TGA-binding (TGA) transcription factors, characterized by the basic region/leucine zipper motif (bZIP), have been recognized as pivotal regulators in plant growth, development, and stress responses through their binding to the as-1 element. In this study, the TGA gene families in melon, watermelon, cucumber, pumpkin, and zucchini were comprehensively characterized, encompassing analyses of gene/protein structures, phylogenetic relationships, gene duplication events, and cis-acting elements in gene promoters. Upon transient expression in Nicotiana benthamiana, the melon CmTGAs, with typical bZI

In plants, systemic acquired resistance (SAR) is established as a result of NPR1-regulated expression of pathogenesis-related (PR) genes. Using gene expression profiling in Arabidopsis, we found that in addition to controlling the expression of PR genes, NPR1 also directly controls the expression of the protein secretory pathway genes. Up-regulation of these genes is essential for SAR, because mutations in some of them diminished the secretion of PR proteins (for example, PR1), resulting in reduced resistance. We provide evidence that NPR1 coordinately regulates these secretion-related genes t

Eukaryotic cold shock domain proteins are nucleic acid-binding proteins that are involved in transcription, translation via RNA chaperone activity, RNA editing, and DNA repair during tissue developmental processes and stress responses. Cold shock domain proteins have been functionally implicated in important developmental transitions, including embryogenesis, in both animals and plants. Arabidopsis thaliana cold shock domain protein 4 (AtCSP4) contains a well conserved cold shock domain (CSD) and glycine-rich motifs interspersed by two retroviral-like CCHC zinc fingers. AtCSP4 is expressed in

Calcium-dependent protein kinases (CDPKs) are crucial calcium sensors involved in plant responses to pathogen infection. Here, we report isolation and functional characterization of the pathogen-responsive rice OsCPK10 gene. The expression of OsCPK10 was strongly induced following treatment with a Magnaporthe grisea elicitor. Kinase activity assay showed that the functional OsCPK10 protein not only autophosphorylated, but also phosphorylated Casein in a calcium-dependent manner. Overexpression of constitutively active OsCPK10 in Arabidopsis enhanced the resistance to infection with Pseudomonas

In animals, plants and fungi, cholera toxin (CTX) can activate signalling pathways dependent on heterotrimeric GTP binding proteins (G-proteins). We transformed tobacco plants with a chimeric gene encoding the A1 subunit of CTX regulated by a light-inducible wheat Cab-1 promoter. Tissues of transgenic plants expressing CTX showed greatly reduced susceptibility to the bacterial pathogen Pseudomonas tabaci, accumulated high levels of salicylic acid (SA) and constitutively expressed pathogenesis-related (PR) protein genes encoding PR-1 and the class II isoforms of PR-2 and PR-3. In contrast, the

This paper reports the expression and localization of linamarase in roots of two cassava (Manihot esculenta Crantz) cultivars of low and high cyanide. Two different patterns of linamarase activity were observed. In the low-cyanide type, young leaves displayed very high enzyme activity during the early plant growing stage (3 months), whereas in root peel, the activity increased progressively to reach a peak in 11-month-old plants. Conversely, in the high-cyanide cultivar (HCV), root peel linamarase activity decreased during the growth cycle, whereas in expanded leaves linamarase activity peaked

Anthocyanins determine the appearance and nutritional quality of sweet cherries, yet their transport mechanism remains largely unclear. Here, we performed a genome-wide identification of the MATE family in sweet cherry and characterized two key anthocyanin-related transporters. A total of 54 PavMATE genes were identified and classified into five subgroups with distinct structures and expression patterns. Expression levels of PavMATE50 and PavMATE51 gradually increased with fruit ripening and color deepening. Subcellular localization revealed that PavMATE50 targeted the Golgi apparatus and PavM

Sodium hydrosulfide (NaHS), a hydrogen sulfide donor, has been reported to regulate postharvest senescence and quality maintenance in horticultural crops. However, its role in sand pear softening and the underlying physiological basis remain unclear. Mature fruits of the climacteric sand pear cultivar "Sucui No. 1" were treated with NaHS and stored at 20°C to evaluate its effects on postharvest quality, ethylene production, cell wall metabolism, and softening-related gene expression. Treatment with 1 mmol L-1 NaHS delayed firmness loss and reduced ethylene production, with limited effects on f

WRKY transcription factors (TFs) are important regulators of plant growth, stress responses, and secondary metabolism; however, their roles in endophyte-mediated dendrobine biosynthesis remain largely unknown in Dendrobium nobile. Here, we identified 63 DnWRKY genes in the D. nobile genome and systematically investigated their potential involvement in endophyte-mediated dendrobine accumulation. Phylogenetic analysis revealed high conservation of DnWRKY proteins within subfamilies. Endophytic fungal co-culture significantly promoted dendrobine accumulation, reaching 0.39 mg g-1 at 14 days, whic

