Exploring sibling patterns in dental treatment under general anaesthetic.

Aims Dental treatment under general anaesthesia (DGA) in children is a major public health concern. In the UK, dental extractions are the leading cause of hospital admissions for 6-10-year-olds. Although family-related risk factors are known, no UK-based study has explored the characteristics of siblings undergoing DGA. The aims of this study were to: 1) describe the sociodemographic characteristics of siblings receiving DGA; 2) outline the diagnoses and treatment; 3) identify predictors of repeat DGA among siblings; and 4) compare within- and between-family factors. Methods This retrospective …

Managing Family Accommodation in OCD: A Sibling-focused Case Study.

Family accommodation (FA) is prevalent in obsessive-compulsive disorder (OCD). When patients refuse treatment, the impacts are more severe-with poorer clinical outcomes, greater FA, and more caregiver burden. In these cases, caregiver interventions are an essential clinical priority. However, clinical documentation of structured, culturally informed interventions for FA and caregiver burden for adults with OCD, in the absence of patient participation, remains limited. This case study describes an eight-session caregiver-focused intervention for a 26-year-old male experiencing burnout while acc

Expanding the phenotypic Spectrum of ZNF711: Autism and epilepsy in two siblings.

Zinc Finger Protein 711 (ZNF711) is a Krüppel-type zinc finger transcription factor highly expressed in the brain and whose mutations have been associated with neurodevelopmental disorders such as intellectual disability, autism spectrum disorder, and epilepsy. While most pathogenic variants previously described affect the C-terminal DNA-binding domain, we report two siblings carrying a rare hemizygous missense variant in the N-terminal transactivation domain, NM_001330574.2: c.65 T > C (p.Ile22Thr), inherited from their heterozygous unaffected mother. Patient #1 presents with autism spectrum …

Glycoproteomics and Functional Characterization of Novel Variants in Siblings with ALG1-CDG.

Congenital disorders of glycosylation (CDGs) are a growing group of inborn errors caused by gene defects in glycan biosynthesis pathways. Genetic testing of two patients from the same family harboring ALG1-related CDG (ALG1-CDG) revealed heterozygous variations in the c.1129 A>C and c.1263+3 A>T. Although most CDGs with abnormal N-glycosylation can be detected by transferrin screening, Western blotting of serum transferrin from the two siblings did not reveal significant glycosylation deficiency. In this study, the biochemical phenotype and pathological molecular basis of the CDGs in the sibli…

A Novel Homozygous GFI1B Mutation in Siblings With Thrombocytopenia and Bleeding Tendency.

Platelet type bleeding disorder 17 (OMIM #187900) is a type of "gray platelet syndrome" and occurs due to a mutation in the GFI1B gene on chromosome 9q34.13. Patients usually present with a history of easy bleeding tendencies, recurrent epistaxis or gum bleeding, and rarely with severe hemorrhage, and they may often be misdiagnosed. We herein report a novel autosomal recessive bleeding disorder in two sisters of the same family caused by a mutation in GFI1B, where both sisters, 13 and 8 years old, respectively presented with a history of recurrent episodes of epistaxis, firstly misdiagnosed an

WDR81 Mutation in Two Siblings: A Case Report and Review of Literature.

INTRODUCTION: The WDR81 gene is critically involved in autophagy, endosomal trafficking, and neurodevelopment. Pathogenic variants in this gene are associated with autosomal recessive disorders, such as cerebellar ataxia, intellectual disability, and disequilibrium syndrome. Despite recent advances, the full phenotypic spectrum remains incompletely characterized. CASE PRESENTATION: We report two female siblings carrying a homozygous missense variant in the WDR81 gene. The elder sister (age 15) presented with developmental regression, optic atrophy, motor neuropathy, and pes equinovarus. The y…

Ocular Findings in Siblings With Alagille Syndrome: A Report of Two Cases.

Alagille syndrome (ALGS) is a multisystem disorder, most often caused by JAG1 variants. Ocular features classically include posterior embryotoxon and optic disc anomalies, but glaucoma is uncommonly reported. We report two Latino siblings with ALGS and glaucoma. They share the previously described JAG1 c.925G>C (p.Gly309Arg) variant, classified as a variant of uncertain significance (VUS). One sibling exhibited a posterior embryotoxon, accompanied by retinal nerve fiber layer (RNFL) thinning and elevated intraocular pressure (IOP), which responded to topical therapy. The other had a microcorne…

Family history of psychiatric conditions and development of siblings of children with autism.

Younger siblings (SIBS) of children with autism exhibit a wide range of clinical and subclinical symptoms including social, cognitive, language, and adaptive functioning delays. Identifying factors linked with this phenotypic heterogeneity is essential for improving understanding of the underlying biology of the heterogenous outcomes and for early identification of the most vulnerable SIBS. Prevalence of neurodevelopmental (NDD) and neuropsychiatric disorders (NPD) is significantly elevated in families of children with autism. It remains unknown, however, if the family history associates with …

Illuminating the landscape of sibling relationship quality: An evidence and gap map.

This paper used an evidence and gap map (EGM) to advance the scientific understanding of sibling relationship quality among children aged 2 to 18 years by synthesizing literature on 277 empirical studies from 1985 to 2022 to delineate patterns of study design, sampling, and measurement. Most existing research has utilized majority of White, middle-to-upper class, and/or two-caregiver family samples. Nearly 85% (n = 235) of studies used quantitative methods to measure sibling relationship quality across eight domains: conflict, warmth/affection, quality, cohesion, hostility, power/control, posi

Is early childhood development impeded by the birth timing of the younger sibling?

BACKGROUND: This study investigated whether the timing of birth of the younger siblings was associated with the risk of the older siblings' developmental vulnerability in early childhood. METHODS: Linkage of population-level birth registration, hospital, and perinatal datasets to Australian Early Development Census (AEDC) records (2009-2015), enabled follow-up of a cohort of 32,324 Western Australia born singletons. Children with scores <10th percentile on an individual AEDC domain (Physical Health and Wellbeing; Social Competence; Emotional Maturity; Language and Cognitive Skills (school-bas…

Abetalipoproteinemia Due to a Novel Splicing Variant in MTTP in 3 Siblings.

Abetalipoproteinemia (ABL) is a rare recessive condition caused by biallelic loss-of-function mutations in the MTTP gene encoding the microsomal triglyceride transfer protein large subunit. ABL is characterized by absence of apolipoprotein B-containing lipoproteins and deficiencies in fat-soluble vitamins leading to multisystem involvement of which neurological complications are the most serious. We present 3 siblings with ABL who were born to non-consanguineous parents of Filipino and Chinese background. Identical twin boys with long-standing failure to thrive and malabsorption were diagnosed…
