mouse model of TBEV infection using the naturally attenuated tick-borne flavivirus Langat virus (LGTV). Ccr5-deficient mice presented with an increase in viral replication within the C

MALT1 Knockdown Alleviates Multiorgan Injury and Inflammation Through Inhibiting TAK1/NF-κB Signaling Pathway-Mediated T-Helper 1 and 17 Differentiations in LPS-induced Inflammation.

Mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1) is a vital regulator of inflammation and innate immune response. The aim of this study was to evaluate the potential of MALT1 knockdown for attenuating lipopolysaccharide (LPS)-induced inflammation. C57BL/6 mice received tail vein injection of knockdown control or Malt1 shRNA, followed by intraperitoneal injection of LPS two weeks later. After 24 h of LPS injection, the mice were euthanized for further analysis. The peripheral naïve CD4+ T cells of mice were isolated, treated with Malt1 overexpression vectors or Malt1 s

"Rogue" neutrophil-subset [DEspR+CD11b+/CD66b+] immunotype is an actionable therapeutic target for neutrophilic inflammation-mediated tissue injury - studies in human, macaque and rat LPS-inflammation models.

BACKGROUND AND OBJECTIVE: The correlation (Rs > 0.7) of neutrophils expressing the dual endothelin1/signal peptide receptor (DEspR+CD11b+/CD66b+) with severity of hypoxemia (SF-ratio) and multi-organ failure (SOFA-score) in patients with acute respiratory distress syndrome (ARDS) suggest the hypothesis that the DEspR+ neutrophil-subset is an actionable therapeutic target in ARDS. To test this hypothesis, we conducted in vivo studies to validate DEspR+ neutrophil-subset as therapeutic target and test efficacy of DEspR-inhibition in acute neutrophilic hyperinflammation models. METHODS: We perfo

Reversal of established CD4+ type 2 T helper-mediated allergic airway inflammation and eosinophilia by therapeutic treatment with DNA vaccines limits progression towards chronic inflammation and remodelling.

Immunostimulatory DNA-based vaccines can prevent the induction of CD4(+) type 2 T helper (Th2) cell-mediated airway inflammation in experimental models, when administered before or at the time of allergen exposure. Here we demonstrate their efficacy in limiting the progression of an established response to chronic pulmonary inflammation and airway remodelling on subsequent allergen challenge. Mice exhibiting Th2-mediated airway inflammation induced following sensitization and challenge with group 1 allergen derived from Dermatophagoides pteronyssinus group species (Der p 1), a major allergen o

Integrated multi-omics reveals that Notoginsenoside R1 mitigates inflammation mediated by NLRP3 and metabolic disruption in muscle contusion.

Muscle contusion triggers sterile inflammation and metabolic stress, in which macrophage-mediated NLRP3 inflammasome activation plays a critical role in secondary tissue injury and delayed repair. Panax notoginseng (SanQi) has long been used for traumatic injury and inflammation-related disorders, but its active constituents and immunopharmacological mechanisms in muscle contusion remain incompletely defined. Here, we investigated the protective effects of SanQi in a rat gastrocnemius contusion model and explored its underlying mechanisms using integrated transcriptomic, metabolomic, zebrafish

Linkage-dependent effects of procyanidin dimers on TNFα-mediated intestinal epithelial barrier dysfunction, redox homeostasis and inflammation.

The gastrointestinal (GI) tract is generally exposed to high concentrations of dietary components, positioning diet as a key determinant of intestinal inflammation and barrier function, implicated in the onset of chronic systemic diseases. Procyanidins (PCAs), oligomeric flavan-3-ols abundant in plant-derived foods, exert GI health benefits despite poor systemic bioavailability. We previously found that galloylated PCA dimers protected intestinal epithelial integrity more effectively than non-galloylated dimers. However, the role of linkage type remains unclear. This study investigated the com

Transcription Factor SPI1 Mediates Chronic Neuropathic Pain by Activating NLRC4 Transcription to Drive Microglial Activation and Inflammation.

Microglial polarization plays a key role in the process of chronic neuropathic pain (CNP). This study aims to investigate the molecular mechanism by which the transcription factor SPI1 mediates microglial polarization and participates in CNP by regulating NOD-like receptor C4 (NLRC4) expression. The GSE124272 dataset (blood samples of 8 intervertebral disc degeneration patients and 8 controls) was obtained from the GEO database, and the differentially expressed genes (DEGs) were screened. The intersection of DEGs with neuropathic pain related genes in GeneCards was taken. Support vector machin

Longitudinal association between insulin resistance and depressive symptoms in older adults: the mediating role of inflammation in the ELSA cohort.

BACKGROUND: Insulin resistance (IR) has been implicated in depression, yet the dynamic interplay between concurrent changes in IR and depressive symptoms over time, and whether systemic inflammation mediates this relationship, remains unclear. METHODS: Data were obtained from 3,404 older adults (aged ≥50 years) in the English Longitudinal Study of Ageing (ELSA) across Waves 2, 4, and 6. IR was assessed using the estimated glucose disposal rate (eGDR). Depressive symptoms were measured with the CES-D scale (0-8). C-reactive protein (CRP) was log-transformed as the mediator. Linear mixed models

Triptolide alleviates pathological neovascularisation in oxygen-induced retinopathy via inhibiting the inflammation mediated by NF-κB pathway.

BACKGROUND: Retinopathy of prematurity (ROP), an oxygen-induced retinopathy (OIR), triggers a series of vascular lesions and inflammatory responses and results in visual impairment or even blindness. Triptolide (TP) possesses many pharmacological properties, including immunosuppressive and anti-tumour effects. However, the effects of TP on ROP and its underlying mechanisms remain unclear. PURPOSE: To investigate whether TP could inhibit the progression of OIR and to elucidate its underlying mechanisms. METHODS: The 7-day-old mice (P7) were kept in a 75% hyperoxia incubator for 5 days to indu

Exosome-Mediated Rewiring of Oxidative Stress-Inflammation-ECM Remodeling Axis Mitigates UVB-Triggered Skin Photoaging.

Photoaging is a chronic and multifactorial form of skin aging primarily induced by ultraviolet (UV) radiation. It is characterized by oxidative stress, DNA damage, chronic inflammation, pigmentary alterations, and extracellular matrix (ECM) degradation, which ultimately lead to wrinkle formation, loss of elasticity, and uneven pigmentation. Exosomes, nanosized extracellular vesicles that mediate intercellular communication, have attracted growing attention in photoaging research. Emerging evidence suggests that exosomes may both contribute to photoaging and promote tissue repair by modulating

5-Bromocytidine protects against hyperuricemia-induced renal injury by suppressing TLR4/NF-κB-mediated inflammation.

Hyperuricemia (HUA), a prevalent metabolic disorder, is an independent risk factor for chronic kidney disease (CKD). Elevated serum uric acid (UA) level leads to renal UA deposition, causing kidney inflammation, impairing renal function, and accelerating HUA progression. In this study, an inflammation model in RAW264.7 cells induced by monosodium urate (MSU) was established to screen for compounds resistant to MSU-induced inflammation. Fortunately, 5-bromocytidine, a nucleic acid analogue, demonstrated potent anti-inflammatory activity in MSU-treated RAW264.7 cells and exhibited excellent ther
