First report of cattle farms with gastrointestinal nematodes resistant to levamisole in Mexico.

The objectives of the present study were: (1) to report the percentage of cattle farms with gastrointestinal nematodes (GINs) resistant to l … should be improved. None of the farmers weighed their animals in order to dose them correctly with anthelmintics. Six cattle farms (54.5%) applied anthelmintics to new arriving animals. This is the first report of levamisole resistant GINs in Mexico. Improving the use of anthelmintics and measures of quarantine for infected cattle will help control the spread …

Targeted supplementation with bioactive plants sustainably improves goat health and decreases antiparasitic drug use on smallholder farms.

Goats play a significant role in farming communities in semi-arid tropical areas with limited cropping capacity; however, production is limited by endoparasites, especially gastrointestinal nematodes (GINs). Control of GINs is typically mediated by anthelmintic drugs, but can be costly or ineffective where anthelmintic-resistant GINs are present. To manage anthelmintic resistance and improve herd health at lower costs, farmers can implement Targeted Selective Treatment (TST) strategies to treat animals based on performance or health traits. The study aimed to quantify the impacts of plant-base

The situation in the chemotherapy of nematode infections. in both human and veterinary medicine, is currently satisfactory. There are differ … ne is, however, governed by the great resistance problem in some animal hosts against all important anthelmintics. There are various reports of resistance against benzimidazoles, levamisole, pyrantel, and macrocyclic lactones in trichostrongylides in small ruminants (sheep, goats) and also Strongylides in horses. Resistance against ivermectin is now arising in …

The potential for histone deacetylase (HDAC) inhibitors as cestocidal drugs.

BACKGROUND: Echinococcosis and cysticercosis are neglected tropical diseases caused by cestode parasites (family Taeniidae). Not only there is a small number of approved anthelmintics for the treatment of these cestodiases, but also some of them are not highly effective against larval stages, such that identifying novel drug targets and their associated compounds is critical. Histone deacetylase (HDAC) enzymes are validated drug targets in cancers and other diseases, and have been gaining relevance for developing new potential anti-parasitic treatments in the last years. Here, we present the a…

Effects of selected Palestinian plants on the in vitro exsheathment of the third stage larvae of gastrointestinal nematodes.

BACKGROUND: Gastrointestinal parasites are one of the main restrictions to small ruminant production. Their pathological importance is primarily related to the major production losses, in quantity or quality, induced by the direct action of worms. Control of these parasites is based exclusively on the frequent use of anthelmintic drugs. However, the resistance to anthelmintics in worm populations after commercialisation of chemical drugs is now widespread. Therefore, there is a need to find new natural resources to ensure sustainable and effective treatment and control of these parasites. The …

Which is the best phenotypic trait for use in a targeted selective treatment strategy for growing lambs in temperate climates?

Targeted selective treatment (TST) requires the ability to identify the animals for which anthelmintic treatment will result in the greatest benefit to the entire flock. Various phenotypic traits have previously been suggested as determinant criteria for TST; however, the weight gain benefit and impact on anthelmintic efficacy for each determinant criterion is expected to be dependent upon the level of nematode challenge and the timing of anthelmintic treatment. A mathematical model was used to simulate a population of 10,000 parasitologically naïve Scottish Blackface lambs (with heritable var

Lessons from the History of Ivermectin and Other Antiparasitic Agents.

The twentieth century's arsenal of chemical anthelmintics brought manifold improvement in human health and, more abundantly, in animal health. The benefits were not only in health per se but also in agricultural economics, livestock management, and the overall production of food and fiber to support expanding human populations. Nevertheless, there remains (due in large part to drug resistance and paucity of available vaccines) a great need for new means of controlling disease caused by parasitic worms. Prudence should persuade us to look to our past for lessons that might help in our quest for…

Changes in the activity of glutathione-S-transferase in muscles and sera from mice infected with Trichinella spiralis after treatment with albendazole and levamisole.

In this study, changes in GST activity before and after treatment with antiparasitic drugs were measured in muscles and sera from mice infected with Trichinella spiralis between the 2nd and 9th weeks post-infection (wpi). Levamisole and albendazole, two anthelmintics with a different mode of action, were used. An about twofold stimulation of GST activity in the 2nd wpi and in the later muscular phase (6-7 wpi) was observed in muscles from animals treated with albendazole. In addition, a statistically significant stimulation of GST activity was observed after levamisole treatment, however, only…

The impact of albendazole treatment on the incidence of viral- and bacterial-induced diarrhea in school children in southern Vietnam: study protocol for a randomized controlled trial.

BACKGROUND: Anthelmintics are one of the more commonly available classes of drugs to treat infections by parasitic helminths (especially nematodes) in the human intestinal tract. As a result of their cost-effectiveness, mass school-based deworming programs are becoming routine practice in developing countries. However, experimental and clinical evidence suggests that anthelmintic treatments may increase susceptibility to other gastrointestinal infections caused by bacteria, viruses, or protozoa. Hypothesizing that anthelmintics may increase diarrheal infections in treated children, we aim to e…

Massive proteinuria: a possible side effect of pyrantel pamoate?

