Muscarinic receptor binding and behavioral effects of atropoine following chronic catecholamine depletion or acetylcholinesterase inhibition in rats.

Rats were subjected to one of two experimental treatments: (1) intraventricular infusion of the catecholamine neurotoxin 6-hydroxydopamine (6-OHDA), known to permanently reduce brain dopamine and norepinephrine levels, or (2) chronic administration of the irreversible acetylcholinesterase inhibitor diisopropylfluorophosphate (DFP). Both treatments are believed to produce relative overactivity of cholinergic systems and to suppress forward locomotion. The anticholinergic agent atropine sulfate yielded excessive forward walking in otherwise chronically akinetic 6-OHDA-treated rats, whereas atrop…

Conventional occlusion versus pharmacologic penalization for amblyopia.

BACKGROUND: Amblyopia is defined as impaired visual acuity in one or both eyes without demonstrable abnormality of the visual pathway, and is not immediately resolved by wearing glasses. OBJECTIVES: In performing this systematic review, we aimed to synthesize the best available evidence regarding the effectiveness and safety of conventional occlusion therapy compared to atropine penalization in treating amblyopia. SEARCH METHODS: We searched the Cochrane Central Register of Controlled Trials (CENTRAL) (which contains the Cochrane Eyes and Vision Trials Register) (2018, Issue 8); Ovid MEDLINE…

Acetylcholine exerts inhibitory and excitatory actions on mouse ileal pacemaker activity: role of muscarinic versus nicotinic receptors.

The effect of acetylcholine (ACh) on pacemaking and spontaneous contractions in the gastrointestinal tract is not well characterized. The cu … tials. The inhibitory effect of ACh was mimicked by donepezil (300 μM) but not nicotine (0.3-7 mM). Atropine (300 μM), but not hexamethonium (300 μM), reversed the inhibitory actions of ACh and bethanechol and revealed excitatory properties manifested as increases in pacemaker frequency. A spatial analysis also revealed that atropine, but not hexamethonium …

Advances in myopia control strategies for children.

Myopia has long been a global threat to public health. Timely interventions are likely to reduce the risk of vision-threatening complications. There are both established and rapidly evolving therapeutic approaches to slow myopia progression and/or delay its onset. The effective methods for slowing myopia progression include atropine eye-drops, defocus incorporated multiple segments (DIMS) spectacle lenses, spectacle lenses with highly aspherical lenslets target (HALT), diffusion optics technology (DOT) spectacle lenses, red light therapy (RLT), multifocal soft contact lenses and orthokeratolog…

[Comparative refractioning in juvenile strabismus patients. Experience with the Auto-Refractor 6600 (author's transl)].

122 strabismus patients have been refracted in order to compare the cycloplegic effect of Cyclopentolat-HCl (Zyklolat) and atropin. In young patients the cycloplegic effect of Zyklolat is not as sufficient as that of atropin is. Moreover the positions of cylindrical axes show differences up to 30 degrees under Zyklolat in comparison with atropin. On the other hand we give a report of our experience using the Auto-Refractor 6600, the before-mentioned measurings were performed with. -- The important field of the Auto-Refractor is the routine refractioning of co-operative patients. [Cycloplegia with cyclopentolate for testing-refraction of children (author's transl)].

A comparative study of the cycloplegic effect of 1% cyclopentolate and atropine or homatropine was done by means of Lindner's method of retinoscopy. In 53% of 1.5- to 6-year-old strabismic patients the hypermetropic refraction was higher (average 0.6 dpt) under the use of atropine, and in 27% of 6- to 11-year-old children cyclopentolate was more effective than homatropine (average 0.5 dpt). Therefore, atropine should be used for children with convergent squint up to 6 years, in order to get full correction of hypermetropia. In other cases cyclopentolate is preferable because of its rapid and s…

The effect of the parasympathetic autonomic nervous system on auditory fatigue.

Volunteers with normal hearing were tested for vegetative balance with the aid of vegetative reflexes and with the aid of the atropine test. The effect of white noise stimulated on hearing thresholds was then investigated together with their recovery. The vegetative system was affected experimentally by intravenous administration of atropine, and the beginning and recession of hearing fatigue was observed. Atropine caused a small change only. In a similarly arranged experiment, 1% pilocarpin administered subcutaneously in a dose of 1.4 minus 1.6 ml, resulted in increased hearing fatigue and re…

Meal-induced insulin sensitization and its parasympathetic regulation in humans.

In animal studies, the whole-body glucose disposal effect of insulin is low in the fasted state or after atropine infusion, but doubles after a meal, consistent with the hepatic insulin-sensitizing substance (HISS) hypothesis. We tested how a standardized test meal and atropine affected the dynamic response to insulin in humans. Insulin sensitivity was assessed in healthy male subjects (aged 28.9 +/- 1.9 years, body mass index 23.3 +/- 0.8 kg.m-2) by using the rapid insulin sensitivity test (RIST), which is a transient euglycemic clamp. After a 24-hour fasting period, dynamic insulin sensitivi…

Acetylcholine receptors do not mediate the immobilization produced by inhaled anesthetics.

