Anticonvulsant role of nitric oxide synthase inhibitors in epilepsy (Review).

Epilepsy is a pathophysiological process triggered by an imbalance between excitatory and inhibitory mechanisms in the brain, resulting in t … e inhibition of nNOS and iNOS activity significantly suppresses glutamatergic hyperexcitability and nitrosative stress associated with N-methyl-D-aspartate receptor activation. nNOS inhibition increases neuronal survival and reduces seizure severity by decreasing intracellular Ca2+ influx. Conversely, endothelial NOS inhibition has been shown to produce proconvulsan …

Dose-Dependent Effects of Nickel on Skeletal Development: Physiological Necessity and the Threshold of Toxicity.

Nickel (Ni) is a ubiquitous trace metal, yet its physiological dynamics and dose-dependent roles in skeletal biology remain unclear. Here we … OS, and induced time-dependent upregulation of Hmox1 and Nos2, consistent with escalating oxidative/nitrosative stress. By contrast, low-dose Ni (0.1 μM) enhanced matrix mineralization, whereas this pro-mineralization effect was attenuated at higher concentrations. In vivo, both Ni deprivation and Ni overload impaired bone formation: a Ni-free diet caused trabecular …

Behavioral standardization and validation of a new model of cancer-induced depression in sarcoma-bearing mice.

Cancer has been correlated with psychiatrical disorders, and incidence of depression is about 3-fold higher in oncological patients. However, there is no validated animal models currently describing sarcoma-associated depression. Herein, we evaluated a preclinical way for estimating behavioral depression-like parameters in Sarcoma-180180180-transplanted mice and investigated the therapeutic efficacy of current clinical antidepressants. Firstly, S-180-bearing Swiss were evaluated using behavioral tests between days 10-11 after S-180 transplantation: open field (OFT), elevated plus maze (EPM), l

Oral Salvia officinalis phenolics abolish mortality and mitigate inflammation in acute TNBS murine colitis.

BACKGROUND: This study is the first to evaluate a phenolic extract of Salvia officinalis in the TNBS murine model of colitis. Inflammatory bowel disease (IBD) involves transmural oxidative and nitrosative stress that up-regulates inducible nitric-oxide synthase (iNOS) and cyclo‑oxygenase-2 (COX-2). Although phenolic constituents of S. officinalis are anti-inflammatory in vitro, their efficacy has never been tested in the trinitrobenzene-sulphonic acid (TNBS) model of colitis. METHODS: Aqueous-ethanolic sage leaves were extracted and standardized [(total phenolics = 6208 ± 200 mg gallic acid e…

Experimental glaucoma triggers a pro-oxidative and pro-inflammatory state in the rat cornea.

BACKGROUND: Increasing evidence suggests that glaucoma affects the ocular surface. We aimed to investigate the cellular mechanisms underlying the glaucoma-associated corneal alterations in an animal model. METHODS: Wistar rats underwent the cauterization of two episcleral veins of the left eye to elevate the intraocular pressure (ipsilateral, G-IL). Control animals received a sham procedure (C-IL). Contralateral eyes did not receive any procedure (G-CL or C-CL). Enzymes related to the redox status, oxidative damage to macromolecules, and inflammatory markers were assessed in corneal lysates.

An H2-infused, nitric oxide-producing functional beverage as a neuroprotective agent for TBIs and concussions.

Traumatic brain injuries (TBIs) are a leading cause of death and disability. Sports-related TBIs are estimated to be more than several million per year. The pathophysiology of TBIs involves high levels of inflammation, oxidative stress, dysregulation of ion homeostasis, mitochondrial dysfunction, and apoptosis. There is also a reduction in cerebral blood flow, leading to hypoxia and reduced removal of metabolic waste, which further exacerbates the injury. There is currently no recognized effective medical treatment or intervention for TBIs, which may in part be due to the difficulty of drug de

The Many Neuroprogressive Actions of Tryptophan Catabolites (TRYCATs) that may be Associated with the Pathophysiology of Neuro-Immune Disorders.

Many, if not all, chronic medical, neurodegenerative and neuroprogressive illnesses are characterised by chronic immune activation, oxidative and nitrosative stress (O&NS) and systemic inflammation. These factors, notably elevated pro-inflammatory cytokines, activate indoleamine 2,3-dioxygenase (IDO) leading to an upregulated tryptophan catabolite (TRYCAT) pathway of tryptophan degradation in the periphery and in the brain. In such conditions the TRYCAT pathway becomes the predominant system for tryptophan degradation in all body compartments. In this paper we review the pathways whereby TRYCA…

Nitrosative and cognitive effects of chronic L-DOPA administration in rats with intra-nigral 6-OHDA lesion.

Besides motor disturbances, other symptoms found in the early stage of Parkinson's disease (PD) are deficits in both learning and memory. The nigro-striatal-cortical pathway is affected in this pathology, with this neuronal circuit involved in cognitive processes such as spatial working memory (SWM). However, cognitive dysfunction appears even when the patients are receiving L-DOPA treatment. There is evidence that the dopamine metabolism formed by L-DOPA generates free radicals such as nitric oxide, which may cause damage through the nitrosative stress (NS). The aim of this study was to evalu…

Blocking cold-inducible RNA-binding protein protects liver from ischemia-reperfusion injury.

Cold-inducible RNA-binding protein (CIRP) is a nuclear protein that has been recently identified as a novel inflammatory mediator in hemorrh … s reduced by anti-CIRP treatment. Anti-CIRP also dramatically decreased the amount of apoptosis and nitrosative stress, evidenced by decrease in TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) staining and inducible nitric oxide synthase and cyclooxygenase 2 levels, respectively. Finally, the 10-day survival rate was increased from 37.5% in the …

The Toll-Like Receptor Radical Cycle Pathway: A New Drug Target in Immune-Related Chronic Fatigue.

In this review we discuss that peripheral and central activation of the Toll-like receptor 2/4 (TLR2/4) Radical Cycle may underpin the patho … its treatments. Increased TLR signaling is driven by activated immuneinflammatory and oxidative and nitrosative stress pathways, pathogen derived molecular patterns, including lipopolysaccharides, and damage associated molecular patterns (DAMPs). Newly formed redox-derived DAMPs, secondary to oxidative processes, may further activate the TLR complex leading to an au …

Nicotine inhibits the proliferation by upregulation of nitric oxide and increased HDAC1 in mouse neural stem cells.

Cigarette smoking (CS) is considered one of the major risk factors to cause neurodegenerative disorders. Nicotine is the main chemical in CS which is responsible for dysfunction of the brain as a neuroteratogen. Also, nicotine dependency is a real mental illness and disease. Recently, chronic nicotine exposure has been shown to cause oxidative/nitrosative stress leading to a deleterious condition to cellular death in different brain regions. However, little is known about the effects of nicotine on mouse neural stem cells (mNSCs). The aim of this study is to investigate the effects of nicotine…
