Upon skin colonization, the prevalent human skin commensal S. epidermidis can elicit a CD8 + T cell response that protects against pathogens or clears tumors. The microbial features that drive this response are undefined, limiting our ability to understand and predict commensal-immune crosstalk and to engineer potent commensal-derived immunotherapies. To uncover these microbial features, we harnessed both the natural variation in CD8 + T cell induction across primary human isolates of Staphylococcus and our ability to genetically manipulate these strains. Stimulatory strains exhibit increased

Ovarian cancer remains one of the deadliest gynaecological cancers due to its often-late diagnosis and the absence of specific symptoms in the early stages. Beyond genetic alterations, the tumour microenvironment, including the vaginal microbiota, plays a decisive role in tumour progression. Recent studies have highlighted the involvement of microbial imbalance 'dysbiosis' in ovarian carcinogenesis, particularly through interactions with the immune system and the modulation of local inflammatory pathways such as Th17-related pathways. In a state of 'eubiosis,' the vaginal microbiota, dominated

Inflammatory bowel disease (IBD) is a complex chronic intestinal inflammatory disorder whose pathogenesis involves aberrant interactions between genetic, environmental, microbial, and immune factors. Neutrophils, as key effector cells of innate immunity, are among the first immune cells to infiltrate the inflamed mucosa in IBD, and their role in the disease course is multifaceted. This review systematically elaborates on the dual functions of neutrophils in IBD. On one hand, activated neutrophils act as crucial "destroyers" promoting the initiation and progression of IBD by releasing effector

Despite the availability of antiseizure medications (ASMs), approximately one-third of patients with epilepsy remain refractory to treatment. Evidence suggests that the gut microbiota modulates central nervous system function through neuroimmune and metabolic pathways. We characterized the fecal microbiome of patients with epilepsy and autoimmune encephalitis (AE) using 16S rRNA sequencing. Ninety-six participants from a tertiary hospital in Brazil were included: mesial temporal lobe epilepsy (MTLE; n = 38), genetic generalized epilepsy (GGE; n = 11), AE (n = 10), and healthy controls (n = 37)

Helicobacter pylori (H. pylori) infection is a global health problem that affects more than half of the world population and leads to gastrointestinal disorders such as gastric ulcer, duodenal ulcer and gastric cancer. The available treatment options are inefficient because of patient non-adherence to the regimen due to the polypharmacy approach of treating H. pylori over longer periods. The bio-availability of antibiotics at the gastric mucosa, where the organism resides, is also a major concern. The continuous adaptation of bacteria such as genetic mutation toward beta-lactamase, macrolide a

Beyond reproductive dysfunction, polycystic ovarian syndrome (PCOS) is a complex endocrine condition that significantly raises metabolic and cardiovascular morbidity in PCOSaffected individuals. There is growing evidence that the gut microbiota plays a vital role in the pathogenesis of PCOS and the cardiometabolic problems associated with it. This review examined the novel premise of the gut-heart-ovary axis, emphasising how gut microbial dysbiosis in PCOS interferes with critical processes related to inflammation, insulin resistance, lipid metabolism, and endothelial function, ultimately lead

Factor H (FH) is the main regulator of the complement alternative pathway, and it is essential to prevent complement-mediated autologous damage. Many pathogens express FH-binding proteins on their surface that recruit human FH as a strategy to avoid or limit their elimination by the host complement. The FH-Related Proteins (FHR-1, FHR-2, FHR-3, FHR-4A, and FHR-5) share sequential and structural homology with FH that result from their common ancestry, but none of them can regulate complement like FH. While the precise contribution of FHRs in human complement is still not completely understood,

Severe anaemia remains a major public health challenge, disproportionately affecting children and women of reproductive age in sub-Saharan Africa. In high-burden settings, the aetiology of severe anaemia is multifactorial, encompassing nutritional deficiencies, infections, and host genetic factors. Beyond its direct clinical consequences, severe anaemia is also associated with increased susceptibility to invasive bacterial infections, particularly those caused by enteric pathogens such as non-typhoidal Salmonella and Escherichia coli. In this review, we present an integrated framework linking

Ulcerative colitis (UC) is a chronic inflammatory bowel disease influenced by genetic, immune, microbial, and environmental factors, with diet emerging as a key modifiable element. This narrative review examines how specific macronutrients, food additives, and dietary patterns affect UC pathogenesis, symptom control, and clinical outcomes. High intake of saturated fats, red and processed meats, and ultra-processed foods promotes dysbiosis and mucosal inflammation, while diets rich in fiber, resistant starches, and omega-3 fatty acids support gut barrier integrity and exert anti-inflammatory ef

