Calcific uraemic arteriolopathy: an update.

PURPOSE OF REVIEW: Calcific uraemic arteriolopathy (CUA) or calciphylaxis is a rare but important cause of morbidity and mortality in patients with chronic kidney disease. The prevalence of CUA is increasing in patients with renal failure, and the condition is also being recognized in nonuraemic patients. RECENT FINDINGS: There has been increasing understanding of the molecular basis of vascular calcification, in particular on the important role of the uraemic microenvironment in the factors implicated in the differentiation of vascular smooth muscle cells into osteoblasts. New options for tr

COBALT COMPOUNDS AS ANTIDOTES FOR HYDROCYANIC ACID.

The antidotal potency of a cobalt salt (acetate), of dicobalt edetate, of hydroxocobalamin and of cobinamide against hydrocyanic acid was examined mainly on mice and rabbits. All the compounds were active antidotes for up to twice the LD50; under some conditions for larger doses. The most successful was cobalt acetate for rabbits (5xLD50), which was effective at a molar cyanide/cobalt (CN/Co) ratio of 5, but had as a side-effect intense purgation. Hydroxocobalamin was irregular in action, but on the whole was most effective for mice (4.5xLD50 at a molar ratio of 1), and had no apparent side ef

Synthesis of glucose oxidase and catalase by Aspergillus niger in resting cell culture system.

The synthesis of glucose oxidase and catalase by Aspergillus niger was investigated using a resting cell culture system without growth being established. Calcium carbonate induced the synthesis of both enzymes and calcium chloride inhibited it. The optimal pH for the biosynthesis of glucose oxidase and catalase was 6.0 and 5.7, respectively. The effects of other bivalent cations, reductive compounds and metabolic products on enzyme synthesis were also tested. The biosynthesis of glucose oxidase and catalase was promoted by MnCO3, thioglycolic acid, pyroracemic acid and gluconic acid.

Salicylate inhibits the renal transport of inorganic sulfate in rat membrane vesicle preparations.

Salicylic acid (SA) administration produces a pronounced enhancement of the renal clearance of inorganic sulfate in rats. The purpose of the present investigation was to determine if SA inhibits the renal transport of inorganic sulfate in rat kidney cortex brush border membrane (BBM) and basolateral membrane (BLM) vesicle preparations. Sodium-dependent cotransport of inorganic sulfate was examined in BBM, and the values for KM and Vmax, determined using nonlinear regression analysis, were 0.52 +/- 0.41 mM and 2.84 +/- 1.26 nmol/mg protein/10 sec, respectively (N = 7). Increasing use of azoles combined with corticosteroids in Denmark.

INTRODUCTION: Calcinosis cutis (CC), the deposit of insoluble calcium salts in the skin and subcutaneous tissues, is a rare condition that occurs in autoimmune connective tissue diseases, including systemic sclerosis (SSc), dermatomyositis and in non-connective tissue diseases (non-CTD), particularly in patients with calciphylaxis and nephropathy, and in chronic ulcers. CC causes significant morbidity and impacts quality of life. This study evaluated CC management strategies of patients in a clinical setting. METHODS: A retrospective descriptive study was conducted at a university dermatology

First detection of Vibrio vulnificus and other potentially pathogenic Vibrio species on different types of waste material in the Gulf of Gdańsk.

BACKGROUND: Vibrio bacteria, including pathogens like Vibrio vulnificus (V. vulnificus) and Vibrio cholerae (V. cholerae) (typical for tropical regions), are increasingly reported in the Baltic Sea, with rising infections linked to climate change. In our study, we examined the presence of Vibrio species on waste materials floating in the coastal zone. MATERIAL AND METHODS: Seventy-five waste items were collected. Samples were rinsed, homogenized, and cultured on CHROMagar Vibrio and thiosulfate-citrate-bile salts-sucrose (TCBS) agar. Vibrio isolates were tested for their tolerance to salinity

Gut sulfide metabolism modulates behavior and brain bioenergetics.

