Pharmacokinetic modelling of [2-13C]uracil metabolism in normal and DPD-deficient dogs.

A physiologically based pharmacokinetic (PBPK) model to simulate the plasma concentration and 13CO2 exhalation after [2-13C]uracil administration to DPD-suppressed dogs was developed. Simulation using this PBPK model should be useful in clinical situations where DPD-deficient patients at risk are to be detected with [2-13C]uracil as an in vivo probe.

Auto-PEEP in respiratory failure.

Intrinsic positive end-expiratory pressure (auto-PEEP) is a common occurrence in patients with acute respiratory failure requiring mechanical ventilation. Auto-PEEP can cause severe respiratory and hemodynamic compromise. The presence of auto-PEEP should be suspected when airflow at end-exhalation is not zero. In patients receiving controlled mechanical ventilation, auto-PEEP can be estimated measuring the rise in airway pressure during an end-expiratory occlusion maneuver. In patients who trigger the ventilator or who are not connected to a ventilator, auto-PEEP can be estimated by simultaneo…

The antipyrine breath test in the rat: a pharmacokinetic model.

Breath tests have been widely advocated for use as non-invasive probes of mixed function oxidase activity in vivo. A catenary sequence of events begins with demethylation and results in the exhalation of 14CO2. Intermediates in this chain include formaldehyde and formate. In this current study [14C]-antipyrine, [14C]-formaldehyde and [14C]-formate have been administered to rats. The data from these one carbon intermediates lead to the conclusion that demethylation is not the rate-limiting step in the antipyrine breath test in the rat. The resultant 14CO2 exhalation rate time profiles have been…

Respiratory-Swallow Coordination and Its Relationship With Pharyngeal Residue, Penetration, and Aspiration in People With Parkinson's Disease.

PURPOSE: Respiratory-swallow coordination (RSC) frequently changes in people with Parkinson's disease (PwPD). Little is known about how these changes relate to impairments in swallowing safety (penetration and aspiration) and efficiency (pharyngeal residue). Therefore, the aims of this study were to assess the relationships between RSC, pharyngeal residue, penetration, and aspiration in PwPD. METHOD: Twenty-four PwPD were recruited to undergo simultaneous assessment of RSC, swallowing safety, and swallowing efficiency. RSC was assessed using respiratory inductive plethysmography and nasal air

[Importance and perspectives of breath analysis].

Quantitative chemical analysis of volatile constituents in exhaled gas can provide useful insights into biochemical processes in the body. Relations between the chemical composition of human exhaled air and inflammatory processes, states of high oxidative activity, inhalation or ingestion of various noxious substances as well as diseases like ARDS, pneumonia or sleep apnea have been described. Clinical interpretation of these findings, however, remains difficult because definite marker substances for certain diseases could not yet be identified, substance concentrations in the exhaled air chan

An Enhanced Mechanistic Model For Capnography, With Application To CHF-COPD Discrimination.

Capnography records CO2 partial pressure in exhaled breath as a function of time or exhaled volume. Time-based capnography, which is our foc … ed using capnogram-derived estimates of respiratory parameters in a simple mechanistic model of CO2 exhalation. We propose an enhanced mechanistic model that can capture specific capnogram characteristics in congestive heart failure (CHF) by incorporating a representation of the inertance associated with fluid in the lungs. Gene expression changes in blood RNA after swimming in a chlorinated pool.

Exposure to disinfection by-products (DBP) such as trihalomethanes (THM) in swimming pools has been linked to adverse health effects in humans, but their biological mechanisms are unclear. We evaluated short-term changes in blood gene expression of adult recreational swimmers after swimming in a chlorinated pool. Volunteers swam 40min in an indoor chlorinated pool. Blood samples were drawn and four THM (chloroform, bromodichloromethane, dibromochloromethane and bromoform) were measured in exhaled breath before and after swimming. Intensity of physical activity was measured as metabolic equival

Volume of rat lungs measured throughout the respiratory cycle using 19F NMR of the inert gas SF6.

The lung volumes of mechanically ventilated rats were measured over the course of the respiratory cycle using the NMR signal strength from inhaled sulfurhexafluoride. Rats with elastase-induced emphysema showed larger lung volumes and slower exhalation than control rats. For humans the technique should be able to provide lung volume measurements at least 20 times a second.

Practical ppb-Level Room Temperature Chemiresistive Nitric Oxide Sensing Assembly Based on Ultrathin Thick Porphyrin MOF Nanosheets.

Nitric oxide is an endogenous biological signaling molecule, and the corresponding fractional exhaled NO serves as an important indicator in … structure in the nanohybrid. The potential clinical utility of the obtained sensors is validated by exhalation tests toward exhalation samples from healthy individuals and asthma patients, respectively. This work provides an effective strategy of developing MOF-based room temperature ppb-level chemiresistive NO sensors for practical FeNO monitoring.

Brief structured respiration practices enhance mood and reduce physiological arousal.

