To assess the role of prolactin (PRL) in the puberty onset and the ovarian aromatase activity, hyper- and hypoprolactinemic conditions were induced by treating chronically prepubertal female rats with PRL and bromocriptine, respectively. The effects of prolactin (PRL), alone and as a modulator of human chorionic gonadotropin (hCG) action, on steroidogenesis and cAMP accumulation in rat luteal cell cultures were examined. PRL levels were evaluated during long-term treatment with cyproterone acetate 100 mg and ethinyloestradiol 100 micrograms/day orally or depot-oestrogens in 214 male-to-female transsexuals. PRL levels increased above normal in all subjects (normal less than 300 mU/l). In 46 (21.4%) subjects PRL levels rose to greater than 1000 mU/l. The incidence of PRL levels greater than 1000 mU/l was 3.7-7.2% per treatment year. The benzazepine, SCH 23390 (10(-5) M), is able to block D2 anterior pituitary receptors and their interaction with a maximal concentration of dopamine (10(-7) M) in an experimental setting involving superfused pituitary slices and prolactin release by lactotrophs as a functional model of such receptors; this finding which correlates nicely with binding studies and drug-induced hyperprolactinaemia supports a SCH 23390 D1 dose-related selectivity; given at doses higher than those inducing behavioural changes, the compound may affect D2 receptor-driven functions, e.g. prolactin secretion.

To determine the frequency of hormonal abnormalities in a Nigerian population of male partners of infertile relationships, serum levels of luteinizing hormone, follicle-stimulating hormone, prolactin, and testosterone were estimated using radioimmunoassay techniques in 454 oligo/azoospermic male partners of infertile marriages. The mol wt (Mr) of intact murine PRL is approximately 23,000. Immunoblotting analysis shows a 21,000 Mr band in fresh rat and mouse pituitary extracts that cross-reacts strongly with PRL antibodies. The prolactin response to thyrotropin-releasing hormone (TRH) and metoclopramide was studied in 16 patients with Sheehan's syndrome and 16 matched controls in the follicular phase. Metoclopramide resulted in a greater prolactin response than TRH did in the controls. However, both stimuli failed to evoke any appreciable prolactin response in the patients with Sheehan's syndrome. Since metoclopramide is generally free of side effects and far cheaper than TRH, we recommend the prolactin response to metoclopramide as the preferred screening test in the diagnosis of Sheehan's syndrome.

The effect of DHEAS, Prolactin and Testosterone hormones to female infertility and hirsutism have been investigated. In primary infertility group average DHEAS value is 491 +/- 95 ng/ml so it is meaningful low (p less than 0.01). In fertil hirsutic group (+4) category average DHEAS value is 4166 ng/ml, meaningful high (p less than 0.01). The relation between DHEAS and Prolactin also remarkable.

Nutritional iron deficiency (ID), like neuroleptic treatment, results in a reduction in dopaminergic activity and a rise in serum prolactin (PRL).
