Insect-Specific Chimeric Viruses Potentiated Antiviral Responses and Inhibited Pathogenic Alphavirus Growth in Mosquito Cells.

Most alphaviruses are transmitted by mosquito vectors and infect a wide range of vertebrate hosts, with a few exceptions. Eilat virus (EILV) in this genus is characterized by a host range restricted to mosquitoes. Its chimeric viruses have been developed as safe and effective vaccine candidates and diagnostic tools. Here, we investigated the interactions between these insect-specific viruses (ISVs) and mosquito cells, unveiling their potential roles in determining vector competence and arbovirus transmission. By RNA sequencing, we found that these ISVs profoundly modified host cell gene expres

Population structure of G. pallidipes tsetse flies in Northwestern Kenya - Towards effective vector control.

BACKGROUND: Tsetse flies are vectors of trypanosome parasites responsible for Human and Animal African Trypanosomiasis (HAT and AAT), diseases that greatly affect health and economic growth across sub-Saharan Africa. Vector control, one of the possible strategies to tackle these diseases, is most effective on genetically isolated tsetse populations. While the population genetic structure of Glossina pallidipes tsetse flies has been well studied in southern Kenya, little is known about populations in northwestern Kenya. This study investigated the presence, genetic diversity, and population str

Efficacy of volatile pyrethroid spatial repellents to reduce exposure to mosquito and sand fly vectors and human infection risk: a systematic review and meta-analysis.

BACKGROUND: Volatile pyrethroid spatial repellents (VPSRs) have emerged as promising complementary tools for vector control. This systematic review and meta-analysis evaluated the protective efficacy (PE) of VPSRs against mosquito- and sand-fly-borne human infections, and against vector numbers in traps. METHODS: Clinical infection and entomological studies that evaluated VPSRs against human infection with Plasmodium, arboviruses (Dengue and Zika), and Leishmania, and against their respective vectors, were systematically reviewed. Random-effects meta-analyses estimated pooled PE values and ex

Patterns of microbiome composition in tsetse fly Glossina palpalis palpalis during vector control using Tiny Targets in Campo, South Cameroon.

Novel vector control tools against African trypanosomiases require a deep understanding of the factors driving tsetse vector fitness or population resilience in their ecosystems. Following evidence of microbiota-mediated host fitness or traits shaping, including insecticide resistance in arthropod populations, we undertook a comparative study of the microbiota in wild-caught tsetse flies during vector control with deltamethrin-impregnated traps called Tiny Targets. The bacterial microbiome composition of tsetse flies collected before and after 6, 12, and 18 months of vector control were charac

Standardising visual control devices for Tsetse: East and Central African Savannah species Glossina swynnertoni, Glossina morsitans centralis and Glossina pallidipes.

BACKGROUND: This study focused on the savannah tsetse species Glossina swynnertoni and G. morsitans centralis, both efficient vectors of human and animal trypanosomiasis in, respectively, East and Central Africa. The aim was to develop long-lasting, practical and cost-effective visually attractive devices that induce the strongest landing responses in these two species for use as insecticide-impregnated tools in population suppression. METHODS AND FINDINGS: Trials were conducted in different seasons and years in Tanzania (G. swynnertoni) and in Angola and the Democratic Republic of the Congo

An Operational Framework for Insecticide Resistance Management Planning.

Arthropod vectors transmit organisms that cause many emerging and reemerging diseases, and their control is reliant mainly on the use of chemical insecticides. Only a few classes of insecticides are available for public health use, and the increased spread of insecticide resistance is a major threat to sustainable disease control. The primary strategy for mitigating the detrimental effects of insecticide resistance is the development of an insecticide resistance management plan. However, few examples exist to show how to implement such plans programmatically. We describe the formulation and im

Bioactivity and laundering resistance of five commercially available, factory-treated permethrin-impregnated fabrics for the prevention of mosquito-borne diseases: the need for a standardized testing and licensing procedure.

Personal protective measures against hematophagous vectors constitute the first line of defense against arthropod-borne diseases. In this regard, a major advance has been the development of residual insecticides that can be impregnated into clothing. Currently, however, information on specific treatment procedures, initial insecticide concentrations, arthropod toxicity, residual activity, and laundering resistance is either fragmentary or non-existent, and no World Health Organization Pesticides Evaluation Scheme or other guidelines exist for the standardized testing and licensing of insectici

Long-lasting transition toward sustainable elimination of desert malaria under irrigation development.

In arid areas, people living in the proximity of irrigation infrastructure are potentially exposed to a higher risk of malaria due to changes in ecohydrological conditions that lead to increased vector abundance. However, irrigation provides a pathway to economic prosperity that over longer time scales is expected to counteract these negative effects. A better understanding of this transition between increased malaria risk and regional elimination, in particular whether it is slow or abrupt, is relevant to sustainable development and disease management. By relying on space as a surrogate for s

Effect of large-scale installation of deltamethrin-impregnated screens and curtains in Bam, a major focus of anthroponotic cutaneous leishmaniasis in Iran.