Controlled breathwork practices have emerged as potential tools for stress management and well-being. Here, we report a remote, randomized, controlled study (NCT05304000) of three different daily 5-min breathwork exercises compared with an equivalent period of mindfulness meditation over 1 month. The breathing conditions are (1) cyclic sighing, which emphasizes prolonged exhalations; (2) box breathing, which is equal duration of inhalations, breath retentions, and exhalations; and (3) cyclic hyperventilation with retention, with longer inhalations and shorter exhalations. The primary endpoints…

Nasal administration of the nitric oxide synthase inhibitor L-NAME induces daytime somnolence.

In preliminary studies, human subjects complained of drowsiness after aerosolization of NG-nitro-L-arginine methyl ester (L-NAME) into the nasal passages. We compared the effects of a nasal aerosol of L-NAME (0.5 M, 4 ml) to those of saline on sleep onset latency and exhaled nasal nitric oxide (NO). L-NAME decreased sleep onset latency and exhaled nasal NO. Vasoconstriction and local effects of L-NAME on NO synthesis are unlikely to explain this effect since oxymetazoline, a vasoconstrictor, decreased exhaled NO but had no effect on sleep onset latency. We conclude that aerosolization of L-NAM

Fraction of air coming from conductive airways has the negative balance in heat dissipation after maximal effort exercise-a physiological basis for exercise-induced bronchoconstriction.

Exhaled breath temperature (EBT) has recently been used as a tool to assess the level of inflammation in airways. The newest technology can also assess EBT coming from different fractions of exhaled air (fEBT). We aimed to assess the changes in fEBT after a maximal cardio-pulmonary exercise test in healthy athletes. Forty-four healthy professional athletes (two females) were included. Their mean (±SD) age was 22.9 ± 4.8 years. An innovative device (FractAir®) was used for fEBT measurement, dividing the exhaled air into three fractions (V1, V2 and V3) coming from different parts of the lungs; t

Development of a Wearable Device for Precise Assessment of Particle Deposition in the Human Respiratory System.

The adverse respiratory health effects of particulate matter (PM) are well documented, yet real-world exposure and deposition within the human respiratory system remain poorly quantified. Here, we report a wearable device capable of simultaneously collecting PM from inhaled and exhaled breath to directly assess particle deposition under real-world conditions. Designed as a face mask, the device draws ambient air for inhalation while capturing exhaled aerosol, enabling direct comparison between inhaled and exhaled particles. Laboratory validation confirmed efficient collection of exhaled partic

[Effects of organohalogenated compounds on the emission of ethane and pentane in expired air].

Effects of two halogenated acetamides (chloroacetamide-iodoacetamide) and of three chlorinated volatile solvents on the ethane-pentane test, as an index of in vivo lipid peroxidation have been studied. Chloroacetamide stimulates alcanes production. Iodoacetamide acts only on pentane exhalation. The three chlorinated solvents seem to induce moderate lipoperoxidation, and the apparent increase of hydrocarbons might also result from decreased metabolism.

Performance evaluation of a recirculated personalized air curtain combined with displacement ventilation in mitigating indoor airborne transmission.

To mitigate the risk of airborne transmission, we explored the integration of a recirculated personalized air curtain (rPAC) with displaceme … raction from coughing and speaking, although its performance varied with respiratory activities and exhalation directions. At 0° and 30°, the rPAC reduced the intake fraction of small coughing droplets by over 90 % and medium droplets by 74 %-98 %, with significant reductions also observed for speaking. However, at 60°, the rPAC increased the intake fraction …

Ultrahigh humidity-resistance ppb-level formaldehyde sensing at room temperature induced by fluorinated dipole based "umbrella" and "bridge".

Formaldehyde (HCHO) is a major indoor pollutant that is extremely harmful to human health even at ppb-level. Meanwhile, ppb-level HCHO is also a potential disease marker in the exhalation of patients with respiratory diseases. Higher humidity resistance and lower practical limit of detection (pLOD) both have to be pursued for practical HCHO sensors. In this work, by assembling indium oxide (In2O3) and fluorinated dipole modified reduced graphene oxide (rGO), we prepared a high-performance room temperature HCHO sensor (In2O3 @ATQ-rGO). Excellent sensing properties toward HCHO under visible illu…

Numerical study on the heat-recovery capacity of the human nasal cavity during expiration.

The characteristics of the thermal field in the human nasal cavity during the expiration period were investigated using computational fluid dynamics. Heat and water-vapor recovery features were quantitatively investigated under realistic distributions of the epithelial surface and air temperature. A constant expiratory flow rate of 250 mL/s was assumed. The epithelial surface temperature was approximately 34.3-34.4 °C in the nasopharynx and 33.5-33.6 °C in the vestibule region, and these values are in good agreement with the measurement data in the literature. We observed that heat-recovery fr

Volatile organic compounds (VOCs) fingerprint of Alzheimer's disease.

Alzheimer's disease (AD) is a profoundly life changing condition and once diagnosis occurs, this is typically at a relatively late stage into the disease process. Therefore, a shift to earlier diagnosis, which means several decades before the onset of the typical manifestation of the disease, will be an important step forward for the patient. A promising diagnostic and screening tool to answer this purpose is represented by breath and exhaled volatile organic compounds (VOCs) analysis. In fact, human exhaled breath contains several thousand of VOCs that vary in abundance and number in correlat
