Emergence of Resistance to Macrolides and Rifampin in Clinical Isolates of Rhodococcus equi from Foals in Central Kentucky, 1995 to 2017.

The objective of this study was to determine the prevalence of Rhodococcus equi strains resistant to macrolides and rifampin over time in clinical samples from foals submitted to diagnostic laboratories in central Kentucky. We performed a retrospective observational study of all clinical samples from foals that were submitted to veterinary diagnostic laboratories in Kentucky between January 1995 and December 2017. Samples were included if the R. equi bacterium was cultured and tested for in vitro susceptibility to erythromycin or rifampin. In vitro susceptibility testing to erythromycin was av…

Anti-inflammatory effect of prophylactic macrolides on children with chronic lung disease: a protocol for a double-blinded randomised controlled trial.

INTRODUCTION: Recent studies suggest that the high mortality rate of respiratory viral infections is a result of an overactive neutrophilic inflammatory response. Macrolides have anti-inflammatory properties, including the ability to downregulate the inflammatory cascade, attenuate excessive cytokine production in viral infections, and may reduce virus-related exacerbations. In this study, we will test the hypothesis that prophylactic macrolides will reduce the severity of respiratory viral illness in children with chronic lung disease by preventing the full activation of the inflammatory casc…

In-vitro activities of tetracyclines, macrolides, fluoroquinolones and clindamycin against Mycoplasma hominis and Ureaplasma ssp. isolated in Germany over 20 years.

Antimicrobial resistance in genital mycoplasmas is increasing and shows global variation. We determined the susceptibilities of 469 mycoplam … s (70.3% and 35.2% susceptibility, respectively). Clarithromycin and josamycin were the most potent macrolides (MIC₉₀ of 0.5 mg/L) against ureaplasmas. Erythromycin had the lowest activity (MIC₉₀ of 8 mg/L) against ureaplasmas like clindamycin which was the most potent agent against M. hominis. Cross-resistance was found between tetracyclines (53-93%), macro …

Efficient Production of 14-Membered Macrolides by the Endophytic Fungus Colletotrichum gloeosporioides GLL-7 and Their Insecticidal Activity against Diaphorina citri.

The citrus psyllid (Diaphorina citri) vectors Huanglongbing. Overuse of insecticides causes resistance and pollution, necessitating safer alternatives. Bioactivity-guided fractionation of the fungus Colletotrichum gloeosporioides GLL-7 yielded two macrolides: Sch725674 and Gliomasolide H (GLH). Sch725674, a major metabolite produced at 0.035 g per gram of culture medium, is easily obtained via methanol trituration. GLH is a novel macrolide; its structure was elucidated by spectroscopy and confirmed by chemical synthesis. GLH exhibited contact toxicity (24 h LC50 = 0.129 mg/mL) against D. citri…

A sequential esterification-ring closing metathesis-Nozaki-Hiyama-Kishi strategy for constructing a natural product-like library of tetrahydrofuran-containing macrolides.

Polyketide-like macrolides (pMLs) represent a privileged class of compounds with a high incidence of bioactivity, however their structural complexity challenges their synthesis and more general study. Here we report the synthesis of a library of tetrahydrofuran-containing pMLs underpinned by a robust and convergent build/couple/pair/couple synthetic approach. The library comprises 170 pMLs originating from 17 building blocks, 10 of which were synthesized using a proline-catalyzed α-chlorination aldol reaction. Northern and southern hemisphere building blocks were coupled using either an oxidat…

Modeling the Effects of Vaccination and Treatment With Third-Generation Macrolides or Oxytetracyclines on Persistence and Impact of Contagious Bovine Pleuropneumonia.

This research assesses the impact of contagious bovine pleuropneumonia (CBPP) vaccination, treatment, and combined vaccination and treatment at the herd and population level in heterogeneous, stochastic, state-transition models of CBPP transmission. Results from published trials with oxytetracycline or third-generation macrolides (tulathromycin and gamithromycin) were used to inform parameters for antibiotic treatment. Societies have evolved and the levels of movement control responsible for the previous success of vaccination programs are no longer possible. Current vaccines, when applied in …

Monitoring of Antimicrobial Resistance to Aminoglycosides and Macrolides in Campylobacter coli and Campylobacter jejuni From Healthy Livestock in Spain (2002-2018).

