The present work examines the capacity of biological systems to encode memories via adaptive changes in molecular networks. In single cells, the rewiring of molecular networks can store information as molecular engrams and instantiate learning processes. In multicellular organisms, single cells might communicate with each other and tune their molecular memories to cooperatively encode multicellular memories in tissues and organs. Learning in the whole brain might consequently be examined as a problem of individual cells learning how to form a memory together by tuning their single-cell memorie

T-cell lymphoblastic lymphoma (T-LBL) is the second most common subtype of non-Hodgkin lymphoma (NHL) in children and adolescents. T-LBL shares considerable biological and clinical features with T-cell acute lymphoblastic leukaemia (T-ALL). The two entities are clinically distinguished by the extent of bone marrow (BM) involvement at diagnosis, with T-ALL defined by ≥25% blasts in the BM. Although the WHO classifies T-LBL and T-ALL as a single entity, emerging data suggest relevant clinical and molecular differences. This review focuses on T-LBL in paediatric patients, summarizing current know

Wall-climbing robots (WCRs) using numerous attachment/grasping mechanisms replace humans in executing repetitive or challenging tasks in space-confined, high-risk, or radioactive environments, garnering substantial research interest. Nevertheless, their application remains limited to environment- and surface-specific scenarios. To this end, we present a contact-adaptable, peeling-resistant, and cross-medium WCR integrated with gecko- and octopus-inspired self-adaptive rigid-soft hybrid tracks. The hollow mushroom-shaped adhesive microstructures (HMSAMSs) on the robot tracks simultaneously coup

The complex and dynamic relationship between plasticity and genetic adaptation in response to a changing environment represents a longstanding and controversial debate in evolutionary biology. In particular, the molecular and cellular mechanisms underlying this relationship have not been explored. Here, we conducted a plain-to-plateau animal translocation experiment using sheep as a model. We obtained brain, heart and lung tissues from normoxia-adapted, normoxia-to-hypoxia translocated and hypoxia-adapted sheep. We generated 27 scRNA-seq and 54 snRNA-seq datasets for tissues from 27 animals an

Semi-occluded vocal tract exercises (SOVTE) are vocal exercises involving partial occlusion in the front part of the vocal tract, often through phonation into a narrow tube. Although SOVTE are widely used in voice therapy and pedagogy, the physiological mechanisms underlying their therapeutic benefits remain unclear. In this computational study, voice production at different vocal fold configurations with and without semi-occlusion was investigated in a three-dimensional model. Semi-occlusion significantly increased phonation threshold pressure under conditions of weak vocal fold adduction. Un

Tropical rivers in Australia and New Guinea (Sahul) provide a rare natural experiment in vertebrate evolution: Unlike other continental systems, their freshwater ichthyofaunas are composed almost entirely of marine-derived lineages rather than primary freshwater fishes. This unique biogeographic setting enables replicated tests of why some marine-to-freshwater transitions give rise to extensive adaptive radiations whereas others remain species-poor, and whether these outcomes reflect ecological opportunity or temporally structured paleoenvironmental constraints. Using a densely sampled, time-c

Various environmental conditions force viruses to continuously evolve to survive. Evolving viruses with improved fitness are subject to positive selection and will pass on their genetic information to the next generation. If virus populations experience similar environmental pressure, they may undergo a dynamic process of molecular adaptation, which is known as convergent or parallel evolution (parallelism). Noteworthy, these phenomena are among the underlying mechanisms of cross-species transmission and emergence of novel viruses in the human population with a significant impact on global hea

Understanding how life is adapting to urban environments represents an important challenge in evolutionary biology. In this work, we investigate a widely cited example of urban adaptation, Culex pipiens form molestus, also known as the London Underground mosquito. Population genomic analysis of ~350 contemporary and historical samples counters the popular hypothesis that molestus originated belowground in London <200 years ago. Instead, we show that molestus first adapted to human environments aboveground in the Mediterranean or Middle East over the course of more than 1000 years, possibly in

BACKGROUND: SARS-CoV-2 infection is associated with a significant risk of hospitalisation, death, and prolonged impact on quality of life. Evaluation of new treatment options and optimising therapeutic management of people hospitalised with SARS-CoV-2 infection remains essential, but rapid changes in pandemic conditions and potential therapies have limited the utility of traditional approaches to randomised controlled trials.
METHODS: ASCOT ADAPT is an international, investigator-initiated, adaptive platform, randomised controlled trial of therapeutics for non-critically ill patients hospital

Deficits in the ankle plantarflexor muscles, such as weakness and contracture, occur commonly in conditions such as cerebral palsy, stroke, muscular dystrophy, Charcot-Marie-Tooth disease, and sarcopenia. While these deficits likely contribute to observed gait pathologies, determining cause-effect relationships is difficult due to the often co-occurring biomechanical and neural deficits. To elucidate the effects of weakness and contracture, we systematically introduced isolated deficits into a musculoskeletal model and generated simulations of walking to predict gait adaptations due to these d

