Network oscillatory dynamics accompany cerebral bioenergetic defence in hypoxia.

A network physiology framework investigated how coordinated interactions among multiple organ systems collectively support the preservation of cerebral bioenergetic function and better distinguish adaptive from maladaptive responses to hypoxia. Twelve healthy males were passively exposed to 6 h of normoxia (21% O2) and hypoxia (12% O2) in a randomised, single-blind, crossover design. Venous blood was assayed for oxidative-nitrosative stress (OXNOS, spectroscopy/chemiluminescence) and neurovascular unit (hs-ELISA) biomarkers. Global cerebral delivery of O2 and glucose were determined by duplex

Poly(ADP-Ribose) Polymerases Inhibition in Cardiovascular Oncology: Protective Mechanisms Versus Clinical Cardiotoxicity.

Poly(ADP-ribose) polymerases (PARPs) are central components of the cellular DNA damage response, coordinating DNA repair, chromatin remodeling, and transcriptional regulation. Beyond their established role in oncology, accumulating evidence identifies PARP-particularly PARP-1-as a key modulator of cardiovascular stress responses. In cardiovascular disease states characterized by oxidative and nitrosative stress, PARP activation exerts context-dependent effects: while moderate activation supports genomic integrity and cell survival, excessive or sustained PARP activity leads to depletion of nic

The FaAP2-11 Transcription Factor Putatively Regulates Cell Wall Remodeling and Stress-Acclimation Responses Under Drought and Salinity Stress.

APETALA2/ethylene-responsive factor (AP2/ERF) transcription factors are key regula-tors of plant responses to abiotic stress, integrating environmental and hormonal signals into coordinated transcriptional programs. RNA-seq analysis of strawberry (Fragaria × ananassa) leaves and roots subjected to drought and salinity identified 58 differentially expressed FaAP2/ERF genes. Promoter analysis revealed cis-regulatory elements associated with abscisic acid, jasmonate, ethylene, salicylic acid, and drought-responsive signaling. Among these genes, FaAP2-11 was selected for functional characterizatio

Asymmetric Structure-Driven Functional Synergy: A Multistimuli-Activated Janus Nanomotor for Self-Amplifying NO Cascade Therapy.

Nitric oxide (NO)-based cancer therapy is fundamentally limited by poor intratumoral penetration of exogenous donors and their inability to sustain localized relevant flux. Here, a multistimuli-activated Janus nanomotor was developed to interconnect nonenzymatic and enzymatic NO-generation pathways, enabling in situ self-cascading NO production. The nanomotor comprises an AuPt3Cu nanozyme asymmetrically coated with a disulfide-bridged mesoporous silica shell (2sMSN) loaded with L-arginine, allowing autonomous propulsion under O2, NO, and NIR irradiation to achieve deep tumor penetration. Upon

Neuroprotective Effects of Senecio brasiliensis Extract Against Chlorpyrifos-Induced Neurotoxicity and Oxidative Stress in Drosophila melanogaster.

Chlorpyrifos (CP) is an organophosphate insecticide that induces neurotoxicity through acetylcholinesterase (AChE) inhibition and redox imbalance, raising environmental and occupational health concerns. Plant-derived compounds from Senecio brasiliensis, native to the Pampa biome, offer multitarget therapeutic potential. This study evaluated the protective effects of the hydroalcoholic extract of S. brasiliensis (HESB) against CP-induced toxicity in Drosophila melanogaster (Harwich strain). Adult females (3-5 days old) were exposed for 48 h to control (1% sucrose), CP (0.25 ppm), HESB (1 mg/mL)

Ruxolitinib attenuates diethylnitrosamine-induced liver injury in mice: involvement of TLR4/MYD88/NF-κB and IL-6/JAK/STAT pathways.