UNLABELLED: Acetylcholine receptors transmit excitatory impulses, are broadly distributed throughout the central nervous system, and are particularly sensitive to the depressant effects of inhaled anesthetics. Thus these receptors are potential mediators of the immobility produced by inhaled anesthetics. We tested this potential in rats by giving intraperitoneal atropine, scopolamine, and mecamylamine to block muscarinic (atropine and scopolamine) and neuronal nicotinic (mecamylamine) acetylcholine receptors. Block with scopolamine (up to 100 mg/kg), atropine (10 mg/kg), mecamylamine (up to 4 …

Properties of muscarinic receptors mediating second messenger responses in the rabbit aorta.

The second messenger response of cultured smooth muscle and endothelial cells of the rabbit aorta to acetylcholine (ACh) was investigated. ACh induced a concentration-dependent accumulation of inositol-monophosphate (InsP1) in the smooth muscle cells and a concentration-dependent reduction in basal production of adenosine 3',5'-cyclic monophosphate (cAMP) in the endothelial cells. Atropine, scopolamine and nitric oxide (NO) inhibited the ACh-induced accumulation of InsP1. The IC50 values were 55 +/- 4.3 nM, 81.2 +/- 6.5 microM and 13.3 +/- 3.5 microM, respectively. On the other hand, the inhib…

Cholinergic neurons mediate intestinal propulsion in the rat.

Cholinergic influences on intestinal propulsion were determined in vivo in fasted rats by measuring the movement of a nonabsorbable radioactive marker along the intestine following treatment with cholinergic drugs. The marker was instilled directly into the intestine via a previously implanted cannula. The direct effects of cholinergic drugs on intestinal contractions were determined in vitro using isolated segments of duodenum and jejunum. Neostigmine (0.1 mg/kg) produced a marked increase in intestinal transit that was blocked by atropine pretreatment (1.0 mg/kg) but not hexamethonium pretre…

Circadian pattern of plasma 17-hydroxycorticosteroid: alteration by anticholinergic agents.

Atropine, administered to cats just prior to the time of the expected circadian rise in levels of 17-hydroxycorticosteroid in plasma, blocks this rise. Atropine does not alter this circadian pattern when administered at other times in the circadian cycle. Results similar to those obtained with atropine have been observed with short-acting barbiturates. Dibenzyline administered just prior to the time of the expected circadian rise is ineffective in blocking this rise. These findings support the hypothesis that the circadian pattern of plasma 17-hydroxycorticosteroid levels reflects activation, …

Pylorospasm: a less common functional disorder mimicking hypertrophic pyloric stenosis, leading to persistent vomiting in a preterm male infant.

INTRODUCTION AND IMPORTANCE: Historically, pylorospasm is defined as a condition in which the pyloric canal of the infant has a functional obstruction as opposed to an organic one, compared to hypertrophic pyloric stenosis (HPS), a well-known cause of post-feeding and repetitive vomiting in infancy. CASE PRESENTATION: We have reported a case of a premature male infant born via surrogacy and fed from a milk bank, whose acute after-meal intermittent vomiting led to a pylorospasm diagnosis. He was successfully treated non-surgically by intravenous (IV) atropine and gained weight. CLINICAL DISCU…

Vago-vagal activation of naloxone-sensitive non-adrenergic, non-cholinergic jejunal contractions in the anaesthetized cat.

Experiments were performed on chloralose-anaesthetized cats, pretreated with guanethidine and with ligated adrenals and sectioned splanchnic nerves. The animals were paralysed and artificially ventilated. Jejunal motility was recorded by an intraluminal balloon, as volume changes at a constant pressure. Cervical or abdominal vagal afferent stimulation was performed before and after the administration of atropine. In both circumstances jejunal contractions were elicited. The latency of onset and the duration of the responses were significantly shorter before than after atropine. Strength-durati…

Protective effect of thiaton, an antispasmodic drug, against indomethacin-induced intestinal damage in rats.

The effect of thiaton [3-(di-2-thienylmethylene)-5-methyl-trans-quinolizidinium bromide], an antispasmodic drug, on indomethacin-induced intestinal damage was examined in rats. The animals were given indomethacin, s.c., and the intestinal mucosa was examined 24 h later. Thiaton or atropine was given s.c. twice, 30 min before and 8 h following indomethacin. Indomethacin caused intestinal damage, accompanied with increase in enterobacterial translocation as well as inducible nitric oxide synthase (iNOS) and myeloperoxidase (MPO) activities, and these changes were significantly prevented by suppl…
