Molecular pathways: beta-adrenergic signaling in cancer.

Beta-adrenergic signaling has been found to regulate multiple cellular processes that contribute to the initiation and progression of cancer, including inflammation, angiogenesis, apoptosis/anoikis, cell motility and trafficking, activation of tumor-associated viruses, DNA damage repair, cellular immune response, and epithelial-mesenchymal transition. In several experimental cancer models, activation of the sympathetic nervous system promotes the metastasis of solid epithelial tumors and the dissemination of hematopoietic malignancies via β-adrenoreceptor-mediated activation of protein kinase …

Age-related changes in sympathetic activity: biochemical measurements and target organ responses.

The aim of this study was to evaluate using several parameters the effect of aging on sympathetic nervous system activity. We measured heart rate, skin conductance level and free and total plasma catecholamines in 60 normal subjects aged 20-80 years, divided into three age groups, in basal supine conditions and in response to standing and to hyperventilation. Basal heart rate was similar in all subjects and failed to correlate with age. Skin conductance level decreased with increasing age and correlated negatively and significantly with age. In response to standing and to hyperventilation, hea…

Spontaneous hypertension is primarily the result of sympathetic overactivity and immunologic dysfunction.

Overactivity of the sympathetic nervous system and immunologic dysfunction have been shown to contribute to development and maintenance of hypertension in the Okamoto spontaneously hypertensive rat (SHR). In this study, the combined effects of reduction in sympathetic activity and immunologic manipulation on spontaneous hypertension have been determined. Neonatal SHRs received sham implants or implants of thymic tissue from Wistar donor rats. In addition, the thymus-implanted SHRs underwent bilateral renal denervation when they were 6 weeks old. At the same time, the sham-implanted SHRs underw…

Assessing Correlation between Heart Rate Variability Markers Based on Laguerre Expansion and Direct Measures of Sympathetic Activity during Incremental Head-up Tilt.

Traditional frequency domain analysis of heart period (HP) variability allows the estimation of the parasympathetic modulation directed to the heart but the sympathetic one remains largely unknown. Recently, sympathetic and parasympathetic activity indexes (SAI and PAI) have been proposed to address this issue. SAI and PAI were derived from HP variability via the application of an orthonormal Laguerre expansion allowing the separation of HP variations driven by sympathetic and parasympathetic outflows. In this study, SAI and PAI were validated against tonic and variability measures of muscle s

The airway neurogenic inflammation: clinical and pharmacological implications.

Bronchial airway microvasculature consists in a developed network of vessels, which plays an important role in normal homeostasis as well as in inflammatory airway processes. Its airway autonomic neural control includes cholinergic and adrenergic innervation, as well as nonadrenergic noncholinergic system. The nerve/vessel interplay is complex and not yet completely clarified. In response to inspired air conditions, the sensory nerves can recruit appropriate reflexes, which can induce different vascular processes, such as vasodilatation, vasoconstriction, plasma extravasation and exudation. Modulation of host immunity by HIV may be partly achieved through usurping host autonomic functions.

Modulation of host immunity has been observed in human immunodeficiency virus (HIV) infections. HIV is believed to influence host immunity through a variety of mechanisms including direct effects on host T cell survival, indirect effects on cytokine profile through modulation of immune cells, and modulation of endocrine functions that affect immunity such as steroids. We hypothesize that HIV infection may also alter host immunity through modulation of host sympatho-vagal balance. Specifically, we propose that HIV drives autonomic balance towards sympathetic bias, which can contribute to a T he

Sympathetic skin response: basic mechanisms and clinical applications.

Sympathetic skin response (SSR), defined as the momentary change of the electrical potential of the skin, may be spontaneous or reflexively evoked by a variety of internal or by externally applied arousal stimuli. Although the suprasegmental structures influencing the SSR in humans are not well known, SSR has been proposed as a non-invasive approach to investigate the function of the sympathetic system. SSR is easy to apply but current procedures are not sufficiently reliable for diagnostic purposes, and show imperfect correlations both with clinical features and other measurements of autonomi

[Pharmacologic treatment of urinary dysfunctions].

After reviewing the nervous control of the lower urinary tract, the author defines the action of various drugs on the bladder, bladder neck, urethra (smooth and striated sphincter). The drugs are classified according to their stimulant or blocking effect on the sympathetic and parasympathetic nervous systems and alpha and betadrenergic receptors. Drugs acting on smooth and striated muscle fibres are then studied and the iatrogenic adverse effects of these drugs are mentioned.

[Neurophysiological mechanisms of the reflex lacrimation].

Neurophysiological mechanisms of the reflex lacrimation were analyzed in anesthetized rabbits. The secretory pattern of the lacrimation elicited by stimulation of the cornea consisted of two phases, that is, a rapid flow phase during stimulation and the subsequent slow flow phase in post-stimulus time. Parasympathetic nerve activities are closely related to the secretory volume in the rapid flow phase of the reflex lacrimation. On the other hand, excitation of the sympathetic nerve depressed the secretion in the rapid flow phase, while it facilitated slightly the secretion in the slow flow pha

Three layers of the opossum stomach: responses to nerve stimulation.

