Patterns of alcohol use and response to digital brief interventions in college students: a secondary analysis of a cluster randomized trial.

BACKGROUND: Drinking among college students in India is rising, influenced by social and cultural shifts. This study aimed to classify drinking behaviors among underage students using Latent Class Analysis (LCA) and assess the effectiveness of digital screening and brief intervention (DSBI) and brief advice (DSBA) across identified classes. METHODS: This was a secondary analysis of a two-stage cluster randomized trial that screened 693 college students (age 18-22 yrs) with the Alcohol Use Disorders Identification Test (AUDIT). LCA was employed to identify latent classes in a main sample (N =

Evidence for a ∼43 GeV γ-ray Line Signal in a Stacking Analysis of the Virgo, Fornax, and Ophiuchus Galaxy Clusters.

As the largest gravitationally bound objects in the Universe, galaxy clusters have provided the first piece of evidence for the presence of dark matter and may be suitable targets for indirect dark matter searches. Among various signals, the GeV-TeV γ-ray line has been taken as the smoking-gun signal of the dark matter annihilation and decay, since no known astrophysical or physical process(es) could generate such a peculiar spectrum. With 15.5 years of Fermi-LAT P8R3 publicly available data, we search for the γ-ray line emission in the directions of 13 massive galaxy clusters at redshifts z≤0

Analysis of Kinase-Peptide Interactions: DGMM Latent Constructs and Clustering Towards New Groups and Stand-Out Structures.

Protein kinases are among the largest families of enzymes in eukaryotes and have been intensively studied as important cellular components and medicinal targets. In this study, interface properties for all experimental high-resolution human protein kinase domain-peptide complexes from the protein data bank (PDB) were calculated, and analyzed statistically. The structures encompassed all the six most populous human kinase groups, and the majority of peptides were found binding either solely to the kinase C lobe or both/between lobes. Analyses of the interface properties revealed numerous unexpe

Identification and clustering analysis of drug-responsive temporally varying genes through high-frequency longitudinal RNA sequencing.

High-frequency longitudinal RNA sequencing has emerged as a powerful approach for capturing dynamic transcriptional responses to therapeutic interventions, yet traditional differential expression analysis fails to identify genes with temporal variability that lack static expression differences. We used our previously developed computational framework for identifying Temporally Varying Genes (TVGs) from daily blood samples collected over 10-21 days in Sprague-Dawley rats treated with hepatotoxic compounds including tetracycline, isoniazid, carbon tetrachloride, and valproate. Our methodology em

Chemical Clustering Analysis of Ambient and Emission Source Particulate Matter Reveals Compositional Determinants of Pulmonary Toxicity Responses.

Comparative toxicological studies of heterogeneous particulate matter (PM) samples are needed to evaluate the influence of particle chemistry on pulmonary toxicity outcomes. Here, groups of mice were exposed by oropharyngeal aspiration of a 100 μg dose of one of seven PM samples, including three coarse and two fine ambient air PM, and 2 fine emission source PM. Acute inflammatory and lung injury markers in the bronchoalveolar lavage fluid (BALF) were assessed. A weighted chemical correlation network analysis (WCCNA) clustered PM chemical constituents into four modules based on comodulation wit

Unsupervised clustering analysis reveals distinct postoperative cortisol trajectories following pituitary adenoma resection in Cushing's disease.

PURPOSE: Cushing’s disease (CD), caused by ACTH-secreting pituitary adenomas, results in hypercortisolism. Transsphenoidal surgery (TSS) is the first-line treatment. Early postoperative cortisol trends are frequently monitored, though standard interpretation is lacking. We sought to determine whether early postoperative cortisol trajectories are associated with biochemical remission at 1 year using unsupervised machine learning. METHODS: We retrospectively reviewed 94 adult patients with CD who underwent TSS between 2011 and 2021 at two academic centers. Serum cortisol was measured every 6 h,

Demixing and Analysis of Complex Biological Raman Hyperspectra Based on Peak Fitting, Amplitude Trend Clustering, and Spectrum Reconstruction.

To better interpret the Raman spectra from mammalian cells, it is often desirable to reduce their complexity by decomposing them into the spectral contributions from individual macromolecules or types of macromolecules. Diverse methods exist for demixing complex spectra, each with different benefits and drawbacks. However, some methods require a library of component spectra that might not be available, while others are hampered by noise and peak congestion that includes many proximal overlapping peaks. Through rapid fitting of individual peaks in every spectrum of a Raman hyperspectral data se

Design of an improved method for cardiac MRI lesion analysis using CMFT-GAN and spatio-temporal clustering process.

Early and precise diagnosis is difficult due to the limitations of cardiac MRI lesion analysis, which include low spatial resolution, undersampled datasets, and insufficient lesion diversity. Because of poor feature regularization, insufficient spatiotemporal dynamics modeling, and a lack of pathology-aware augmentation, current systems frequently generate high false positives. We present a thorough enhancement and clustering framework that reinterprets lesion analysis in cardiac MRI in order to get past these obstacles. Our method combines motion-aware lesion consistency, synthesizes various

Unbiased cell clustering analysis of vaccine-induced T cell responses in the Imbokodo HIV-1 vaccine trial.

BACKGROUND: HIV-1 T cell responses are associated with viral control and may be protective in a prophylactic vaccination setting. Traditional methods for analysing these responses might be biased towards specific functionalities or epitopes. This study presents an unsupervised and unbiased clustering analysis workflow, using the Leiden algorithm followed by selection of antigen-specific clusters using MIMOSA positivity calls, for high-dimensional flow cytometry data to identify distinct T cell populations associated with protection in the HVTN 705/HPX2008/Imbokodo HIV-1 vaccine efficacy trial.

Comprehensive analysis of traumatic rupture of the aorta in road traffic crashes: incorporating epidemiological insights and K-prototype clustering.

OBJECTIVES: The identification of crash characteristics associated with traumatic rupture of the aorta (TRA) can significantly enhance countermeasures against TRA. Conventional epidemiological approaches struggle to adequately handle the substantial variability of traffic crash data. Consequently, this study aims to integrate conventional epidemiological analysis with data-driven cluster analysis to more comprehensively analyze TRA-related crash characteristics. METHODS: A total of 350 unweighted TRA crashes were extracted from traffic crash databases including comprehensive crash details and…

Symptom clusters and their impact on quality of life in multiple myeloma survivors: Secondary analysis of BMT CTN 0702 trial.

Autologous haematopoietic cell transplantation (autoHCT) and continuous post-transplant maintenance therapy are the standard of care in transplant-eligible multiple myeloma (MM) patients. We sought to describe symptom burden and identify symptom clusters occurring in MM patients after autoHCT using data from the BMT CTN 0702 randomized controlled trial comparing the outcomes of three treatment interventions after an autoHCT in 758 MM patients. We analysed individual transplant-related symptoms assessed via the FACT-BMT questionnaire at enrolment and annually for 4-year post-autoHCT. We also de

Multi-omics analysis of the gut microbiome and metabolites associated with the psychoneurological symptom cluster in children with cancer receiving chemotherapy.

BACKGROUND: Children with cancer receiving chemotherapy commonly report a cluster of psychoneurological symptoms (PNS), including pain, fatigue, anxiety, depression, and cognitive dysfunction. The role of the gut microbiome and its functional metabolites in PNS is rarely studied among children with cancer. This study investigated the associations between the gut microbiome-metabolome pathways and PNS in children with cancer across chemotherapy as compared to healthy children. METHODS: A case-control study was conducted. Cancer cases were recruited from Children's Healthcare of Atlanta and hea
