Melatonin, melanogenesis, and hypoxic stress in the newt, Triturus carnifex.

Groups of 6 specimens each of the newt Triturus carnifex were treated with melatonin to see if the hormone inhibited melanogenesis in the Kupffer cells of the liver (melanomacrophages), a process markedly stimulated by hypoxia. A dose of 500 microg/g in 27% ethanol, injected intraperitoneally, induced loss of consciousness and tetany of all the skeletal muscles, which on the contrary appeared relaxed in animals pre-anesthetised by immersion in chlorbutol at 0.2%. Anesthetised specimens injected with melatonin showed a significantly lower increase in hepatic pigmentation after acute hypoxia, a …

[Melatonin/circadian rhythm. Is there a feedback between epiphysis and hypophysis?].

BACKGROUND: The study evaluates the circadian rhythm of melatonin in relation to sex and age and identified contemporary alterations in the secreton of some hypophyseal hormones, suggesting that melatonin may exert a modulatory action on the latter. METHODS: The melatonin metabolite (6-hydroxymelatonin sulfate) was assayed in urine samples from 48 subjects of both sexes aged between 25 and 60 years old using the RIA method described by Arendt, modified for the ise of iodine markers. A blood sample was taken from the same subjects to assay hypophyseal hormones. RESULTS: Melatonin secretion do…

Melatonin is rhythmic in newborn seals exposed to continuous light.

We have investigated the effect of continuous light and darkness on plasma levels of melatonin in relation to the extremely large and active pineal gland typically found in newborn seals. Plasma levels of melatonin in captive newborn harp (Phoca groenlandica) and hooded seals (Cystophora cristata) were generally extremely high, with peak concentrations ranging from 0.8 ng/ml to 62.3 ng/ml. Moreover, plasma melatonin showed a similar, pronounced rhythmicity, both outdoors under natural light conditions (hooded seal only) and indoors under either 30 h of continuous light (490 lux) or 30 h of dar…

Effect of brefeldin A on melatonin secretion of chick pineal cells.

Melatonin is secreted from the pineal gland in a circadian manner. It is well established that the synthesis of melatonin shows a diurnal rhythm reflecting a daily change in serotonin N-acetyltransferase (NAT) activity, and the overall secretion of melatonin requires a cellular release process, which is poorly understood. To investigate the possible involvement of Golgi-derived vesicles in the release, we examined the effect of brefeldin A (BFA), a reversible inhibitor of Golgi-mediated secretion, on melatonin secretion of cultured chick pineal cells. We show here that treatment with BFA compl…

Human melatonin suppression by light: a case for scotopic efficiency.

Human adult males were exposed to combinations of two illuminances and two spectral power distributions over the course of four nighttime sessions. A dose-dependent response of acute melatonin suppression to light was found, but photopic (cone-based) illuminance did not adequately predict suppression. When melatonin suppression was plotted against scotopic (rod-based) illuminance, the data formed a nearly monotonic function, implicating rods, or a rod-dominated mechanism, in the human melatonin regulation system. The results do not, however, rule out mechanisms other than rods, including novel…

[Melatonin: is there a rationale for the clinical use of this hormone in sleep therapy?].

Melatonin, the hormone of the pineal gland, is under discussion as multipotent hormone for different indications. The question whether there is a therapeutical benefit for all these indications could not be evaluated, because more clinical studies are needed. Based on the data of basic research as well as first clinical experience, the use of melatonin to treat sleep disorders caused by a deficit of melatonin might be appropriate. Other indications, derived from animal studies where the results often could not be applied to humans, have still to be proved. Given the fact that melatonin is not …

The therapeutic potential of melatonin in migraines and other headache types.

A large number of individuals suffer from migraine headaches. Several theories attempt to explain migraine etiology. One such theory holds that since environmental stimuli are well known to trigger migraine headaches, the pineal gland may be involved in migraine etiology. Specifically, a pineal gland irregularity may be the physical origin of migraine headaches, with subsequent physiological changes being secondary. Research has found the pineal hormone melatonin is low in migraine patients. Additionally, several studies found administering melatonin to migraine sufferers relieved pain and dec…

Dual effects of melatonin on barbiturate-induced narcosis in rats.

Melatonin affects the circadian sleep/wake cycle, but it is not clear whether it may influence drug-induced narcosis. Sodium thiopenthal was administered intraperitoneally into male rats pre-treated with melatonin (0.05, 0.5, 5 and 50 mg/kg). Melatonin pre-treatment affected in a dual manner barbiturate narcosis, however, no dose-effect correlation was found. In particular, low doses reduced the latency to and prolonged the duration of barbiturate narcosis. In contrast, the highest dose of melatonin (50 mg/kg) caused a paradoxical increase in the latency and produced a sustained reduction of t…

Melatonin and epileptic seizures in patients with acute intermittent porphyria.

