Surveillance of Plasmodium falciparum pfcrt haplotypes in southwestern uganda by high-resolution melt analysis.

BACKGROUND: Chloroquine (CQ) resistance is conferred by mutations in the Plasmodium falciparum CQ resistance transporter (pfcrt). Following CQ withdrawal for anti-malarial treatment, studies across malaria-endemic countries have shown a range of responses. In some areas, CQ sensitive parasites re-emerge, and in others, mutant haplotypes persist. Active surveillance of resistance mutations in clinical parasites is essential to inform treatment regimens; this effort requires fast, reliable, and cost-effective methods that work on a variety of sample types with reagents accessible in malaria-ende

Glutathionylation at Cys-111 induces dissociation of wild type and FALS mutant SOD1 dimers.

Mutation of the ubiquitous cytosolic enzyme Cu/Zn superoxide dismutase (SOD1) is hypothesized to cause familial amyotrophic lateral sclerosis (FALS) through structural destabilization leading to misfolding and aggregation. Considering the late onset of symptoms as well as the phenotypic variability among patients with identical SOD1 mutations, it is clear that nongenetic factor(s) impact ALS etiology and disease progression. Here we examine the effect of Cys-111 glutathionylation, a physiologically prevalent post-translational oxidative modification, on the stabilities of wild type SOD1 and tw

Targeting autophagy in ALS: a complex mission.

Several neurodegenerative diseases share a common neuropathology, primarily featuring the presence of abnormal protein inclusions in the brain containing specific misfolded proteins. Strategies to decrease the load of protein aggregates and oligomers are considered relevant targets for therapeutic intervention. Many studies indicate that macroautophagy is a selective and efficient mechanism for the degradation of misfolded mutant proteins related to neurodegeneration, without affecting the levels of the corresponding wild-type form. In fact, activation of autophagy by rapamycin treatment decre

Conformation studies of various hemoglobins by natural-abundance 13 C NMR spectroscopy.

Studies of variously liganded hemoglobins (both from human and rabbit) by natural-abundance (13)C NMR spectroscopy have revealed apparent conformational differences that have been interpreted on the basis of two quaternary structures for the alpha(2)beta(2) tetramer, and variable tertiary structures for the individual alpha and beta subunits. In solution, rabbit hemoglobins appear to have somewhat more flexibility than human hemoglobins.

LNA-Modified DNAzyme Nanomachines Enable Physiological-Temperature Recognition of Double-Stranded DNA.

Sequence-specific detection of double-stranded DNA (dsDNA) under physiological conditions remains challenging because the complementary strand is inaccessible to conventional oligonucleotide probes, often requiring thermal denaturation or protein-assisted strand separation. Here, we developed a binary DNAzyme (BiDz) sensor for sequence-specific dsDNA detection at 37 °C. While the all-DNA BiDz failed to recognize dsDNA efficiently, incorporating alternating locked nucleic acid (LNA)/DNA residues into the analyte-binding arms enabled efficient strand invasion and markedly improved signal generat

Mechanisms and efficacy of wet heat, dry heat, and steam-based treatments for bacterial spore inactivation.

Dry and wet heat treatments are widely used physical methods for microbial inactivation, and their effectiveness in destroying bacterial spores has been extensively demonstrated. This review systematically discusses the bactericidal effects and underlying mechanisms of dry heat, wet heat, and steam-based treatments on bacterial spores. Wet heat inactivates spores through heat-induced protein denaturation and dipicolinic acid release, resulting in multi-level structural damage, including spore coat collapse, cortical peptidoglycan hydrolysis, and core hydration. On the other hand, dry heat trea…

Anti-arthritic and anti-inflammatory activity of linalool against formaldehyde and complete Freund's adjuvant induced arthritis in rats.

Arthritis, characterized by persistent joint inflammation, poses a significant challenge due to its detrimental effects on cartilage function and subchondral bone health. This study investigates the potential pharmacological effects of linalool, a monoterpene alcohol commonly found in essential oils, as a promising treatment for arthritis. In vitro assays, including red blood cell membrane stability and protein denaturation tests, were conducted to preliminarily assess the anti-arthritic potential of linalool. Preclinical evaluations were performed on rats with arthritis induced by formaldehyd…

A Rapid Electrochemical DNA Biosensor for Serum Uric Acid Using the Pre-equilibrium Binding Mechanism.

The precise and rapid detection of uric acid (UA) in human serum is pivotal for diagnosing and managing metabolic disorders such as gout, kidney disease, and hyperuricemia. However, current equilibrium-based assays suffer from long incubation time, interference from serum components, and a narrow dynamic range. Herein, we present a robust electrochemical DNA assay (EC-DNA) that can respond rapidly (within 1 min) to serum UA under pre-equilibrium conditions, with an extended dynamic range (1-1000 μM). Our assay employs a Mg2+-dependent UA aptamer that can form a metastable hairpin-like structur

Exploring Multivalent Architectures for Binding and Stabilization of N-Acetylgalactosamine 6-Sulfatase.