The liver is a main target organ for xenobiotic-induced acute injury, with such toxicity frequently mediated through the oxidative stress generation and the subsequent activation of inflammatory signaling pathways like TLR4/MYD88/NF-κB and IL-6/JAK/STAT pathways. The current study investigated the potential protective effect of ruxolitinib (RUX) on liver injury induced by diethylnitrosamine (DEN). Mice were administered RUX prophylactically as a single dose orally, then DEN (150 mg/kg, i.p.) after an hour. Interestingly, RUX significantly alleviated liver injury induced by DEN as evidenced by

Novel SGLT2 inhibitors with endothelial-protective potential: a combined computational and experimental approach.

OBJECTIVES: Sodium-glucose co-transporter 2 (SGLT2) inhibitors are a class of anti-diabetic drugs with cardiovascular benefits independent of glycemic control, partly via endothelial protection. Identifying novel inhibitors with both high affinity and vascular-protective effects remains a priority. METHODS: Structure-based high-throughput virtual screening (HTVS) of 14 million ligands was performed using Schrödinger Glide. Compounds with better docking scores than empagliflozin (-13.615) were filtered for ADME properties using QikProp. Ten lead compounds were evaluated in human coronary arter

Nitric oxide pathway: Emerging therapeutic strategies for MASLD.

Metabolic dysfunction-associated steatotic liver disease (MASLD) affects approximately 30-40% of the global population, making it the most prevalent chronic liver disorder worldwide. Its progressive form, metabolic dysfunction-associated steatohepatitis (MASH), may advance to fibrosis, cirrhosis, and hepatocellular carcinoma, imposing a substantial socioeconomic and clinical burden. The recent FDA approval of Resmetirom for non-cirrhotic MASH has marked a significant therapeutic milestone; however, the multifactorial nature of MASLD continues to necessitate the identification of complementary

Oxidative Stress Biomarkers in Silicosis: A Systematic Review.

BACKGROUND: Silicosis is a progressive fibrotic lung disease caused by chronic inhalation of crystalline silica. Increasing evidence indicates that oxidative stress plays a central role in linking silica exposure to inflammation, tissue injury, and fibrosis. We conducted a systematic review to critically appraise the current evidence on the imbalance between oxidant and antioxidant markers in patients with silicosis compared with unexposed healthy controls. METHODS: A systematic literature search was conducted in PubMed, Scopus, and Google Scholar from their inception to 30 November 2025. Eli

CorA2-mediated magnesium transport is essential for stress adaptation and virulence of Streptococcus agalactiae.

Magnesium (Mg2+) homeostasis is critical for bacterial survival and virulence. Streptococcus agalactiae is a major piscine and mammalian pathogen, yet the molecular mechanisms governing its Mg2+ acquisition remain undefined. Here, we identify and characterize CorA2 as an essential Mg2+ transporter in a hypervirulent S. agalactiae strain HN016. Through biochemical and genetic assays including a corA2 deletion mutant (ΔcorA2), a complemented strain, and structure-guided point mutants (GGGG gain of function and D206K loss of function), we demonstrate that CorA2 is essential for Mg2+ uptake. CorA2

Nitric Oxide, Oxidative Stress and Endothelial Dysfunction in Migraine: Recent Advances and Molecular Mechanisms.

Migraine is a highly prevalent and disabling neurovascular disorder that represents a major global health burden due to its significant impact on quality of life and socioeconomic costs. Increasing evidence suggests that migraine pathophysiology involves complex interactions between neuronal hyperexcitability, vascular dysregulation, oxidative stress, and neuroinflammatory processes. Oxidative and nitrosative stress are increasingly recognized as key contributors to migraine mechanisms, influencing mitochondrial dysfunction, cortical spreading depression, and trigeminovascular activation. Immunomodulatory and locomotor regulations via Diosgenin treatment in lipopolysaccharide-induced chronic fatigue syndrome (CFS)/ depressive despair symptom: an in vivo assessment.

Diosgenin is one of the several bioactive chemicals found in fenugreek. In traditional Chinese medicine and modern pharmacological studies, it is useful as anti-cancer, antiviral, anti-inflammatory, and hepatoprotective agents. However, studies investigating its role against antigen-induced neuroinflammation, providing new insights at cellular levels in its pharmacological potential, have not been completely investigated yet. Diosgenin was tested against lipopolysaccharide (LPS)-induced neuroinflammation in mice using forced swim test, tail suspension test, thermal hyperalgesia, locomotor acti

Oxidative stress and metabolic dysfunction in oral lichen planus pathogenesis.

