Simultaneous detection of SARS-CoV-2 proteins and nucleic acids by coupling PNIPAM with circle-to-circle amplification.

In the past few years, the global uncontrollable spread of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has underscored the need for rapid, sensitive, and accurate detection methods. Given the continuous mutation characteristics of viruses, single-target detection carries the risk of false-positive or false-negative results, whereas dual-target detection can effectively mitigate such risks. Therefore, thermosensitive polymer poly(N-isopropylacrylamide) (PNIPAM) was integrated with an isothermal amplification method-circle-to-circle amplification (C2CA) in this study, enabling i

Prevalence and genetic variation of the M, N, and S2 genes of feline coronavirus in Shandong Province, China.

Feline coronavirus (FCoV) is the causative agent of feline infectious peritonitis and diarrhoea in kittens worldwide. In this study, a total of 73 feline diarrhoeal faecal samples were collected from animal hospitals and pet markets in ShanDong province from 2017 to 2019. FCoV was detected in 58.23% (46/73) of the samples, using the RT-PCR method. The results showed that the detection rate of FCoV in healthy cats and sick cats was 41.7% (10/24) and 81.6% (40/49), respectively. Full gene amplification and sequencing of the N, M, and S2 genes of FCoV isolates were performed. An amino acid mutati

Host factor MxA restricts Dabie bandavirus infection by targeting the viral NP protein to inhibit NP-RdRp interaction and ribonucleoprotein activity.

Severe fever with thrombocytopenia syndrome (SFTS) is an emerging infectious disease with high case mortality rates, which is caused by Dabi … DBV infection. Mechanistic target analysis revealed that MxA specifically interacts with the viral nucleocapsid protein (NP) in a manner independent of RNA. Minigenome reporter assay showed that in agreement with its targeting of NP, MxA inhibits DBV ribonucleoprotein (RNP) activity. In detail, MxA interacts with the NP N-terminal and disrupts the interaction …

Hepatitis B Virus.

This infographic about hepatitis B virus explores its replication cycle, natural history of infection and pathogenesis, and how this can be controlled and treated. Hepatitis B virus (HBV) is a common worldwide blood-borne pathogen. Chronic hepatitis B can progress to an inactive carrier state, and then, in some patients, give rise to cirrhosis and cancer of the liver, leading to death. An HBV surface-antigen vaccine is effective, but treatments are currently not curative. HBV replicates via reverse transcription. Its covalently closed circular (ccc) DNA in the nucleus encodes a pregenomic RNA

Increased anti-SARS-CoV-2 antibody titers in plasma-derived intravenous immunoglobulin collected in Southern China.

Although the COVID-19 pandemic is no longer considered an emergency, the ongoing spread of the virus poses a persistent threat to public health. COVID-19 poses a challenge to the system due to the risk of saturation and the associated economic burden. To assess changes in the anti-SARS-CoV-2 antibody profile, we analyzed nine batches of intravenous immunoglobulin (IVIG) prepared from plasma samples collected between November 2020 and June 2023. Antibody changes against SARS-CoV-2 spike, nucleocapsid, and membrane proteins, and neutralizing antibodies were measured. Antibody titers against wild…

Non-Neutralising Antibodies and T Cell Responses to SARS-CoV-2 Detected in Unvaccinated SARS-CoV-2-Naive Spanish Adolescents.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection in children and adolescents usually follows a mild or asymptomatic course. In this brief report, we investigated the cellular and humoral responses to SARS-CoV-2 in six healthy Spanish female adolescents, aged 14 to 16, using blood samples collected early in 2021, well before COVID-19 vaccinations. Using IFNγ-ELISPOT assays, we detected T cell recall responses in four of these adolescents to a peptide pool encompassing the SARS-CoV-2 Spike protein. The same subjects also exhibited IgM antibodies against the Nucleocapsid and…

Chemokines as potential biomarkers for predicting the course of COVID-19 - a review of the literature.

Since the beginning of the COVID-19 pandemic, research has been ongoing to find the best diagnostic parameters to identify patients with a high risk of severe infection. Numerous studies have examined chemokine biomarkers in COVID-19 as a biomarker for high risk patients. The four main structural proteins of the SARS-CoV-2, spike protein, membrane protein, envelope protein and nucleocapsid protein enable the virus to penetrate host cells and stimulate the immune system. SARS-CoV-2 enters host cells via ACE2 in upper respiratory tract the virus entries by binding to the spike protein. Uncontrol…

Cloning and phylogenetic analysis of N protein gene from Rift Valley Fever Virus (RVFV).

Rift Valley Fever (RVF) is a mosquito-borne viral zoonosis caused by RVFV in humans and livestock. Currently, there are no approved vaccines or antiviral therapies available. Additionally, in Saudi Arabia, there is a lack of a routine screening system to monitor RVFV in humans and animals which hinders to design and develop the preventive measures as well as the prediction of future outbreaks and the potential re-emergence of RVFV. Hence, we have performed the cloning, sequencing, and phylogenetic analysis, of nucleocapsid (N) protein gene. The sequence analysis showed high similarities with R…

Evidence for increased SARS-CoV-2 susceptibility and COVID-19 severity related to pre-existing immunity to seasonal coronaviruses.

The importance of pre-existing immune responses to seasonal endemic coronaviruses (HCoVs) for the susceptibility to SARS-CoV-2 infection and the course of COVID-19 is the subject of an ongoing scientific debate. Recent studies postulate that immune responses to previous HCoV infections can either have a slightly protective or no effect on SARS-CoV-2 pathogenesis and, consequently, be neglected for COVID-19 risk stratification. Challenging this notion, we provide evidence that pre-existing, anti-nucleocapsid antibodies against endemic α-coronaviruses and S2 domain-specific anti-spike antibodies…

Developing effective siRNAs to reduce the expression of key viral genes of COVID-19.

The COVID-19 pandemic has been raging worldwide for more than a year. Many efforts have been made to create vaccines and develop new antiviral drugs to cope with the disease. Here, we propose the application of short interfering RNAs (siRNAs) to degrade the viral genome, thus reducing viral infection. By introducing the concept of the probability of binding efficiency (PBE) and combining the secondary structures of RNA molecules, we designed 11 siRNAs that target the consensus regions of three key viral genes: the spike (S), nucleocapsid (N) and membrane (M) genes of SARS-CoV-2. The silencing …

The hepatitis delta virus: Replication and pathogenesis.

Hepatitis delta virus (HDV) is a defective virus and a satellite of the hepatitis B virus (HBV). Its RNA genome is unique among animal viruses, but it shares common features with some plant viroids, including a replication mechanism that uses a host RNA polymerase. In infected cells, HDV genome replication and formation of a nucleocapsid-like ribonucleoprotein (RNP) are independent of HBV. But the RNP cannot exit, and therefore propagate, in the absence of HBV, as the latter supplies the propagation mechanism, from coating the HDV RNP with the HBV envelope proteins for cell egress to delivery …

Unexpected features and mechanism of heterodimer formation of a herpesvirus nuclear egress complex.

Herpesvirus nucleocapsids escape from the nucleus in a process orchestrated by a highly conserved, viral nuclear egress complex. In human cytomegalovirus, the complex consists of two proteins, UL50 and UL53. We solved structures of versions of UL53 and the complex by X-ray crystallography. The UL53 structures, determined at 1.93 and 3.0 Å resolution, contained unexpected features including a Bergerat fold resembling that found in certain nucleotide-binding proteins, and a Cys3His zinc finger. Substitutions of zinc-coordinating residues decreased UL50-UL53 co-localization in transfected cells, …