In the context of global climate warming, studies have yet to fully clarify how pollutants affect the gut microbiome and antibiotic resistance genes (ARGs) in nontarget soil fauna. This study investigates the interactive effects of pesticide exposure (imidacloprid) and elevated temperature on the gut bacterial community and ARGs in the model soil collembolan Folsomia candida. Our results demonstrate warming exacerbates the toxicity of imidacloprid in collembolans. While exposure to both warming and pesticide significantly altered the gut microbial composition of F. candida, impairing microbial

Plant-microbe partnerships constitute a complex and intricately woven network of connections that have evolved over countless centuries, involving both cooperation and antagonism. In various contexts, plants and microorganisms engage in mutually beneficial partnerships that enhance crop health and maintain balance in ecosystems. However, these associations also render plants susceptible to a range of pathogens. Understanding the fundamental molecular mechanisms governing these associations is crucial, given the notable susceptibility of plants to external environmental influences. Based on quo

Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly known as non-alcoholic fatty liver disease, has become the most common form of chronic liver disease in children. The spectrum of pediatric MASLD ranges from simple steatosis to steatohepatitis, fibrosis, cirrhosis, and in rare cases, hepatocellular carcinoma. Its pathogenesis involves a complex interplay among genetic, epigenetic, and environmental factors, along with alterations in the gut microbiota and its associated metabolites. Given the staggering prevalence and the distinct etiopathogenesis of pediatric MASLD, c

The global rise in inflammatory bowel disease (IBD) and other non-communicable diseases (NCDs) over the past five decades has coincided with the widespread adoption of a Western diet and lifestyle. These conditions, characterised by chronic inflammation, are shaped by complex interactions between genetic, environmental, immunological, and microbial factors. The Western diet rich in, refined sugars, unhealthy fats, ultra-processed foods and excess salt, is increasingly recognised as a major contributor to immune dysfunction, microbial dysbiosis, and compromised intestinal barrier integrity, all

Systemic lupus erythematosus (SLE) is an autoimmune disease characterized by chronic inflammation that affects multiple organs, with its prevalence varying by ethnicity. Intestinal dysbiosis has been observed in both SLE patients and murine models. Additionally, intestinal barrier impairment is thought to contribute to the ability of pathobionts to evade and breach immune defenses, resulting in antigen cross-reactivity, microbial translocation, subsequent immune activation, and, ultimately, multiple organ failure. Since the detailed mechanisms underlying these processes are difficult to examin

Survival strategy of bacteria is expanded by extrachromosomal elements (ECEs). However, their genetic diversity and functional roles for adaptability are largely unknown. Here, we discover a novel family of intracellular ECEs using 56 saliva samples by developing an efficient microbial DNA extraction method coupled with long-read metagenomics assembly. Even though this ECE family was not hitherto identified, our global prevalence analysis using 476 salivary metagenomic datasets elucidates that these ECEs reside in 74% of the population. These ECEs, which we named, "Inocles", are giant plasmid-

For forty generations, two lines of White Leghorn chickens have been selected for high (HAS) or low (LAS) antibody response to a low dose injection of sheep red blood cells (SRBCs). Their gut is home to billons of microorganisms and the largest number of immune cells in the body; therefore, the objective of this experiment was to gain understanding of the ways the microbiome may influence the differential antibody response observed in these lines. We achieved this by characterizing the small intestinal microbiome of HAS and LAS chickens, determining their functional microbiome profiles, and by

Methanobrevibacter smithii (M. smithii), initially isolated from human feces, has been recognised as a distinct taxon within the Archaea domain following comprehensive phenotypic, genetic, and genomic analyses confirming its uniqueness among methanogens. Its diversity, encompassing 15 genotypes, mirrors that of biotic and host-associated ecosystems in which M. smithii plays a crucial role in detoxifying hydrogen from bacterial fermentations, converting it into mechanically expelled gaseous methane. In microbiota in contact with host epithelial mucosae, M. smithii centres metabolism-driven micr

The increased spreading of antibiotic resistance among different infectious agents has been a fast-growing public health challenge worldwide; this is because of the discovery of new resistance mechanisms and the reduction in quality and effective treatments of general pathogenic infections. This has caused unsuccessful microbial responses to standard therapy, which could lead to a higher risk of mortality, prolonged illness, and more expenditures for health care. Most parasites, bacteria, fungi, and viruses can produce a higher degree of multidrug resistance (MDR) with increased mortality and