Oral lichen planus (OLP) is a chronic, immune-mediated inflammatory disease of the oral mucosa that can cause persistent pain and has a recognized, though relatively low, risk of malignant transformation. Recent studies have identified oxidative and nitrosative stress as key contributors to the pathophysiology of OLP. Excessive production of reactive oxygen and nitrogen species, combined with reduced antioxidant defenses, leads to cellular damage, including lipid peroxidation, protein modification, and DNA injury. This oxidative imbalance drives keratinocyte apoptosis and sustains inflammatory

Neuroinflammation and Oxidative Stress in Parkinson's Disease, Alzheimer's Disease, and COVID-19: Microglia-Neutrophil Interaction.

Abnormal activation of the immune system and oxidative stress are crucial factors in neurodegenerative disorders, such as Parkinson's disease and Alzheimer's disease. Microglia, neutrophils, oxidative stress mediators such as reactive oxygen species (ROS), lipid peroxidation products (e.g., malondialdehyde), and nitrosative stress markers (e.g., nitrite and nitrate) play important roles in neuroinflammatory mechanisms. Microglial cells acquire a proinflammatory phenotype through interactions with endogenous or exogenous compounds, including cell debris, abnormally modified proteins (including

Nitric oxide signaling in Alzheimer's disease: A double-edged sword.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cognitive decline, memory impairment, and behavioral changes, primarily affecting the elderly population. Despite extensive research, the exact pathogenesis of AD remains elusive, with proposed mechanisms involving amyloid-β accumulation, tau protein hyperphosphorylation, oxidative stress, neuroinflammation, and neuronal apoptosis. Nitric oxide (NO), a gaseous signaling molecule synthesized by nitric oxide synthases (NOS), plays dual roles in AD pathophysiology-acting as both a neuroprotective and neurotoxic

Medical Gas Plasma Inactivates Adenoviruses via Capsid Oxidation.

Pandemics threaten societies, driving the need for new therapeutics. Medical gas plasma, a partially ionized gas, has been proposed as an antiviral agent, but its mechanisms of action are elusive. We employed a certified argon plasma jet and dynamic infection monitoring. Gas plasma exposure reduced adenovirus infectivity in lung cells in treatment time-dependently, with 90s yielding infectivity losses of 96%. The antioxidants N‑acetylcysteine and tyrosine, and the nitric oxide scavenger carboxy‑PTIO, abrogated gas plasma-mediated virus inactivation, implicating hydroxyl radicals and peroxynitr

Atmospheric pollutants and airborne bacteria: adaptation mechanisms, virulence modulation, and public health implications.

Outdoor air pollution is a major public health issue. Many studies correlate ambient air pollution with acute and chronic pulmonary disease. However, its interactions with airborne bacteria remain insufficiently characterized. In particular, the mechanisms linking pollutants to microbial adaptation and pathogenicity are not clearly established. An increasing body of evidence shows that airborne bacteria respond actively to atmospheric pollutants. These responses affect their survival, behavior, and functional traits. However, a comprehensive synthesis of pollutant-driven microbial adaptation a

Ovary Metal Toxicity Remediation by Agro-Food Waste: Evidence for a Regulatory Mechanism of Oxidative Stress by Banana (Musa cavendish) Peel Extract.

Banana (Musa cavendish) peel, usually discarded as waste, is a polyphenol-rich source with antioxidant and chelating properties. This study evaluated its ability to mitigate ovarian toxicity induced by a heavy metal mixture (HMM) consisting of Hg, Mn, Pb, and Al in female rats. Animals received the HMM with or without banana peel extract at 200, 400, and 800 mg/kg dosages for 60 days. Co-treatment dose-dependently reduced ovarian metal accumulation, attenuated oxidative and nitrosative stress (MDA, NO), restored antioxidant enzyme activities (SOD, CAT, GSH, GPx), and modulated pro-inflammatory
