Binding mechanism and antagonism of the vesicular acetylcholine transporter VAChT.

The vesicular acetylcholine transporter (VAChT) has a pivotal role in packaging and transporting acetylcholine for exocytotic release, serving as a vital component of cholinergic neurotransmission. Dysregulation of its function can result in neurological disorders. It also serves as a target for developing radiotracers to quantify cholinergic neuron deficits in neurodegenerative conditions. Here we unveil the cryo-electron microscopy structures of human VAChT in its apo state, the substrate acetylcholine-bound state and the inhibitor vesamicol-bound state. These structures assume a lumen-facin

Role of the nicotinic acetylcholine receptor in Alzheimer's disease pathology and treatment.

Alzheimer's Disease (AD) is the major form of senile dementia, characterized by neuronal loss, extracellular deposits, and neurofibrillary tangles. It is accompanied by a loss of cholinergic tone, and acetylcholine (ACh) levels in the brain, which were hypothesized to be responsible for the cognitive decline observed in AD. Current medication is restricted to enhancing cholinergic signalling for symptomatic treatment of AD patients. The nicotinic acetylcholine receptor family (nAChR) and the muscarinic acetylcholine receptor family (mAChR) are the target of ACh in the brain. Both families of r

Nobiletin differentially regulates muscarinic and nicotinic receptor-mediated ERK signaling.

Nobiletin, a citrus polymethoxyflavone with neuroprotective properties, activates extracellular signal-regulated kinase 1/2 (ERK1/2); however, its effects on cholinergic receptor-mediated ERK signaling remain unclear. This study aimed to investigate the effects of nobiletin on the muscarinic and nicotinic receptor-dependent ERK signaling in neuronal nitric oxide synthase-expressing PC12 cells. The results showed that nobiletin inhibited acetylcholine- and bethanechol-induced ERK1/2 phosphorylation, an effect attenuated by increasing agonist concentrations, consistent with a competitive-like pa

Pathological Appearance of a Case of Preclinical Multiple System Atrophy: A Comparison With Advanced Cases.

We aimed to investigate the frequency of multiple system atrophy (MSA) in a large number of forensic autopsies and characterize the pathological appearance of preclinical MSA. We investigated a series of 1930 brains from forensic autopsies. In addition to performing immunohistochemistry for phosphorylated α-synuclein, the levels of 3 autonomic nervous system markers (catecholaminergic, serotonergic, and cholinergic) were used to assess the peripheral nerve (heart and superior cervical ganglion) and medulla oblongata. The results were compared to those of healthy control and Parkinson disease (

Light and electronmicroscopy of the bowel in an unusual form of Hirschsprung's disease.

In a histological and histochemical study of multiple biopsies of unaffected segments of the bowel from 15 patients with Hirschsprung's Disease (H.D.), the AChE or non-specific esterase and the NADPH tetrazolium reductase enzyme reactions proved to be useful in identification of myenteric plexus islands; and acid phosphatase for the delineation of individual neurones. In the affected segment (usually aganglionic), this myenteric plexus tissue was not reactive for esterase, but individual nerve fibres among muscle fibres of the two muscle coats showed the enzyme product in a third of the cases.

High-affinity choline uptake (HACU) and choline acetyltransferase (ChAT) activity in neuronal cultures for mechanistic and drug discovery studies.

Acetylcholine (ACh) is the neurotransmitter used by cholinergic neurons at the neuromuscular junction, in parasympathetic peripheral nerve terminals, and in important memory-related circuits in the brain, and takes part in other critical functions. ACh is synthesized from choline and acetyl coenzyme A by the enzyme choline acetyltransferase (ChAT). The formation of ACh in cholinergic nerve terminals requires the transport of choline into cells from the extracellular space and the activity of ChAT. High-affinity choline uptake (HACU) represents the majority of choline uptake into the nerve term…

Choline Ameliorates Disease Phenotypes in Human iPSC Models of Rett Syndrome.

Rett syndrome (RTT) is a postnatal neurodevelopmental disorder that primarily affects girls. Mutations in the methyl-CpG-binding protein 2 (MECP2) gene account for approximately 95 % of all RTT cases. To model RTT in vitro, we generated induced pluripotent stem cells (iPSCs) from fibroblasts of two RTT patients with different mutations (MECP2 (R306C) and MECP2 (1155Δ32)) in their MECP2 gene. We found that these iPSCs were capable of differentiating into functional neurons. Compared to control neurons, the RTT iPSC-derived cells had reduced soma size and a decreased amount of synaptic input, ev

A dominant mutation in a neuronal acetylcholine receptor subunit leads to motor neuron degeneration in Caenorhabditis elegans.

Inappropriate or excessive activation of ionotropic receptors can have dramatic consequences for neuronal function and, in many instances, leads to cell death. In Caenorhabditis elegans, nicotinic acetylcholine receptor (nAChR) subunits are highly expressed in a neural circuit that controls movement. Here, we show that heteromeric nAChRs containing the acr-2 subunit are diffusely localized in the processes of excitatory motor neurons and act to modulate motor neuron activity. Excessive signaling through these receptors leads to cell-autonomous degeneration of cholinergic motor neurons and para

Barbiturate anaesthesia reduces the neurotoxic effects of quinolinate but not ibotenate in the rat pedunculopontine tegmental nucleus.