The objective of this study was to assess melatonin production in patients with acute intermittent porphyria (AIP), with and without known epileptic seizures, as a guide to whether melatonin may have anti-convulsive or pro-convulsive effects in AIP. Melatonin concentration in urine, sampled over eight hours on two consecutive nights, was analysed in eight AIP patients with epileptic seizures and in 14 AIP relatives without epilepsy. The AIP patients with epileptic seizures had a significantly lower urinary excretion of melatonin, compared with their AIP relatives without epilepsy, which may in…

Facilitation of benzodiazepine discontinuation by melatonin: a new clinical approach.

BACKGROUND: Benzodiazepines are the most frequently used drug for the treatment of insomnia. Prolonged use of benzodiazepine therapy is not recommended. However, many patients, particularly older patients, have difficulties discontinuing therapy. Melatonin, a hormone that is produced at night by the pineal gland, promotes normal sleep in humans and augments sleep induction by benzodiazepine therapy. OBJECTIVE: To assess whether the administration of melatonin could facilitate the discontinuation of benzodiazepine therapy in patients with insomnia. METHODS: Thirty-four subjects receiving benz…

Effects of melatonin on the oxidative stress induced by thyrotoxicosis in rats.

High concentrations of thyroid hormones can affect the metabolism of oxygen in aerobic conditions and stimulate free radicals generation in mitochondria. Reactive oxygen species are toxic to biomembranes and lead to peroxidation of lipids unless they are not removed by free radical scavengers. A few years ago melatonin (MEL), a pineal hormone, was described as a scavenger of free radicals, especially of hydroxyl and peroxyl radicals. The goal of the study was to examine the parameters of oxidative stress [the levels of conjugated dienes (CD), malondialdehyde (MDA) and Schiff bases (SB)] in mal…

Use of melatonin to treat sleep disorders in tuberous sclerosis.

The results of a therapeutic trial of the use of melatonin in patients with tuberous sclerosis complex who also have severe sleep problems are reported. We used a randomized double-blind placebo-controlled crossover design. Seven patients with confirmed diagnoses of tuberous sclerosis and significant sleep disorder were recruited. We employed three outcome measures: total sleep time, time to sleep onset, and number of awakenings. Patients treated with melatonin had a small but clinically significant improvement in total sleep time (mean improvement 0.55 hours, P<0.05). They also tended to have…

Lack of calmodulin antagonism of melatonin in T-lymphocyte activation.

Despite various reported effects of the pineal hormone melatonin on the immune system, its mechanism of action on immune cells is still unknown. Since melatonin has been suggested as a physiological antagonist to calmodulin in certain cell types, we investigated effects of melatonin on calmodulin-dependent IL-2 production and proliferation of activated T-lymphocytes. It was found, however, that, in contrast to the calmodulin antagonists trifluoperazine and W7, melatonin neither inhibited the IL-2 production of activated lymphoblastoid Jurkat T-cells nor decreased the mitogen response of periph…

Comparative aspects of the pineal/melatonin system of poikilothermic vertebrates.

The pineal gland of poikilothermic vertebrates originates as an evagination from the diencephalic roof between the habenular and the posterior commissures, and associates with a parapineal organ to form the so-called pineal complex. The pinealocytes may be photosensitive, secretory or intermediate cells between both. Melatonin, the indoleamine secreted by the pineal, exhibits a circadian secretory rhythm that conveys environmental information to the organism. The peak melatonin secretion occurs during the night, although there are a few examples of an increase in indoleamine secretion during t…

The effect of light on plasma melatonin levels in premature infants.

Recent studies have shown that full term neonates actively secrete melatonin and that light deprivation during the first 72 h of life significantly increases plasma melatonin levels. In order to evaluate pineal gland activity and responsiveness to light in premature infants, we measured plasma melatonin levels in 23 healthy infants, 33-36 weeks of gestation, during their first week of life. Nine infants (Group A) remained under constant illumination conditions for 48 hour prior to melatonin measurements. Fourteen infants (Group B) were exposed for the same time period to an artificial alternat…

Inhibition of melatonin secretion onset by low levels of illumination.

Melatonin is a hormone released during darkness under the control of the hypothalamic circadian pacemaker. It has been shown that melatonin is suppressed by light as a function of intensity, with low levels of illumination producing small effects and more intense light greater, but not complete inhibition. The studies which lead to these conclusions administered light subsequent to the secretion pattern being well established. Light as low as 250 lux administered during the normal onset of secretion can reduce melatonin to below detectable levels. The onset of melatonin secretion was delayed f…

Effect of melatonin on active transport of serotonin into blood platelets.

Previous research has demonstrated that the circadian rhythm of melatonin negatively correlates with that of the sensitivity of platelet serotonin uptake sites. Moreover, serotonin has been found to be a competitive inhibitor of melatonin binding to platelets. The goal of the present study was to investigate whether melatonin directly affects platelet serotonin transport. Blood samples from 12 healthy subjects were assayed for both serotonin transport into platelets. Active transport of serotonin (at a concentration of 10(-6) M) into blood platelets was measured in the presence of melatonin at…
