Maternal residential exposure to specific agricultural pesticide active ingredients and birth defects in a 2003-2005 North Carolina birth cohort.

BACKGROUND: Previously we observed elevated odds ratios (ORs) for total pesticide exposure and 10 birth defects: three congenital heart defects and structural defects affecting the gastrointestinal, genitourinary and musculoskeletal systems. This analysis examines association of those defects with exposure to seven commonly applied pesticide active ingredients. METHODS: Cases were live-born singleton infants from the North Carolina Birth Defects Monitoring Program linked to birth records for 2003-2005; noncases served as controls (total n = 304,906). Pesticide active ingredient exposure was a

Low doses of prenatal ethanol exposure and maternal separation alter HPA axis function and ethanol consumption in adult male rats.

Adverse maternal behaviors during pregnancy and unfavorable postnatal experiences during development are associated with an increased risk of developing psychiatric disorders, as well as, a vulnerability to alcohol addiction in adulthood. Here, we examined the effects of combined ethanol exposure during late pregnancy and postnatal maternal separation (MS) on HPA responsiveness, anxiety behavior and preference for alcohol consumption in adult male rats. Animals exposed to both conditions revealed a decrease in blood levels of allopregnanolone accompanied by increased anxiety behavior. In addit

Maternal di-(2-ethylhexyl) phthalate exposure during pregnancy causes fetal growth restriction in a stage-specific but gender-independent manner.

Di (2-ethylhexyl) phthalate (DEHP) is male developmental toxicant that impairs testis development with reduced anogenital distance. The present study aimed to investigate whether maternal DEHP exposure during pregnancy causes intrauterine growth restriction (IUGR) in a gender-specific manner and to identify the critical window of DEHP-induced fetal IUGR. Pregnant mice were administered with DEHP (0, 50 or 200mg/kg) by gavage. Fetal IUGR was observed not only in males but also in females when litters were exposed to DEHP on gestational day (GD)0-GD17. Interestingly, fetal weight and crown-rump

Maternal exposure to criteria air pollutants and congenital heart defects in offspring: results from the national birth defects prevention study.

BACKGROUND: Epidemiologic literature suggests that exposure to air pollutants is associated with fetal development. OBJECTIVES: We investigated maternal exposures to air pollutants during weeks 2-8 of pregnancy and their associations with congenital heart defects. METHODS: Mothers from the National Birth Defects Prevention Study, a nine-state case-control study, were assigned 1-week and 7-week averages of daily maximum concentrations of carbon monoxide, nitrogen dioxide, ozone, and sulfur dioxide and 24-hr measurements of fine and coarse particulate matter using the closest air monitor withi…

Neurodevelopmental effects of low-level prenatal mercury exposure from maternal fish consumption in a Mediterranean cohort: study rationale and design.

BACKGROUND: Mercury is a neurotoxic environmental pollutant. However, the literature on the neurodevelopmental effect of low-level prenatal mercury exposure from maternal fish intake is inconsistent. We assessed the association between prenatal mercury exposure and infant neurodevelopment in coastal areas of 4 Mediterranean countries. METHODS: This was a prospective cohort study that planned to enroll approximately 1700 mother-infant pairs. Pregnant women and their newborn children were recruited in selected hospitals of the study areas. Biological samples, including maternal hair and cord bl

Fetal exposure to a maternal low-protein diet is associated with altered left ventricular pressure response to ischaemia-reperfusion injury.

Rats exposed to protein restriction as fetuses develop hypertension as adults. Hypertension increases the risk of myocardial ischaemia and infarction. We investigated whether rats exposed to low-protein diets in utero are more susceptible to myocardial ischaemia-reperfusion (IR) injury. Pregnant Wistar rats were fed control or low-protein (MLP) diets throughout pregnancy. At 4 and 8 weeks postnatal age systolic blood pressure was determined in the offspring using tail-cuff plethysmography. At 6 months of age, rats were treated with saline or N-acetylcysteine (NAC) for 48 h. Rapidly excised hea

Maternal pollen allergy may be more important than birch pollen exposure during pregnancy for atopic airway disease in the child.

In 1993 extremely high levels of birch pollen were recorded in Stockholm, Sweden. We investigated the effects of this exposure on sensitization and development of atopic airway disease in children. The aim of this study was to assess the influence of maternal birch sensitization and symptoms of pollen allergy, as well as exposure to birch pollen during pregnancy, on sensitization and development of atopic airway disease in children. A total of 387 children with atopic heredity (70% had atopic mothers) and born in Stockholm 1993 or 1994 were investigated at age 4.5-5 yr. The children were exami

Fetal exposure to low protein maternal diet alters the susceptibility of young adult rats to sulfur dioxide-induced lung injury.

The maternal diet is an important determinant of glutathione-related metabolism in rats. Glutathione (GSH) may play a major role in the detoxification of sulfur dioxide (SO2) within the lungs. The effects of fetal exposure to a low protein maternal diet upon later susceptibility to pulmonary injury induced by chronic SO2 exposure were evaluated in young adult rats. Pregnant rats were fed purified diets containing 180 g casein/kg (control diet) or 120, 90 or 60 g casein/kg (experimental diets). After parturition, all dams were fed a standard non-purified diet (189 g protein/kg diet). The pups t

Early-Life exposure to environmental pollutants and endocrine disruptors: emerging risks and mechanistic insights in maternal, fetal, and child health.