We have previously demonstrated that ibotenate (IBO) injected into the pedunculopontine tegmental nucleus (PPTg) damages all neurones there while quinolinate (QUIN) makes relatively selective lesions of cholinergic neurones. We now compare the effects of two anaesthetics, sodium pentobarbitone and Avertin (tribromoethanol/tert-amylalcohol dissolved in ethanol, saline and phosphate buffer) on three doses of IBO and QUIN in the PPTg. Diaphorase-positive cell loss after QUIN was attenuated under barbiturate, the relative selectivity of QUIN for diaphorase-positive neurones was lost and lesion vol

[Effect of serotonin on the contractile activity of the cat duodenum].

In cats, distention of an isolated segment of the ileum with a saline injection elicited its contraction which could be abolished with blockade of muscarinic receptors. Serotonine acting from the ileum lumen enhanced the response, whereas acting from the vascular bed induced a contractile response which could be decreased with the beta-adrenoreceptor-blocking agent propranolol. The findings suggest a possibility of the serotonine participation in realisation of peristaltic reflex, the character of its modulating effect being dependent on activation of excitatory cholinergic and inhibitory nona

Cholinergic stimulation of the rat hypothalamus: effects of liver glycogen synthesis.

Cholinergic stimulation of the lateral hypothalamic neurons with intrahypothalamic microinjections of acetylcholine or carbachol caused a marked increase in the content of the active form of glycogen (starch) synthase in the liver. Total activity of the enzyme (active plus inactive forms) was not increased significantly. The lowest effective dose of acetylcholine was 5 X 10(-10) mole, and the optimum dose was 5 X 10(-9) mole. Similar applications of other neurotransmitters, such as norepinephrine, dopamine, serotonin, and gamma-aminobutyric acid, did not affect the enzyme's activity.

Striatal astrocytes transdifferentiate into functional mature neurons following ischemic brain injury.

To determine whether reactive astrocytes stimulated by brain injury can transdifferentiate into functional new neurons, we labeled these cells by injecting a glial fibrillary acidic protein (GFAP) targeted enhanced green fluorescence protein plasmid (pGfa2-eGFP plasmid) into the striatum of adult rats immediately following a transient middle cerebral artery occlusion (MCAO) and performed immunolabeling with specific neuronal markers to trace the neural fates of eGFP-expressing (GFP(+)) reactive astrocytes. The results showed that a portion of striatal GFP(+) astrocytes could transdifferentiate

Enteric botulinum neurotoxins facilitate infection by Salmonella and Shigella.

Botulism is a life-threatening disease caused by botulinum neurotoxins (BoNTs) released by Clostridium bacteria. BoNTs induce a flaccid paralysis by blocking neurotransmitter release from peripheral cholinergic neurons via soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) cleavage. The enteric nervous system (ENS) controls antimicrobial defense reactions within the intestine through neuron-released mediators. Here, we show that subclinical doses of BoNT/A and BoNT/B, which are not sufficient to cause botulism when released in the intestinal lumen, paralyze ENS neuro…

lynx1, an endogenous toxin-like modulator of nicotinic acetylcholine receptors in the mammalian CNS.

Elapid snake venom neurotoxins exert their effects through high-affinity interactions with specific neurotransmitter receptors. A novel murine gene, lynx1, is highly expressed in the brain and contains the cysteine-rich motif characteristic of this class of neurotoxins. Primary sequence and gene structure analyses reveal an evolutionary relationship between lynx1 and the Ly-6/neurotoxin gene family. lynx1 is expressed in large projection neurons in the hippocampus, cortex, and cerebellum. In cerebellar neurons, lynx1 protein is localized to a specific subdomain including the soma and proximal

Light-modulated neural control of sphincter regulation in the evolution of through-gut.

The development of a continuous digestive tract, or through-gut, represents a key milestone in bilaterian evolution. However, the regulatory mechanisms in ancient bilaterians (urbilaterians) are not well understood. Our study, using larval sea urchins as a model, reveals a sophisticated system that prevents the simultaneous opening of the pylorus and anus, entry and exit points of the gut. This regulation is influenced by external light, with blue light affecting the pylorus via serotonergic neurons and both blue and longer wavelengths controlling the anus through cholinergic and dopaminergic

Distribution of choline acetyltransferase and acetylcholinesterase in the vocal motor system of zebra finches.

The distribution of choline acetyltransferase immunoreactivity (ChAT-IR) was surveyed in the vocal motor system of adult male and female zebra finches and was compared with the pattern of histochemical acetylcholinesterase (AChE-His). In the vocal motor system the most prominent accumulation of ChAT-IR somata was found in lobus parolfactorius (LPO) including Area X. Immunoreactive neuropil was found to be concentrated in pericellular networks of fibers in male's Area X while the corresponding area in females could not be demarcated within the LPO. The density of ChAT-IR fiber networks was much

Stem cells and derived growth factors in Alzheimer's disease: mechanistic insights and translational perspectives.

Alzheimer's disease (AD) is a significant healthcare concern due to the complex interactions between amyloid-β accumulation, tau pathology, neuroinflammation, synaptic dysfunction, and progressive neuronal death. Stem-cell-based therapies have demonstrated potential to treat this complex condition through immunomodulation, trophic support, and neuroregeneration. Beyond direct cell replacement, stem-cell-derived growth factors have important therapeutic functions by providing strong neurotrophic and cytoprotective effects. These bioactive compounds improve neuronal survival, restore synaptic pl
