Intermittent physical stress during early- and mid-adolescence differentially alters rats' anxiety- and depression-like behaviors in adulthood.

Prior work showed that exposing rats to stress from weaning through to late adolescence (PD23-51) increased anxiety- and depression-related responses in adulthood. In the current study, we tested the hypothesis that the outcome of adolescent stress depends on the specific timing of adversity in adolescence. Male and female rats were exposed to intermittent, physical stress during either early (PD22-33) or mid -(PD35-46) adolescence, and then their anxiety- and depression-related responses to acute stressors were tested in adulthood. Early adolescent stress decreased rats' open-arm exploration

Post-weaning isolation promotes food intake and body weight gain in rats that experienced neonatal maternal separation.

Neonatal maternal separation (MS) in rats has been reported to result in permanent dysfunctions of the hypothalamic-pituitary-adrenal axis a … sed immobility in forced swim test, compared to NH pups, regardless of their housing conditions. In elevated plus maze test, group-caged MS pups spent less time in the open arms and more time in the closed arms than group-caged NH pups, but social isolation did not further affect the arm stay of MS pups. However, statistical analyses revealed an interaction between MS an …

Cachexia in mice blunts improved cognitive flexibility induced by calorie restriction.

Approximately 50-80 % of cancer patients suffer from cachexia, a metabolic syndrome involving inflammation, appetite loss, and muscle and fat wasting. Another common co-morbidity of cancer patients is cognitive impairment, and clinical evidence suggests the incidence of cachexia is linked to more severe cognitive symptoms. Given the difficulty of studying changes in cognitive function in human cancer patients, we set out to examine key aspects of cognitive performance in a mouse model of pancreatic cancer (pancreatic ductal adenocarcinoma; PDAC) cachexia, using an in-cage operant device (Feedi

Epigenetic regulation of the glucocorticoid receptor gene through methylation is linked to post-traumatic stress disorder.

Post-traumatic stress disorder (PTSD) emerges in a subset of individuals following exposure to traumatic events and is characterized by marked variability in susceptibility and resilience. Epigenetic mechanisms, particularly DNA methylation, have been implicated in psychiatric disorders; however, their specific role in regulating stress-related genes in the context of PTSD remains insufficiently understood. This study investigated the epigenetic regulation of the metabotropic glutamate receptor 5 (mGluR5) gene in the hippocampus and its association with individual differences in PTSD vulnerabi

Assessment of the anxiolytic, antidepressant, and antioxidant potential of Parquetina nigrescens (Afzel.) Bullock in Wistar rats.

ETHNOPHARMACOLOGICAL RELEVANCE: The recent growing concerns about the multisystemic nature of mental health conditions in the global population are facilitating a new paradigm involving alternative natural, nutritional, and complementary therapies. Herbal remedies despite accounts in literature of their ethnobotanical as alternative remedies for diverse ailments, remain underexplored for psychiatric disorders like anxiety, depression, and insomnia. AIM OF THE STUDY: Hence, the anxiolytic, antidepressant, and antioxidant properties of a hydro-ethanolic leaf extract of Parquetina nigrescens (PN

Amelioration of pain and anxiety in sleep-deprived rats by intra-amygdala injection of cinnamaldehyde.

BACKGROUND: Sleep disorders are accompanied by increased anxiety and somatic pain. In addition, it has been observed that anxiety and pain have a boosting effect on each other, resulting in continued sleep disturbances. Amygdala's (CeA) central nucleus plays a crucial role in these processes. Cinnamaldehyde (Cinn) is an aromatic compound with anti-anxiety, antioxidant, and sleep-promoting properties. The present study uses sleep-deprived rats to examine the effects of an intra-CeA injection of Cinn on pain and anxiety. METHODS: Sleep deprivation (SD) was induced using the platform technique.

Naringenin ameliorates neuropsychiatric deficits in mice fed a methionine-choline-deficient diet.

Neuropsychiatric dysfunction is increasingly being acknowledged as a disabling complication of non-alcoholic steatohepatitis (NASH), but there are no therapeutic approaches. We investigated in the present study the neuroprotective effectiveness of naringenin, a citrus flavonoid with known anti-inflammatory and neurotrophic effects, in a murine NASH model induced by an 8-week methionine-choline-deficient (MCD) diet. Male C57BL/6 mice (n = 8/group) were treated with naringenin (50 mg/kg/day, i.p.) during the final 4 weeks. In behavioral tests, naringenin counteracted cognitive impairment in nove

Biotransformation and its effects on anxiolytic activity and toxicity of Polygalae Radix saponins.

