Neurotological auditory brain stem response findings in human immunodeficiency virus-positive patients without neurologic manifestations.

Neurologic manifestations of human immunodeficiency virus (HIV) infection, rather than being a late complication of the disease, are principally correlated with the early central nervous system (CNS) localization of HIV. The CNS may be infected in the early stages of acquired immunodeficiency syndrome (AIDS) without evidence of neurologic disorders. Evoked potentials (visual, auditory, and somatosensory) and electronystagmographic test batteries have proven to be very sensitive in showing subclinical CNS disorders due to HIV. In this study, auditory brain stem response (ABR) and electronystagm…

Study of brain stem auditory-evoked potentials (BAEPs) and visual-evoked potentials (VEPs) in leprosy.

A study of brain stem auditory-evoked potentials (BAEPs) and visual-evoked potentials (VEPs) was done on 25 newly diagnosed patients with leprosy whose diagnosis was confirmed by skin biopsy. The results were compared with 25 age- and sex-matched healthy controls. In BAEPs the important observations were the prolonged latency of wave V in 13 (52%), delayed interpeak latency (IPL) of wave I-III in 5 (20%) cases, of wave III-V in 12/25 (40%), suggesting a conduction abnormality of the VIII cranial nerve in its peripheral part, in its nucleus and in its connection in the brain stem. In VEPs, a de…

Post-transcriptional control of tryptophan hydroxylase gene expression in rat brain stem and pineal gland.

We have cloned a segment of the rat tryptophan hydroxylase (TPH) gene and used it to investigate differences in mRNA levels in two tissues: brain stem and pineal gland. The cloned, 13-kb genomic region contains five exons corresponding to cloned TPH cDNA sequence. Most intron/axon junctions are homologous to those found in the closely related genes encoding tyrosine and phenylalanine hydroxylase. The gene segment contains a region of low complexity that is repeated in the genome, but the remainder of the segment is single-copy sequence. TPH mRNAs can be detected by RNase protection assays and …

Auditory nerve-brain stem evoked potentials in cats during manipulation of the cerebral perfusion pressure.

In order to study the effects of various degrees of cerebral ischemia on the auditory nerve-brain stem evoked potentials (BAEP), the cerebral perfusion pressure (CPP), defined as the difference between mean arterial blood pressure (MAP) and intracranial pressure (ICP), was systemically manipulated in anesthetized, paralyzed and ventilated cats. The CPP was varied by decreasing MAP, either by hemorrhage or by the infusion of a vasodilating drug, and elevating ICP by infusion of mock CSF into the cisterna magna, or by MAP depression and ICP elevation simultaneously. Even though the lower limit o…

[The role of brain stem lesions in cerebral circulatory disorders accompanying severe cranio-cerebral injuries].

Clinical and anatomical comparisons were undertaken in 23 cases of mortality due to severe craniocerebral injuries. The set of clinical examinations included circulation and brain gas exchange studies. Cerebral circulatory hypoxy was revealed in all the patients, but in cases of its compensated form all signs of brain stem injury were lacking, while in cases of its non-compensated form foci of brain stem injury could be revealed at autopsy. Timely elimination of brain stem compression in cases of its anatomic integrity preserved helps to convert the non-compensated form of cerebral circulatory…

Protocol for morphogen-guided differentiation of brain cell types using human induced pluripotent stem cells.

Modeling neurological disorders is challenging due to differing functional genomics and phenotypes among species. Here, we present a protocol for morphogen-guided differentiation of human induced pluripotent stem cells (hiPSCs) into microglia, astrocytes, and mixed cortical cultures (MCCs) for studying human brain disorders. We describe steps for enhancing microglial production using hypoxia and implementing quality-control measures for astrocyte and MCC differentiations. We detail knockout serum replacement procedures for serum-free astrocytes. This protocol enables cell-type-specific investi

YTHDF1-modified neural stem cells confer neuroprotection and promote functional recovery following traumatic brain injury.