Hypoxia-inducible factors (HIFs) are master transcription factor regulating hypoxic responses in vertebrates. Species of Schizothoracine, a sub-family of cyprinidae, are highly endemic to the hypoxic Qinghai-Tibetan Plateau (QTP). What roles the HIFs play in hypoxic adaptation in the Schizothoracine fish is little known. In this study, the HIF-1α/B gene from Gymnocypris dobula (Gd) was characterized. The predicted protein for Gd-HIF-1α/B contains the main domains (bHLH, PAS, PAC, ODD, N-TAD, and C-TAD). Moreover, a specific mutation that the proline hydroxylation motif (LXXLAP) mutated into Px

Probiotics are living microorganisms that are increasingly used as gastrointestinal therapeutics by virtue of their innate or engineered genetic function. Unlike abiotic therapeutics, probiotics can replicate in their intended site, subjecting their genomes and therapeutic properties to natural selection. We exposed the candidate probiotic E. coli Nissle (EcN) to the mouse gastrointestinal tract over several weeks, systematically altering the diet and background microbiota complexity. In-transit EcN accumulates genetic mutations that modulate carbohydrate utilization, stress response, and adhe

Hippos are considered the closest living relatives to cetaceans and they have some similar adaptations for aquatic living, such as a modified respiratory tract. Behavioral observations of male and female common hippos (Hippopotamus amphibius) at Disney's Animal Kingdom® and the Adventure Aquarium were conducted to describe and examine movements of the nostrils during respiration (inspiration, expiration, and inter-breath interval). The hippo nostril is a crescent shaped opening with lateral and medial aspects that are mobile and can be adducted and abducted to regulate the nostril opening. Not

Proline accumulation plays an important role in the response and adaptation of plants to abiotic stress. Gaseous signaling molecules such as nitric oxide (NO), carbon monoxide (CO), and hydrogen sulfide (H2S) are involved in complicated events of cell signaling. However, the regulatory mechanisms of gaseous signaling molecules on proline synthesis and degradation are still unclear. This review summarized the biosynthesis and degradation of proline. The role of gaseous signaling molecules and their cross-talk on proline metabolic regulation in plants was discussed along with the future perspect

Respiratory infectious diseases are the third cause of worldwide death. The nasopharynx is the portal of entry and the ecological niche of many microorganisms, of which some are pathogenic to humans, such as Neisseria meningitidis and Moraxella catarrhalis. These microbes possess several surface structures that interact with the actors of the innate immune system. In our attempt to understand the past evolution of these bacteria and their adaption to the nasopharynx, we first studied differences in cell wall structure, one of the strongest immune-modulators. We were able to show that a modific

Candida albicans is an important human fungal pathogen. Resistance to all major antifungal agents has been observed in clinical isolates of Candida spp. and is a major clinical challenge. The rise and expansion of drug-resistant mutants during exposure to antifungal agents occurs through a process of adaptive evolution, with potentially complex population dynamics. Understanding the population dynamics during the emergence of drug resistance is important for determining the fundamental principles of how fungal pathogens evolve for resistance. While few detailed reports that focus on the popula

During the evolution of hominids, childhood and adolescence have been added as new life-history phases. The transition from infancy to childhood (ICT) confers a predictive adaptive response to energetic cues that strongly influence adult height, whereas the transition from juvenility to adolescence establishes longevity and the age of fertility. Evolutionary short-term adaptations to energy crises apparently use epigenetic mechanisms that defer the ICT, culminating in short stature. The study of hunter-gatherers gives us an indication of pre-demographic transition populations and their life st

We developed a microwell plate, high-throughput, screening method aimed at quantitating the tolerance of a panel of Gram-positive and Gram-negative bacteria to metals (Frankia sp., Escherichia coli, Cupriavidus metallidurans, Rhizobium leguminosarum, and Streptomyces scabies). Microbial viability was quantified using MTS; a tetrazolium salt converted to a water-soluble formazan through microbial reduction. In this paper, we present the stepwise development of the method, highlighting the main elements underlying its reliability, and compare results obtained with literature. We conclude the met

BACKGROUND: Sleeping sickness due to Trypanosoma brucei (T.b.) gambiense is still a major public health problem in some central African countries. Historically, relapse rates around 5% have been observed for treatment with melarsoprol, widely used to treat second stage patients. Later, relapse rates of up to 50% have been recorded in some isolated foci in Angola, Sudan, Uganda and Democratic Republic of the Congo (DRC). Previous investigations are not conclusive on whether decreased sensitivity to melarsoprol is responsible for these high relapse rates. Therefore we aimed to establish a parasi

Genotype-specific sensitivity of the hepatitis C virus (HCV) to interferon-ribavirin (IFN-RBV) combination therapy and reduced HCV response to IFN-RBV as infection progresses from acute to chronic infection suggest that HCV genetic factors and intrahost HCV evolution play important roles in therapy outcomes. HCV polyprotein sequences (n = 40) from 10 patients with unsustainable response (UR) (breakthrough and relapse) and 10 patients with no response (NR) following therapy were identified through the Virahep-C study. Bayesian networks (BNs) were constructed to relate interrelationships among H