Antimicrobial resistance (AMR) in Campylobacter spp. (Campylobacter coli and Campylobacter jejuni) is a concern due to its importance in public health, particularly when it involves aminoglycosides and macrolides, drugs of choice for treatment of human cases. Co-resistance to these two antimicrobial classes involves transfer of genetic elements and/or acquisition of mutations in different genetic loci, which can in turn spread through vertical or horizontal gene transfer (HGT) phenomena, with each route having different potential implications. This study aimed at evaluating the association bet…

Characterization of 28 unknown impurities in 16-membered macrolides by liquid chromatography coupled with ion trap/time-of-flight mass spectrometry.

In terms of risk assessment, the study of the impurity profile is important to ensure the safety and effectiveness of drugs in clinical application. Sixteen-membered macrolides are produced by microbial fermentation, and many closely related substances in the product make the components and impurities complicated. In this study, methods were developed to separate and identify the impurities in three representative 16-membered macrolides (josamycin, midecamycin and meleumycin) using a high-performance liquid chromatography coupled to high-resolution ion trap/time-of-flight mass spectrometry (IT…

Synthesis and Antibacterial Activity of New N-Alkylammonium and Carbonate-Triazole Derivatives within Desosamine of 14- and 15-Membered Lactone Macrolides.

Desosamines of azithromycin (AZM) and clarithromycin (CLA) were modified by N-alkylation or nucleophilic substitution at the carbonyl/CuAAC sequence. Biological studies revealed a higher antibacterial potency of quaternary N-alkylammonium bromides of CLA as compared to AZM. SAR studies of CLA salts, including biological, conformation and molecular-docking analysis, enriched by physicochemical parameters, showed the importance of less bulky and unsaturated substituent for an efficient docking mode at the ribosomal tunnel and good antibacterial potency against clinical and standard Streptococcus

A review of non-cystic fibrosis bronchiectasis in children with a focus on the role of long-term treatment with macrolides.

Bronchiectasis is a rare chronic airway disease arising from several respiratory and systemic diseases. The grade of evidence for specific treatment of childhood bronchiectasis unrelated to cystic fibrosis (CF) is low with very few randomized controlled trials. Treatment has been based mainly on evidence from studies in adults with non-cystic fibrosis bronchiectasis and patients with cystic fibrosis. Recently, long-term treatment with macrolides has been proposed. These molecules offer the advantage of anti-inflammatory and immunomodulatory properties in addition to their antibacterial propert…

Miuramides A and B, Trisoxazole Macrolides from a Mycale sp. Marine Sponge That Induce a Protrusion Phenotype in Cultured Mammalian Cells.

Morphology-guided cell-based screening of the extract of a Mycale sp. marine sponge led to the isolation of two trisoxazole macrolides, miuramides A (1) and B (2), which induced characteristic morphological changes in 3Y1 cells. The structure of 1 including absolute configuration was elucidated by a combination of the analysis of spectroscopic data, derivatization, and degradation. Both compounds exhibit potent cytotoxicity against 3Y1 cells.

The role of newer macrolides in the treatment of community-acquired respiratory tract infection. A review of experimental and clinical data.

The macrolide class of antibiotics is well established and often recommended for use in the treatment of community-acquired respiratory tract infection (RTI). The newer agents clarithromycin and azithromycin are frequently prescribed as first- or second-line therapy, and have been considered as superior to erythromycin in microbiological activity and clinical efficacy. In-vitro data show that clarithromycin and azithromycin have good activity (MIC < or = 0.5 microg/ml) against certain RTI pathogens. However the activity of both compounds is intrinsically low against Haemophilus influenzae whil

Frequency of macrolide resistance-associated mutations in Mycoplasma genitalium: a multicenter molecular analysis based in Brazil.

Mycoplasma genitalium is an emerging sexually transmitted pathogen with increasing antimicrobial resistance worldwide. In Brazil, molecular testing for M. genitalium and resistance-associated mutations is not routinely available, and national data remain scarce. We analyzed 118 M. genitalium-positive urethral and vaginal swab samples collected between 2018 and 2023 from sites across all five Brazilian geographic regions. PCR amplification and sequencing of 23S rRNA, parC, and gyrA identified macrolide and fluoroquinolone resistance-associated mutations. Among 79 samples successfully sequenced