The host-microbiome interface is rich in metabolite exchanges and exquisitely sensitive to diet. Hydrogen sulfide (H2S) is present at high concentrations at this interface and is a product of both microbial and host metabolism. The mitochondrial enzyme, sulfide quinone oxidoreductase (SQOR), couples H2S detoxification to oxidative phosphorylation; its inherited deficiency presents as Leigh disease. Since an estimated two-thirds of systemic H2S metabolism originates in the gut, it raises questions as to whether impaired sulfide clearance in this compartment contributes to disease and whether it

Hydrogen sulfide aggravates neutrophil infiltration, vascular remodeling and elastase-induced abdominal aortic aneurysm in male mice.

BACKGROUND: Abdominal aortic aneurysm (AAA) has an 80% mortality rate upon rupture, with no pharmacological treatments available to slow its progression. Hydrogen sulfide (H₂S), produced by cystathionine γ-lyase (CSE), has anti-inflammatory and antioxidant properties, but its role in AAA remains unclear. METHODS: We evaluated the impact of sodium thiosulfate (STS), a clinically relevant H₂S donor, in a periadventitial elastase-induced AAA model in normotensive male wild-type and Cse-/- mice. Complementary in vitro studies were conducted on primary human vascular smooth muscle cells (VSMCs) to

Hydrogen sulfide-dependent activation of human sulfide quinone oxidoreductase.

Hydrogen sulfide (H2S) is a respiratory poison and also a product of our own metabolism. The toxicity of H2S is mitigated by the activity of mitochondrial sulfide quinone oxidoreductase (SQOR), which oxidizes H2S while concomitantly reducing coenzyme Q. An unusual cysteine trisulfide cofactor distinguishes SQOR from other members of the flavin disulfide reductase superfamily. In the opening step of the catalytic cycle, an estimated 105-fold rate enhancement is afforded by nucleophilic addition of the sulfide anion to the trisulfide versus a disulfide cofactor. The source of the bridging sulfan

Probiotic acoustic biosensors for noninvasive imaging of gut inflammation.

Inflammatory bowel diseases (IBD) affect millions of people globally, result in severe symptoms, and are difficult to diagnose and monitor - often necessitating the use of invasive and costly methods such as colonoscopies or endoscopies. Engineered gut bacteria offer a promising alternative due to their ability to persist in the gastrointestinal (GI) tract and sense and respond to specific environmental signals. However, probiotics that have previously been engineered to report on inflammatory and other disease biomarkers in the Gl tract rely on fluorescent or bioluminescent reporters, whose s

Rapid Improvement of Refractory Cutaneous Hypereosinophilic Syndrome with Abrocitinib Plus Methylprednisolone: A Case Report.

Hypereosinophilic syndrome (HES) is a rare disorder characterized by persistent eosinophilia and end-organ damage. Cutaneous manifestations are frequently refractory to conventional therapies, including corticosteroids and immunosuppressants, creating a significant clinical challenge. The Janus kinase (JAK)-STAT pathway has been implicated in eosinophil activation and survival, suggesting a potential role for JAK inhibitors in management. In this context, we present the case of a 58-year-old female with a 10-year history of refractory generalized erythema, papules, and severe pruritus progress

Global survey of hgcA-carrying genomes in marine and freshwater sediments: Insights into mercury methylation processes.

Mercury (Hg) methylation is a microbially mediated process that produces methylmercury (MeHg), a bioaccumulative neurotoxin. A highly conserved gene pair, hgcAB, is required for Hg methylation, which provides a basis for identifying Hg methylators and evaluating their genomic composition. In this study, we conducted a large-scale omics analysis in which 281 metagenomic freshwater and marine sediment samples from 46 geographic locations across the globe were queried. Specific objectives were to examine the prevalence of Hg methylators, to identify horizontal gene transfer (HGT) events involving
