EIF4A3 targeted therapeutic intervention in glioblastoma multiforme using phytochemicals from Indian medicinal plants - an integration of phytotherapy into precision onco-medicine.

Glioblastoma Multiforme (GBM), an aggressive brain tumor (grade-IV astrocytoma), poses treatment challenges. Poor prognosis results from the rapid growth, highlighting the role of EIF4A3 in regulating non-coding RNAs. EIF4A3 promotes the expression of several non-coding RNAs, viz, Circ matrix metallopeptidase 9 (MMP9), a prominent oncogene, by interacting with the upstream region of the circMMP9 mRNA transcript and acts on cell proliferation, migration, and invasion of GBM. However, research shows that EIF4A3 knockdown inhibits glioblastoma progression and increases apoptosis. In this study, w

Gastrin and cholecystokinin 2 receptor, and gastrin-releasing peptide receptor in cancer: From pathogenesis to precision medicine.

Gastrin regulates gastric acid secretion and mucosal homeostasis via the cholecystokinin-2 receptor (CCK2R). Aberrant signaling via CCK2R (cholecystokinin family) and the distinct class A GPCR, gastrin-releasing peptide receptor (GRPR; bombesin family), drives oncogenesis across diverse GI and non-GI malignancies through calcium mobilization, MAPK/PI3K activation, EGFR transactivation, and constitutive splice variants like CCK2Ri4sv. Furthermore, precursor forms such as progastrin remodel the tumor microenvironment by interacting with non-CCK binding sites (p78, Annexin A2), converting fibrobl

From uncertainty to clarity: The evolution of Nuclear Medicine towards precision theranostics and AI-driven digital twins.

Nuclear medicine has progressed from an exploratory discipline, historically constrained by limited spatial resolution and qualitative interpretation, into a rigorously quantitative clinical specialty. This evolution represents a systematic conversion of uncertainty into actionable information. Uncertainty that was once implicit and observer-dependent is increasingly measured, modelled, and managed across the full imaging-therapy continuum. Early tracer work established the foundational diagnostic logic of assessing function before structure. Subsequent instrumentation advances, culminating in

The molecular puzzle of recurrent implantation failure: integrating genetic, epigenetic, and immune mechanisms for precision reproductive medicine.

Recurrent implantation failure (RIF) is a frustrating condition for the majority of infertile couples, occurring when pregnancy does not happen after several embryo transfers through assisted reproductive technologies (ART). ART has progressed over the last few years, but RIF continues for many patients, and the real reason remains unknown in most cases. RIF is a multifactorial condition influenced by both genetic and epigenetic mechanisms. Genetic contributions to RIF can be broadly classified into embryo-related factors (such as chromosomal aneuploidies and single-gene mutations), maternal f

Molecular Mechanisms and Therapeutic Strategies for Immune Checkpoint Inhibitors in Breast Cancer: From Pathogenesis to Precision Medicine.

The advent of immunotherapy, and particularly the development of immune checkpoint inhibitors (ICIs), has revolutionized the landscape of breast cancer treatment, especially for triple-negative breast cancer. However, some patients still do not benefit from immunotherapy. Breast cancer is inherently an immunotherapy "cold" tumor, which results in suboptimal clinical outcomes when ICIs are used as monotherapy. Identifying additional immunotherapeutic targets and drugs, along with developing novel strategies for combination therapy, is crucial for addressing the challenges posed by immunotherapy

Representative Biobanks as Infrastructure for Equitable Precision Medicine: Implications for Clinical Trials, Artificial Intelligence, and Translational Research.

The shift toward precision medicine has transformed clinical trial design and drug delivery, yet persistent gaps in data diversity continue to contribute to health disparities. Broad representation and precision stratification are not competing goals; rather, representative datasets are the necessary foundation for identifying clinically meaningful molecular, demographic, environmental, and social subgroups. Despite regulatory mandates and advances in molecular biology, artificial intelligence (AI), and adaptive trials, patient datasets remain heavily skewed toward European populations, with c…