Morquio A syndrome is a lysosomal disorder caused by the deficiency of the lysosomal enzyme N-acetylgalactosamine 6-sulfatase (GALNS, EC 3.1.6.4). Currently, enzyme replacement therapy (ERT) is used to treat Morquio A through the infusion of the recombinant enzyme VIMIZIM® (elosulfase alfa, BioMarin). Unfortunately, the recombinant enzyme exhibits low conformational stability in vivo. A promising approach to address this issue is the coadministration of recombinant human GALNS (rhGALNS) with a pharmacological chaperone (PC), a molecule that selectively binds to the misfolded protein, stabilize

Synergistic coating of Laponite swollen-layer and heparin composite for enhanced antifouling and antithrombotic performance.

When implantable medical devices come into contact with blood, acute thrombosis and inflammation often occur due to the adhesion and denaturation of plasma proteins, ultimately degrading device performance. To address this issue, we developed a gel-like coating with superhydrophilic properties, incorporating a heparin based outer layer designed to minimize foreign body reactions and enhance hemocompatibility. The coating was engineered using a layer-by-layer (LbL) assembly of poly-l-lysine (PLL), laponite (Lap), and heparin (Hep), utilizing electrostatic interactions. The successful formation

Plasmonic Cu2- xS ySe1- y Nanoparticles Catalyzed Click Chemistry Reaction for SERS Immunoassay of Cancer Biomarker.

Nature enzyme-based immunoassays have been widely used in fundamental scientific research and clinical diagnosis. However, the limitations of natural enzyme, such as the low physical/chemical stability or susceptibility to protein denaturation, greatly restrained its applications. In this article, we reported a new enzyme-free SERS immunoassay by utilizing plasmonic Cu2- xS ySe1- y nanoparticles (NPs) as nanocatalyst to catalyze the click chemistry between the azido and alkynyl substrate which is used as the SERS signal reporter. The unique vibration of C≡C of alkynyl in the Raman-silent regio…

Lyophilized bone marrow cell extract functionally restores irradiation-injured salivary glands.

OBJECTIVE: Bone marrow cell extract (BMCE) was previously reported to restore salivary gland hypofunction caused by irradiation injury. Proteins were shown to be the main active factors in BMCE. However, BMCE therapy requires multiple injections and protein denaturation is a concern during BMCE storage. This study aimed to preserve, by lyophilization (freeze-drying), the bioactive factors in BMCE. METHODS: We developed a method to freeze-dry BMCE and then to analyze its ingredients and functions in vivo. Freeze-dried (FD) BMCE, freshly prepared BMCE (positive control), or saline (vehicle cont…

Etiologies of sperm oxidative stress.

Sperm is particularly susceptible to reactive oxygen species (ROS) during critical phases of spermiogenesis. However, the level of seminal ROS is restricted by seminal antioxidants which have beneficial effects on sperm parameters and developmental potentials. Mitochondria and sperm plasma membrane are two major sites of ROS generation in sperm cells. Besides, leukocytes including polymer phonuclear (PMN) leukocytes and macrophages produce broad category of molecules including oxygen free radicals, non-radical species and reactive nitrogen species. Physiological role of ROS increase the intrac

Chaperone therapy for Krabbe disease: potential for late-onset GALC mutations.

Krabbe disease is an autosomal recessive leukodystrophy caused by a deficiency of the galactocerebrosidase (GALC) enzyme. Hematopoietic stem cells transplantation is the only available treatment option for pre-symptomatic patients. We have previously reported the chaperone effect of N-octyl-4-epi-β-valienamine (NOEV) on mutant GM1 β-galactosidase proteins, and in a murine GM1-gangliosidosis model. In this study, we examined its chaperone effect on mutant GALC proteins. We found that NOEV strongly inhibited GALC activity in cell lysates of GALC-transfected COS1 cells. In vitro NOEV treatment st

Molecular cloning and analysis of stage and tissue-specific expression of Cathepsin L-like protease from Clonorchis sinensis.

Cathepsin L of parasite plays multiple roles in growth, food uptake, and invasion into host and pathogenesis, which makes it a valuable target for diagnosis, vaccine, and drug. In this study, we identified a cDNA encoding cathepsin L homolog (CsCPL) from the library of Clonorchis sinensis adult by bioinformatics analysis. Sequence encoding proenzyme of CsCPL (removal of signal peptide, CsproCPL) was highly expressed in form of inclusion body in Escherichia coli, and soluble rCsproCPL (about 1 mg/ml) in high purity were obtained after denaturation, purification, and renaturation.