Arsenic (As) contamination in paddy soils poses a threat to human health through rice consumption. Arsenate (AsV) uptake contributes to grain As accumulation, yet its upstream regulatory mechanisms remain largely unknown. Here, we identified a WRKY transcription factor, OsWRKY1, as a key regulator of AsV uptake in rice plant, whose expression is strongly induced by AsV at both transcript and protein levels. Loss of OsWRKY1 function increased rice root As concentration by 2.8-fold under hydroponic conditions and raised grain As concentration by 19-22% in soil-grown plants compared with wild typ

Geranylgeranyl diphosphate synthase (GGPS) is a key enzyme in terpenoid biosynthesis, but its role in Phoebe zhennan remains largely unknown. Here, 14 PzGGPS genes were identified and classified into three groups with conserved gene structures and motifs. Among them, PzGGPS12 was identified as a key gene for terpenoid biosynthesis, wood fragrance formation, and drought tolerance. Functional analyses showed that PzGGPS12 catalyzes the production of terpene precursors and promotes the accumulation of mono-, sesqui-, di-, and triterpenoids, thereby enhancing drought resistance through metabolic f

Transcriptional repression is a central mechanism regulating plant growth and development, yet how plant-specific corepressors achieve robust gene silencing remains unclear. Here, we identify the rice corepressor TPR2 (TOPLESS-RELATED 2) as a phase-separating protein that forms nuclear condensates through the cooperative action of an intrinsically disordered region (IDR1) and a plant-specific N-terminal tetramerization domain. Structure-guided mutagenesis disrupting tetramerization or deleting IDR1 markedly impaired condensate formation in vivo and in vitro. Complementation assays showed that

Cuticular wax covers the aerial surfaces of land plants and protects them from environmental stresses. Diketones (β-diketone and hydroxy-β-diketone) are the most important components of cuticular wax, and are mainly associated with the glaucous appearance in wheat (Triticum aestivum). The WAX1 (W1) locus consists of three genes, TaDMH, TaDMP, and TaDMC, which are reported to directly participate in biosynthesis of wheat wax diketones. However, the regulatory mechanism underlying diketone biosynthesis and formation of glaucousness is still largely unclear in wheat. Here, we tested the involveme

Cadmium (Cd) contamination in rice threatens food security and public health. Identifying and characterizing key genes governing Cd uptake and translocation is crucial for breeding low-Cd varieties. In this study, we characterized a Cd-responsive member of major facilitator superfamily (MFS), ABNORMAL SHOOT IN YOUTH (OsASY), which shares high sequence homology with the known Cd transporter OsCd1. Functional analysis revealed that OsASY localizes to the plasma membrane and functions as an influx transporter mediating Cd uptake and translocation. OsASY is constitutively expressed in rice, and it

Phosphate (Pi) deficiency impairs plant development and decreases productivity owing to its critical functions in terms of cell structure, metabolism, and photosynthesis. Here, we assessed the determinate growth program of Arabidopsis primary roots exposed to Pi scarcity to clarify the role of citrate and malate in the expression of the mitotic cyclin CycB1 and changes in the meristem, transition zone, and elongation zone. The transfer of seedlings from high-Pi medium to low-Pi medium, but not to high-Pi medium, inhibits root growth, cell division, and elongation, aspects that are restored upo

Tobacco root rot is a destructive soil-borne disease that constrains sustainable tobacco production in Yunnan. Fusarium oxysporum and Fusarium solani species complex are currently considered the principal pathogens associated with this disease. Therefore, effective biocontrol agents are needed, and understanding their pathogen-side modes of action is essential for improving biological management strategies. This study screened 100 bacterial strains isolated from the tobacco rhizosphere using plate confrontation assays and identified A5 as a broad-spectrum antagonistic strain. A5 consistently i

This review synthesizes the molecular mechanisms of cold tolerance in pepper, integrating multi-omics data, genome editing, and precision breeding strategies to accelerate the development of cold-resilient cultivars. Cold stress is a significant environmental factor that affects the growth, productivity, and fruit quality of Capsicum annuum by impairing membrane integrity photosynthesis and cellular redox homeostasis. Although pepper has several endogenous cold-responsive regulators such as CaNAC035 and CabHLH035, along with antioxidant defense systems, its cold tolerance remains limited due to

In most fleshy fruits, including apple, cherry, plum, and grape, citric acid content declines during the late stages of fruit development, a process that largely determines fruit flavor. In contrast, pear (Pyrus spp.) exhibits a less common developmental pattern characterized by continued citric acid accumulation during late fruit development. Although similar patterns have been reported in a limited number of other fruit species and germplasms, the molecular mechanisms underlying this developmental trajectory remain largely unknown. Here, comparative analyses of organic acid dynamics among cu