Exposure to environmental pollutants and endocrine-disrupting chemicals (EDCs) in early life is a critical public health issue because it can alter developmental programming that impacts susceptibility to disease throughout life. The prenatal and early postnatal periods are times of vulnerability when exposure to environmental stressors may impact endocrine signalling, placental function, epigenetic regulation, and metabolic homeostasis. There is increasing evidence that exposure to bisphenols, phthalates, per- and polyfluoroalkyl substances (PFAS), pesticides, heavy metals, and air pollutants

Maternal exposure to intimate partner violence and its association with diarrhoea prevalence and treatment among children under five in Nepal.

BACKGROUND: Diarrhoeal disease remains a critical public health concern, and a leading cause of death among children under the age of five. Increasingly, studies are also showing a relationship between child diarrhoea and maternal exposure to intimate partner violence (IPV). This study assessed the relationship between maternal experience of different forms of IPV and child diarrhoea prevalence and treatment-seeking in Nepal. METHODS: Study participants included children under five years whose mothers were asked about IPV exposure in the 2022 Nepal Demographic and Health Survey (n = 1,959). Th…

Long-term developmental alterations in intestinal microbiota, metabolism, and T-cell homeostasis in offspring rats following maternal tetrabromobisphenol A exposure.

Tetrabromobisphenol A (TBBPA), a widely used brominated flame retardant, has been associated with adverse health effects. An exploratory NHANES analysis of urinary bisphenol A (BPA) was performed to provide general population-level context for bisphenol-related metabolic and inflammatory associations because urinary TBBPA measurements were unavailable. We investigated whether maternal TBBPA exposure (170 mg·kg⁻¹·day⁻¹) was associated with developmental alterations in offspring intestinal homeostasis. Maternal TBBPA exposure was associated with colonic inflammatory features, increased pro-infla

Maternal prenatal co-exposure to air pollution and psychological distress shapes the neonatal gut: microbiota-mediated pathways to early neurodevelopment.

Early life gut microbiota function as biological sensors for maternal prenatal exposure and play a crucial role in infant neurodevelopment. During pregnancy, air pollution and psychological distress are regarded as general and specific external exposures, respectively; however, the joint influence of these two domains on shaping early life gut microbiota remains unexplored. In this study, 309 mother-infant pairs were recruited from the obstetrics departments of two tertiary hospitals. We collected data on maternal prenatal air pollution exposure and psychological distress, obtained meconium sa

Maternal occupational/environmental exposure to selected organic solvents and small-for-gestational-age births: a systematic review and meta-analysis.

BACKGROUND: Although studies have suggested an association between organic solvent exposure and small-for-gestational age (SGA), conclusions remain inconsistent due to variations in sample size and study design, with insufficient epidemiological evidence. OBJECTIVE: We investigated the association between organic solvent exposure and SGA. METHODS: Following the PRISMA guidelines, searched PubMed, MEDLINE, Embase, and Science Direct databases for cohort and case-control studies. Exposure was classified based on job-exposure matrices (JEMs), expert assessment, or self-reported history. SGA was

Investigating the association between COVID-19 exposure and fetal and maternal health before public health vaccinations: A retrospective cohort study.

OBJECTIVES: Pregnant women and neonates face a higher risk of adverse outcomes from infectious diseases, such as COVID-19. COVID-19 vaccines were developed rapidly, but we do not fully understand how the disease affected pregnancies and births before these vaccines were available. This study investigated how COVID-19 impact on pregnant women and their neonates before vaccines became widely available. The aim was to investigate the association between maternal COVID-19 infection and pregnancy and neonatal outcomes. STUDY DESIGN: This retrospective cohort study included pregnant women and their

Maternal di-n-butyl phthalate exposure promotes renal fibrosis in IUGR offspring rats via HBB downregulation and Nrf2 pathway activation.

Di-n-butyl phthalate (DBP) is an environmental endocrine disruptor with developmental nephrotoxicity, but its long-term effects on renal fibrotic remodeling in offspring remain incompletely understood. In this study, a maternal DBP exposure model was established in pregnant Sprague-Dawley rats (750 mg/kg/day, GD14-GD18), and adult male offspring were examined at postnatal day 42. DBP-exposed offspring showed intrauterine growth restriction, reduced relative kidney weight, renal collagen deposition, and epithelial-mesenchymal transition (EMT)-related molecular changes. Transcriptomic analysis i

Maternal di(2-ethylhexyl) phthalate (DEHP) exposure alters offspring gut microbiota and the colonic transcriptome in a sex-specific manner.

Maternal exposure to the plasticizer DEHP during gestation and lactation programs lasting sex-specific alterations in offspring intestinal health. This study investigated the transgenerational effects of perinatal DEHP exposure on offspring intestinal health using a C57BL/6 mouse model. Dams were exposed to 0, 40, or 400 ppm DEHP via their diet during gestation and lactation. We analyzed the growth phenotypes, intestinal histology, gut microbiota, and colonic transcriptomics of the offspring. We found that perinatal DEHP exposure did not affect the growth of dams or offspring but induced signi…