Traumatic brain injury (TBI) is a significant contributor to global morbidity and mortality, with limited effective treatment options available. Neural stem cells (NSCs) have shown great potential in the treatment of TBI. However, the relatively low differentiation rate of neurons largely hinders the therapeutic efficacy of brain tissue repair. Here, we found that following TBI, the expression level of YTHDF1 in the hippocampus significantly increased and then decreased. Previous reports have also indicated that YTHDF1 mRNA is preferentially expressed in the mouse hippocampus, a key region inv

Brain pericytes derived from human pluripotent stem cells retain vascular and phagocytic functions under hypoxia.

BACKGROUND: The integrity and function of the blood‑brain barrier (BBB) are largely regulated by pericytes. Pericyte deficiency leads to BBB breakdown and neurological dysfunction in major neurological disorders including stroke and Alzheimer's disease (AD). Transplantation of pericytes derived from induced pluripotent stem cells (iPSC‑PC) has been shown to restore the BBB and improve functional recovery in mouse models of stroke and pericyte deficiency. However, the molecular profile and functional properties of iPSC‑PC under hypoxic conditions, similar to those found in ischemic and neurodeg

Bio-Orthogonal Engineering of Neural Stem Cells with Membrane-Bound Microsatellites for Enhanced Brain Repair.

Regulating the differentiation of implanted stem cells into neurons is crucial for stem cell therapy of traumatic brain injury (TBI). However, due to the migratory nature of implanted stem cells, precise and targeted regulation of their fate remains challenging. Here, neural stem cells (NSCs) are bio-orthogonally engineered with hyaluronic acid methacryloyl (HAMA) microsatellites capable of sustained release of differentiation modulators for targeted regulation of their neuronal differentiation and advanced TBI repair. By employing bio-orthogonal covalent reactions and optimizing the microsate

Relative biological effectiveness and neural stem cell fate in carbon ion-irradiated human brain organoids.

BACKGROUND AND PURPOSE: Heavy ion therapy offers a promising approach for treating brain tumors due to the high precision of ion beams and biological effectiveness. However, the mechanisms underlying contrast-enhanced brain lesions, which occur more frequently after ions than conventional X-rays, remain to be elucidated. MATERIALS AND METHODS: Cerebral organoids were generated from human embryonic stem cells and exposed to high-energy carbon-ions at early (day 20) and late (day 80) developmental stages. Analyses of gene expression were performed using RT-PCR analysis, and protein expression w

Streptococcus agalactiae strain COH1 transcriptome in association with stem cell-derived brain-like endothelial cells.

Streptococcus agalactiae (Group B Streptococcus) strain COH1 is a representative strain of serotype III, multi-locus sequence type 17, which is disproportionately associated with neonatal meningitis. Here we report the transcriptome of COH1 when interacting with human brain endothelial cells compared with COH1 alone.

Genetically engineered human induced pluripotent stem cells for the production of brain-targeting extracellular vesicles.

BACKGROUND: Extracellular vesicles (EVs) are cell-secreted membrane vesicles that have become a promising, natural nanoparticle system for delivering either naturally carried or exogenously loaded therapeutic molecules. Among reported cell sources for EV manufacture, human induced pluripotent stem cells (hiPSCs) offer numerous advantages. However, hiPSC-EVs only have a moderate ability for brain delivery. Herein, we sought to develop a stable hiPSC line for producing EVs with substantially enhanced brain targeting by genetic engineering to overexpress rabies viral glycoprotein (RVG) peptide fu

Coxsackievirus B3 infects and disrupts human induced-pluripotent stem cell derived brain-like endothelial cells.

Coxsackievirus B3 (CVB3) is a significant human pathogen that is commonly found worldwide. CVB3 among other enteroviruses, are the leading causes of aseptic meningo-encephalitis which can be fatal especially in young children. How the virus gains access to the brain is poorly-understood, and the host-virus interactions that occur at the blood-brain barrier (BBB) is even less-characterized. The BBB is a highly specialized biological barrier consisting primarily of brain endothelial cells which possess unique barrier properties and facilitate the passage of nutrients into the brain while restric

Endogenous Neural Stem Cell-induced Neurogenesis after Ischemic Stroke: Processes for Brain Repair and Perspectives.

