Exploring the Effects of Antioxidants on αSynuclein-Induced Motor Deficits in Drosophila Larvae.

Parkinson's disease (PD) is marked by dopaminergic neuron loss, motor deficits, and oxidative stress. Misexpression of mutant human α-synuclein in Drosophila larvae induces PD-like motor impairments, making it a powerful and accessible model for studying neurodegeneration. In this Course-Based Undergraduate Research Experience (CURE), we used a novel Ring Maze assay to quantify larval locomotion following expression of A53T and E46K α-synuclein variants. Both induced robust crawling deficits. Notably, antioxidant treatment with vitamin C or açaí extract significantly improved motor performance…

Cyflumetofen induces hepatic steatosis and disrupts lipid metabolism in zebrafish larvae.

Cyflumetofen is a highly effective acaricide, and both it and its metabolites are often present in the environment as pollutants. Therefore, the safety of cyflumetofen for non-target organisms requires further attention. This study used zebrafish larvae to evaluate the effects of cyflumetofen on liver development. After 72-h exposure, cyflumetofen specifically manifested as significantly reduced liver area and histological damage (cellular vacuolization, nuclear loss) in the 2.0 and 4.0 μg/mL exposure groups. Concurrently, Oil Red O, Nile Red, and BODIPY 493/503 staining all showed that cyflum…

Dose-dependent effects of strontium on the calcification physiology of Ruditapes philippinarum larvae.

Strontium (Sr) is a calcium-analog that can accumulate in coastal environments from natural and anthropogenic sources, posing emerging risks to ecosystems and human health. By readily entering calcium-based biomineralization pathways, Sr can disrupt the calcification physiology of marine calcifiers, yet the dose-dependent effects remain poorly constrained. Here, we assessed the impact of Sr across a large concentration gradient (0-80 mg L-1) on the early larval development of the Manila clam (Ruditapes philippinarum). Complete absence of Sr prevented initiation of prodissoconch I (PD I) format…

HarmGR13 mediates myo-inositol taste perception in Helicoverpa armigera larvae.

Myo-inositol, a sugar alcohol produced by most plants, serves as a nutrient and feeding stimulant for many phytophagous insects. Inositol-sensitive taste sensilla have been characterized in many Lepidoptera larvae, but their molecular bases remain unclear. In this study, we determined the gustatory receptors (GRs) for myo-inositol in larva of Helicoverpa armigera, a worldwide crop pest. First, electrophysiological analyses revealed that medial sensilla styloconica strongly responded to myo-inositol and ribose, with weaker responses to xylose, and one GRN inside sensillum may mediate the respon…

Toxic effects of prolonged propofol exposure on cardiac development in zebrafish larvae.

BACKGROUND: Propofol, commonly used as an intravenous anesthetic during pregnancy, can easily penetrate the placental barrier, potentially affecting fetal heart development. This study aims to investigate propofol's impact on developing zebrafish heart structure and function, and identify potential drug targets. METHODS: Zebrafish embryos were exposed to different concentrations of propofol (0.5, 1, and 5 mg/L) to observe changes in zebrafish larval heart structure and function (heart rate). In vitro cell experiments were conducted to assess the effects of propofol at different concentrations…

Effects of the Novel Triazole Fungicide Ipfentrifluconazole on Different Endpoints in Zebrafish Larvae.

The potential hazards of triazole fungicides to non-target organisms necessitate environmental risk assessment. This study, therefore, focused on characterizing the differential toxicity of the enantiomers of Ipfentrifluconazole (IFZ), a new triazole fungicide, in zebrafish larvae using a multi-endpoint approach. Acute toxicity tests determined the LC50 values of 1.709 mg/L for rac-IFZ, 1.531 mg/L for (+)-IFZ, and 1.809 mg/L for (-)-IFZ, indicating a higher toxicity of the (+)-enantiomer. To avoid overt mortality while revealing organ-level effects, we chose a concentration of approximately 20…

Assaying Nociception Behaviors in Drosophila Larvae During Parasitoid Wasp Attacks.