Drug-induced renal injury represents a frequent clinical entity. The most common drugs associated with acute tubular necrosis are aminoglycosides, amphotericin B, radiocontrast agents, and cyclosporine, but no data exist about the potential renal toxicity due to anthelmintics administration. Anthelmintics are commonly considered quite safe agents, and side effects such as gastrointestinal, neurologic, hematologic, or hepatic injury have been only rarely described. We report a 4-year-old boy with persistent massive proteinuria without any other symptoms/signs suggesting nephrotic syndrome (NS).…

Fungus-benzimidazole interactions: a prerequisite to deploying egg-parasitic fungi Paecilomyces lilacinus and Verticillium chlamydosporium as biocontrol agents against fascioliasis and amphistomiasis in ruminant livestock.

In vitro trials investigating the effects of albendazole and triclabendazole anthelmintics on the growth profiles of the egg-parasitic fungi Paecilomyces lilacinus and Verticillium chlamydosporium were undertaken. In addition, in vivo trials were conducted in goats fed on millet grain cultures of each fungus and administered albendazole and triclabendazole anthelmintics. In vitro growth revealed V. chlamydosporium to be more sensitive to albendazole compared to P. lilacinus. In contrast, triclabendazole had the least inhibitory effect on in vitro growth of both P. lilacinus and V. chlamydospor…

Modelling Cooperia oncophora: Quantification of key parameters in the parasitic phase.

Cooperia oncophora is one of the most common intestinal nematodes in cattle. It is also the dose-limiting species for the most frequently used anthelmintics, and consequently, the species usually involved in reports of anthelmintic resistance. However, little information is available on its population dynamics, hindering the parameterisation of transmission models to support understanding of the impact of anthelmintic resistance, climate change and alternative control strategies on nematode epidemiology. This systematic review and meta-analysis provides estimates for key life history traits of…

Endoparasite control practices on Saskatchewan dairy farms.

A questionnaire was administered to dairy producers in Saskatchewan in 2016 to determine basic pasture management practices, how producers use parasite control products, and attitudes towards the threat of endoparasites. All 161 dairy producers in Saskatchewan were invited to participate and the survey response rate was 39.8% (64/161). Most respondents (78.3%) were not concerned with endoparasites in their cattle or the threat of anthelmintic resistance. Yet 79.7% of producers reported using anthelmintics in all classes of cattle (lactating cows, dry cows, weaned heifers, and bred heifers). Th…

Transcriptional responses in Parascaris univalens after in vitro exposure to ivermectin, pyrantel citrate and thiabendazole.

BACKGROUND: Parascaris univalens is a pathogenic parasite of foals and yearlings worldwide. In recent years, Parascaris spp. worms have developed resistance to several of the commonly used anthelmintics, though currently the mechanisms behind this development are unknown. The aim of this study was to investigate the transcriptional responses in adult P. univalens worms after in vitro exposure to different concentrations of three anthelmintic drugs, focusing on drug targets and drug metabolising pathways. METHODS: Adult worms were collected from the intestines of two foals at slaughter. The fo…

Can efficient management of sheep gastro-intestinal nematodes be based on random treatment?

Targeted selective treatment has been proposed as a method to reduce gastro-intestinal nematode infections in ruminants and lower the selective pressure that leads to anthelmintic resistance. Since nematodes are highly aggregated within their host population, treating only the most heavily infected hosts offers an efficient and sustainable strategy to reduce infection within the flock and slow the spread of anthelmintic resistance. Although effective methods to correctly identify such hosts are available, their feasibility is often limited in the field conditions. Instead, treating part of the

The significance of the gastrointestinal parasites of Asian buffalo in Sri Lanka.

Buffalo aged 3-15 months on institutional farms and in the villages of an irrigation project were monitored for gastrointestinal nematodes by faecal egg counts over two years. Apart from treatment for Toxocara vitulorum at 10-16 days of age, no anthelmintics were used. Half of the strongylid egg counts were zero and 90% were less than 500 epg of faeces on the institutional farms, and 67% were zero and 97% less than 500 epg in the villages. No problems arose during four years without anthelmintics on the institutional farms and during two years in the villages. Autopsies demonstrated that the e…

Australian guidelines for equine internal parasite management.

Over the past few decades, the emergence of resistance amongst intestinal parasites of horses to all available anthelmintic classes has emphasised the need for a paradigm shift in parasite control approaches within the Australian equine industry. Findings of a recent Australia-wide research project have provided new insights into intestinal parasites (i.e. strongyles and ascarids) and parasite control from the perspectives of Australian horse breeders and equine veterinarians. The published data have revealed recent trends in parasite prevalence and distribution, breeders' and veterinarians' a