Ischemic stroke is a very common cerebrovascular accident that occurred in adults and causes higher risk of neural deficits. After ischemic stroke, patients are often left with severe neurological deficits. Therapeutic strategies for ischemic stroke might mitigate neuronal loss due to delayed neural cell death in the penumbra or seek to replace dead neural cells in the ischemic core. Currently, stem cell therapy is the most promising approach for inducing neurogenesis for neural repair after ischemic stroke. Stem cell treatments include transplantation of exogenous stem cells but also stimulat

Establishment and characterization of human pluripotent stem cells-derived brain organoids to model cerebellar diseases.

The establishment of robust human brain organoids to model cerebellar diseases is essential to study new therapeutic strategies for cerebellum-associated disorders. Machado-Joseph disease (MJD) is a cerebellar hereditary neurodegenerative disease, without therapeutic options able to prevent the disease progression. In the present work, control and MJD induced-pluripotent stem cells were used to establish human brain organoids. These organoids were characterized regarding brain development, cell type composition, and MJD-associated neuropathology markers, to evaluate their value for cerebellar

Elevated Asparagine Biosynthesis Drives Brain Tumor Stem Cell Metabolic Plasticity and Resistance to Oxidative Stress.

Asparagine synthetase (ASNS) is a gene on the long arm of chromosome 7 that is copy-number amplified in the majority of glioblastomas. ASNS copy-number amplification is associated with a significantly decreased survival. Using patient-derived glioma stem cells (GSC), we showed that significant metabolic alterations occur in gliomas when perturbing the expression of ASNS, which is not merely restricted to amino acid homeostasis. ASNS-high GSCs maintained a slower basal metabolic profile yet readily shifted to a greatly increased capacity for glycolysis and oxidative phosphorylation when needed.

Human Pluripotent Stem Cell-Derived Neural Cells and Brain Organoids Reveal SARS-CoV-2 Neurotropism.

Neurological complications are common in patients with COVID-19. While SARS-CoV-2, the causal pathogen of COVID-19, has been detected in some patient brains, its ability to infect brain cells and impact their function are not well understood, and experimental models using human brain cells are urgently needed. Here we investigated the susceptibility of human induced pluripotent stem cell (hiPSC)-derived monolayer brain cells and region-specific brain organoids to SARS-CoV-2 infection. We found modest numbers of infected neurons and astrocytes, but greater infection of choroid plexus epithelial

Protective effects of mesenchymal stem cells on ischemic brain injury: therapeutic perspectives of regenerative medicine.

Cerebral ischemic injury as the main manifestation of stroke can occur in stroke patients (70-80%). Nowadays, the main therapeutic strategy used for ischemic brain injury treatment aims to achieve reperfusion, neuroprotection, and neurorecovery. Also, angiogenesis as a therapeutic approach maybe represents a promising tool to enhance the prognosis of cerebral ischemic stroke. Unfortunately, although many therapeutic approaches as a life-saving gateway for cerebral ischemic injuries like pharmacotherapy and surgical treatments are widely used, they all fail to restore or regenerate damaged neur

Time for Radical Changes in Brain Stem Nomenclature-Applying the Lessons From Developmental Gene Patterns.

The traditional subdivision of the brain stem into midbrain, pons, and medulla oblongata is based purely on the external appearance of the human brain stem. There is an urgent need to update the names of brain stem structures to be consistent with the discovery of rhomobomeric segmentation based on gene expression. The most important mistakes are the belief that the pons occupies the upper half of the hindbrain, the failure to recognize the isthmus as the first segment of the hindbrain, and the mistaken inclusion of diencephalic structures in the midbrain. The new nomenclature will apply to al…