Nociception in fruit fly (Drosophila melanogaster) larvae is characterized by a stereotyped escape behavior. When a larva encounters a noxious (potentially harmful) stimulus, it responds by curving its body into a c-shape and rolling in a corkscrew-like manner around its long-body axis. This rolling behavior may serve to quickly remove the larva from the source of the noxious stimulus, and is particularly adaptive to escape from a common natural predator of fruit fly larvae: parasitoid wasps (Leptopilina boulardi). L. boulardi completes its life cycle by using fruit fly larvae as hosts for its…

Environmental enrichment influences the relationship between lateralization and neophobia in zebrafish larvae.

Neophobia, the aversion to novelty, is a conserved survival trait that influences individuals’ decision-making. In many species, juveniles s … for investigating the phenotypic plasticity of neophobia. Previous studies showed that two-week-old larvae are more inclined to explore novel stimuli, while older larvae exhibit neophobic responses, indicating an age-dependent shift in exploratory behaviour. Evaluation of developmental and behavioral toxicity of Resmetirom in zebrafish larvae (Danio rerio).

Resmetirom, a promising oral THR-β agonist for treating metabolic dysfunction-associated steatohepatitis (MASH), has been studied extensively for its therapeutic efficacy, yet its effects on embryonic development remain largely unknown. This study aimed to evaluate the potential embryotoxicity of Resmetirom and investigate its impact on embryonic growth and development. We employed the zebrafish (Danio rerio) as the model organisms, and exposed embryos to varying concentrations of Resmetirom. Key developmental endpoints, including hatchability, mortality, malformation rates, developmental timi

Porphyromonas gingivalis induces neuroinflammation in a gingipain-dependent manner in zebrafish larvae.

Porphyromonas gingivalis (Pg), a keystone pathogen in the development of periodontitis, has been implicated in neurodegenerative diseases such as Alzheimer's disease (AD). The gingipains, Pg crucial virulence factors, have been detected in the brains of AD patients and suggested to be involved in neuroinflammation, which leads to neuronal death. However, the mechanisms underlying Pg brain invasion and its inflammatory effect remain poorly understood. In this study, using a zebrafish larval model, we investigated the gingipain-dependent effects of systemic and local hindbrain infection with Pg,…

Zebrafish larvae locomotion bioassay: application on nanofractionated Naja nigricollis venom.

Snakebite envenoming remains a major global health issue, particularly in underserved regions. To better understand venom composition and toxic effects, crude snake venoms can be fractionated into distinct toxin groups using high-throughput nanofractionation analytics. These fractions can then be assessed using venomics to identify specific toxins, in conjunction with bioassays to evaluate their bioactivity. Traditional in vivo testing in rodents is limited by legal and ethical concerns, the high number of fractions, and the small quantities of toxins in each. In this study, zebrafish larvae w…

Zebrafish larvae as a model for investigating dual effects of fluoride on bone development.

Long-term excessive ingestion of fluoride is a severe public health threat globally. Skeletal fluorosis, a significant manifestation of prolonged fluoride exposure, is characterized by aberrant bone structure and alterations in bone function. However, there is currently a shortage of an efficient, fast, and easy-to-operate biological model for application in the field of fluorosis research. Zebrafish larvae, with human - like skeletal traits, high reproduction, rapid development, and transparency, are commonly used in bone disease studies. This study evaluates the potential of zebrafish larvae…

Insights into the developmental and cardiovascular toxicity of bixafen using zebrafish embryos and larvae.

Bixafen (BIX), a member of the succinate dehydrogenase inhibitor (SDHI) class of fungicides, has seen a surge in interest due to its expanding market presence and positive development outlook. However, there is a growing concern about its potential harm to aquatic life, largely due to its resistance to breaking down in the environment. In this study, we thoroughly examined the toxicological impact of BIX on zebrafish as a model organism. Our results revealed that BIX significantly hindered the development of zebrafish embryos, leading to increased mortality, hatching failures, and oxidative st

An Image Processing Tool for Automated Quantification of Bacterial Burdens in Zebrafish Larvae.

Zebrafish larvae are used to model the pathogenesis of multiple bacteria. This transparent model offers the unique advantage of allowing quantification of fluorescent bacterial burdens (fluorescent pixel counts [FPC]) in vivo by facile microscopical methods, replacing enumeration of bacteria using time-intensive plating of lysates on bacteriological media. Accurate FPC measurements require laborious manual image processing to mark the outside borders of the animals so as to delineate the bacteria inside the animals from those in the culture medium that they are in. Here, we have developed an a…