INTRODUCTION: Installation of deltamethrin-impregnated screens and curtains was assessed as a preventive measure against transmission of anthroponotic cutaneous leishmaniasis (ACL) by Phlebotomus sergenti in Bam, a well-known focus of ACL in Iran with a population of nearly 100,000. METHODS: This was a quasi-experiment based on official data from an ACL control program in which one section of the city with about one-quarter of the population received the intervention. In the analysis, the rest of the city was used as control. Data covered June 2007 through October 2010. RESULTS: Comparison o

Spinosad toxicity to Simulium spp. larvae and associated aquatic biota in a coffee-growing region of Veracruz State, Mexico.

Spinosad is a naturally derived insecticide that has shown potential as a mosquito larvicide. To determine the activity of spinosad against blackflies, late-instar larvae from a community comprising Simulium triittatum (63.6%) and seven other species, including three known vectors of onchocerciasis in Mexico (S. metallicum, S. ochraceum, and S. callidum), were subjected to concentration-mortality laboratory bioassays following World Health Organization guidelines. Cephalic capsule measurements confirmed the relatively homogeneous distribution of experimental larvae. The 50% lethal concentratio

Natural vectors of dirofilariasis in rural and urban areas of the Tuscan region, central Italy.

Entomological investigations by means of dog- and human-baited traps were carried out in summers 2000-2002 in urban and rural areas of the Tuscan region in central Italy. The aim of the study was to define the mosquito species involved in the transmission of Dirofilaria nematodes and to assess the risk that their presence might represent for animal and human health. Nocturnal fieldwork on host-seeking activity and feeding preferences was followed by microscopic identification of the mosquito species attracted and by molecular identification of Dirofilaria parasites in mosquitoes. In total, 3,6

Sampling techniques for adult Afrotropical malaria vectors and their reliability in the estimation of entomological inoculation rate.

Various entomological indicators and sampling techniques are used to monitor and evaluate the impact of many vector control interventions. A number of methods have been used in sampling mosquitoes for the purpose of estimating the entomological inoculation rate (EIR) and each is subject to some bias or shortcomings. It was the aim of this paper to critically evaluate the most common mosquito sampling techniques in relation to their reliability in the estimation of EIR. The techniques include man-landing, light trap, light trap/bednet combination and odour-baited traps. Although man-landing tec

[Epidemiological surveillance of Chagas disease].

Chagas disease still constitutes an important medical problem in affected countries. In some, the extent of the disease is still unknown and control programs have not been implemented. In others the disease has been reduced due to regular control programs and other economic and social factors. Epidemiological surveillance with community participation to guard against disease transmission is now the basic challenge. Applied research and in-depth reformulation of health systems are required to establish efficient and sustainable Chagas disease surveillance programs, considering low density of pe

The control of anopheline mosquitos by the spraying of Deltamethrin on raffia curtains used in miners' huts in areas endemic for malaria.

The residual potential of an aqueous solution of Deltamethrin (FW 25 mg i.a./m) was evaluated on raffia curtains. These are sheets of synthetic material used in the construction of huts to house miners. Experiments were conducted during 420 days and the curtains were always rolled up in the daytime and unrolled in late afternoon. Data analyzed by logarithmic regression indicated that raffia treated with Deltamethrin had higher mortality indices than that covered with DDT. The residual capacity of Deltamethrin on raffia was high. The mortality percentage was above 85% after 360 days and dropped

An historical review of malaria, kala-azar and filariasis in Bangladesh in relation to the Flood Action Plan.

This paper reviews the evidence of a link between flood control and vector-borne disease in Bengal/Bangladesh. Malaria is historically associated with reduced flooding and embankment construction in the flood plains of Bengal. The land west and south of the Jamuna river was highly malarious in 1916 but is not so today. The lands east of the Jamuna now have a higher, though still small, risk. The reduction in health risk can be attributed to the intensification of land use and human population density. Although there are many mosquito species, the abundance of the former malaria vector appears

Community participation in the control of tsetse flies. Large scale trials using the pyramid trap in the Congo.

An experiment of Glossina palpalis control was carried out by rural communities in 55 villages of the Niari river sleeping sickness focus (Bouenza region, Republic of the Congo). It was based on the use of a new trap, not requiring insecticide impregnation, in which the captured Glossina are preserved. The results show that this simple, cheap trap is an effective method of control, resulting in a considerable decrease in the tsetse population and is easily operated by the villagers. Screening surveys, using the indirect immunofluorescent test, conducted regularly in this focus, showed a decrea

Variations in the ecology of Glossina spp. with special reference to Nigerian populations of Glossina tachinoides.

During the last decade, an increasing number of instances have been noted of tsetse flies occupying unusual, and often man-made, habitats. Recently, some aberrant populations have become important, either because they have drawn attention to new aspects of trypanosomiasis transmission or because they have necessitated modifications of established tsetse control methods. From observations made on atypical tsetse populations in Nigeria, the author has classified the various types of intra-specific behaviour patterns observed and offers an interpretation. At least 1 species, Glossina tachinoides,