Saponins, accounting for over 2.0% (w/w) in Polygalae Radix extracts, act as both active and toxic constituents, with their glycosyl structures hypothesized to modulate efficacy and toxicity. This study aimed to modify the glycosyl structures of Polygalae Radix (PR) saponins via snail enzyme biotransformation and evaluate changes in the activity of inhibiting anxiety-like behavior in the EPM and gastrointestinal toxicity. The biotransformation process was optimized using one-way experiments and response surface methodology, identifying optimal conditions as 46 °C, pH 6.4, 72-h reaction time, a

Effect of gestational exposure to dim light at night on the behavior of rat dams and offspring.

Recently, dim light at night (dLAN) has become nearly ubiquitous. Whereas nighttime light exposure is associated with an increased risk of adverse health outcomes, little is known about the effects of low-intensity light exposure at night during pregnancy on offspring. We investigated the behavioral perturbations caused by prenatal exposure to dLAN at different stages of ontogeny. Rat dams were subjected to a light-dark regimen with ~ 2 lx dLAN throughout pregnancy. After parturition, the animals were exposed to a regular lighting cycle, and the dams' behavior in the homecage and the pups' dev

Role of opioid tapering in rescuing anxiety and social novelty deficits in morphine-withdrawn male rats.

Abstinence from morphine in patients with opioid use disorder leads to both physical and emotional withdrawal symptoms. A common treatment strategy involves administering long-acting opioids to alleviate these symptoms while gradually reducing the dose, helping patients adapt to morphine-free conditions. This study examined how tapering regimens of methadone, buprenorphine, or morphine affect anxiety-like behaviors and social functioning in morphine-dependent rats. Male Wistar rats were rendered morphine-dependent over a 10-day period and subsequently treated for 30 days with gradually decreas

Ketamine attenuates the effects of intermittent social defeat on anxiety, social interaction and cocaine-induced conditioned place preference in male mice.

Mice exposed to intermittent social defeat (ISD) stress in late adolescence exhibit short-term anxiety- and depression-like behaviours and demonstrate greater sensitivity to the rewarding effects of cocaine in adulthood. Furthermore, the development of depression-like symptoms predicts subsequent enhanced vulnerability to cocaine reward. The aim of this study was to investigate whether ketamine, a non-competitive glutamate N-methyl-D-aspartate (NMDA) receptor antagonist with antidepressant properties, could prevent the short-term effects of ISD on anxiety- and depression-like behaviours and th

Exercise mitigates a gut microbiota-mediated reduction in adult hippocampal neurogenesis and associated behaviours in rats.

Lifestyle factors, especially exercise, impact the manifestation and progression of psychiatric and neurodegenerative disorders such as depression and Alzheimer's disease, mediated by changes in hippocampal neuroplasticity. The beneficial effects of exercise may be due to its promotion of adult hippocampal neurogenesis (AHN). Gut microbiota has also been showed to be altered in a variety of brain disorders, and disturbances of the microbiota have resulted in alterations in brain and behaviour. However, whether exercise can counteract the negative effects of altered gut microbiota on brain func

The involvement of orexin 1 and cannabinoid 1 receptors within the ventrolateral periaqueductal gray matter in the modulation of migraine-induced anxiety and social behavior deficits of rats.

Orexin 1 receptors (Orx1R) and cannabinoid 1 receptors (CB1R) are implicated in migraine pathophysiology. This study evaluated the potential involvement of Orx1R and CB1R within the ventrolateral periaqueductal gray matter (vlPAG) in the modulation of anxiety-like behavior and social interaction of migraineurs rats. A rat model of migraine induced by recurrent administration of nitroglycerin (NTG) (5 mg/kg/i.p.). Infralimbic cortex controls fear memory generalization and susceptibility to extinction during consolidation.

Lesioning or inactivating the infralimbic (IL) subregion of the medial prefrontal cortex before acquisition produces more generalized and extinction-resistant fear memories. However, whether and how it modulates memory specificity and extinction susceptibility while consolidation takes place is still unknown. The present study aims to investigate these questions using muscimol-induced temporary inactivation and anisomycin-induced protein synthesis inhibition in the rat IL following contextual fear conditioning. Results indicate that the IL activity immediately after acquisition, but not six ho