The three muscle layers of the opossum stomach were compared in their responses to electrical field stimulation. Strips were cut from the muscular wall of the stomach with their longer axes in the direction of the longitudinal, circular, and oblique muscle layers and stimulated at 1-min intervals with 20-sec trains of 1-msec pulses at 10 Hz and supramaximal strength at 500 ma. Strips either contracted, relaxed, or did not respond during the stimulus trains. Responses were abolished by tetrodotoxin, 10(-6) M. Contractions, but not relaxations, were abolished by atropine, 10(-6) M. Relaxations w

The sympathetic and parasympathetic nature of neuropeptide Y-immunoreactive nerve fibres in the major salivary glands of the rat.

The distribution and origin of neuropeptide Y in the major salivary glands of the rat was studied by indirect immunofluorescence technique. Numerous nerve fibres immunoreactive for the peptide were seen in the parotid and sublingual glands. Most of the fibres were located around blood vessels and salivary acini. In the submandibular gland the number of immunoreactive nerve fibres around the acini was lower in comparison with that in the parotid and sublingual glands. Some immunoreactive nerve fibres were also found around or along intra- and interlobular ducts in all major salivary glands. A l

Vascular Mural Cells Promote Noradrenergic Differentiation of Embryonic Sympathetic Neurons.

The sympathetic nervous system controls smooth muscle tone and heart rate in the cardiovascular system. Postganglionic sympathetic neurons (SNs) develop in close proximity to the dorsal aorta (DA) and innervate visceral smooth muscle targets. Here, we use the zebrafish embryo to ask whether the DA is required for SN development. We show that noradrenergic (NA) differentiation of SN precursors temporally coincides with vascular mural cell (VMC) recruitment to the DA and vascular maturation. Blocking vascular maturation inhibits VMC recruitment and blocks NA differentiation of SN precursors. Inh…

Shy-Drager syndrome. Neuropathological correlation and response to levodopa therapy.

The post-mortem examination of the nervous system of a patient with Shy-Drager syndrome successfully treated with levodopa (Sharpe et al, 1972) revealed features of striato-nigral degeneration and amyotrophic lateral sclerosis, a cerebellar system degeneration and a loss of approximately 75% of sympathetic preganglionic neurons. Lewy bodies were not present and no detectable changes were observed in the sympathetic prevertebral ganglia. While the limited and transient beneficial effect of levodopa on the bradykinesia in our case is possibly due to the progressive loss of striatal dopaminergic

Increased Sympathetic Outflow Induces Adaptation to Acute Experimental Pain.

BACKGROUND: There are interrelationships between the autonomic nervous system and pain. This study aims to explore the effect of different autonomic manipulations on pain perception and modulation. METHODS: Twenty healthy subjects (10 men and 10 women, mean age 25 ± 3 years) participated in this single-blinded, semi-randomized, controlled study, which included 2 study visits. Warm detection thresholds, heat pain thresholds, conditioned pain modulation (CPM), and pain adaptation were tested before and after administration of phenylephrine, clonidine, yohimbine, and saline. RESULTS: Changes in

Autonomic nervous system activity changes in patients with hypertension and overweight: role and therapeutic implications.

The incidence and prevalence of hypertension is increasing worldwide, with approximately 1.13 billion of people currently affected by the disease, often in association with other diseases such as diabetes mellitus, chronic kidney disease, dyslipidemia/hypercholesterolemia, and obesity. The autonomic nervous system has been implicated in the pathophysiology of hypertension, and treatments targeting the sympathetic nervous system (SNS), a key component of the autonomic nervous system, have been developed; however, current recommendations provide little guidance on their use. This review discusse…

[Variability of sinus rhythm and cardiac failure].

Under pathological conditions, the autonomic nervous system, which continually regulates cardiac function, may have adverse effects on pump function and electrical activation. The sinus rhythm is an indirect marker of the different components of the autonomic nervous system and changes in the heart rate reflect the permanent variation of vagal and sympathetic tone. Analysis of variability in the temporal or frequential domaines, provides information about the modulation of the different components of the autonomic nervous system. In the post-infarction period, studies have shown that the heart

Anti-Leu 19 monoclonal antibody detects an antigen on autonomic nerves in human skin.

The monoclonal antibody anti-Leu 19 recognizes the NKH-1 antigen of active natural killer cells. In immunohistological preparations of the skin, the antibody reacts with cutaneous nerves and labels nerve fibers in the region of the sweat glands, the blood vessels and the hair arrector muscles. This staining pattern corresponds to the distribution of the autonomic sympathetic nervous system of the skin. On the other hand, anti-Leu 7 labels a myelin-associated glycoprotein which is to be found in the myelin sheath of sensory cutaneous nerves. The reactivity of neural structures of the skin can t…
