Improved cognitive flexibility in serotonin transporter knockout rats is unchanged following chronic cocaine self-administration.

Cocaine dependence is associated with orbitofrontal cortex (OFC)-dependent cognitive inflexibility in both humans and laboratory animals. A critical question is whether cocaine self-administration affects pre-existing individual differences in cognitive flexibility. Serotonin transporter knockout (5-HTT(-/-) ) mice show improved cognitive flexibility in a visual reversal learning task, whereas 5-HTT(-/-) rats self-administer increased amounts of cocaine. Here we assessed: (1) whether 5-HTT(-/-) rats also show improved cognitive flexibility (next to mice); and (2) whether this is affected by co

The human oxidative DNA glycosylase NEIL1 excises psoralen-induced interstrand DNA cross-links in a three-stranded DNA structure.

Previously, we have demonstrated that human oxidative DNA glycosylase NEIL1 excises photoactivated psoralen-induced monoadducts but not genuine interstrand cross-links (ICLs) in duplex DNA. It has been postulated that the repair of ICLs in mammalian cells is mainly linked to DNA replication and proceeds via dual incisions in one DNA strand that bracket the cross-linked site. This process, known as "unhooking," enables strand separation and translesion DNA synthesis through the gap, yielding a three-stranded DNA repair intermediate composed of a short unhooked oligomer covalently bound to the d

Transitions at CpG dinucleotides, geographic clustering of TP53 mutations and food availability patterns in colorectal cancer.

BACKGROUND: Colorectal cancer is mainly attributed to diet, but the role exerted by foods remains unclear because involved factors are extremely complex. Geography substantially impacts on foods. Correlations between international variation in colorectal cancer-associated mutation patterns and food availabilities could highlight the influence of foods on colorectal mutagenesis. METHODOLOGY: To test such hypothesis, we applied techniques based on hierarchical clustering, feature extraction and selection, and statistical pattern recognition to the analysis of 2,572 colorectal cancer-associated

How well can we predict native contacts in proteins based on decoy structures and their energies?

One strategy for ab initio protein structure prediction is to generate a large number of possible structures (decoys) and select the most fitting ones based on a scoring or free energy function. The conformational space of a protein is huge, and chances are rare that any heuristically generated structure will directly fall in the neighborhood of the native structure. It is desirable that, instead of being thrown away, the unfitting decoy structures can provide insights into native structures so prediction can be made progressively. First, we demonstrate that a recently parameterized physics-ba

Phase specific analysis of herpes zoster associated pain data: a new statistical approach.

Herpes zoster or shingles is a frequent occurrence in both elderly individuals and immunocompromised hosts. The pain associated with herpes zoster is the most debilitating complication of the disease. It can be described as acute pain and post-herpetic neuralgia or zoster associated pain (ZAP). The latter definition encompasses pain from the onset of disease through its resolution and provides a convenient analytic tool for evaluation of antiviral therapy. A heuristic examination of ZAP historical data suggests the existence of three phases of pain resolution: the acute, subacute and chronic p

[Hypothetical concept: the physiopathological entity of monoaminergic psychoses].

The importance of amino acid transport across the blood-brain barrier as the limiting factor in the metabolism of monoamines has been emphasized by many recent publications. Particularly critical is the transport of tryptophan, since tryptophan hydroxylase is not saturated. This transport is regulated by complex mechanisms, both at the periphery (total and free plasmatic levels and levels of the other essential amino acids) and centraly (by feedback mechanism initiated at the pre- and post-synaptic levels). The hydroxylated derivatives of tryptophan and tyrosine, i.e. 5-HTP and L-DOPA, most pr

Column Generation for Action-Based Group Handover Optimization in Large-Scale LEO Satellite Networks.

Low Earth orbit (LEO) satellite constellations are an important component of non-terrestrial networks (NTNs) in future sixth-generation (6G) systems, owing to their wide coverage and low propagation delay. However, due to the high-speed orbital movement of LEO satellites, the serving satellites of massive ground terminals change frequently, resulting in frequent and highly concurrent handover requests. Existing group handover methods usually follow a two-stage paradigm that first forms terminal groups according to similarity features and then performs target satellite selection or handover sch

Topology-aware dynamic co-evolution for robust graph learning.

Graph Neural Networks (GNNs) have achieved remarkable success in learning from graph-structured data. However, recent studies have revealed their vulnerability to adversarial attacks, where imperceptible perturbations to the graph structure can easily mislead predictions. Existing defense methods predominantly rely on global heuristic strategies, which often lead to over-smoothing in deep architectures and fail to capture localized, fine-grained structural anomalies. In this paper, we propose Topology-Aware Dynamic Co-evolution (TADC), a novel framework that reformulates graph defense as a clo

Investigating Sensorimotor Decision-Making in Depressed Individuals Using a Bayesian Approach.

Depression is among the most prevalent and disabling mental health conditions worldwide. It is associated with low mood and motivation, cognitive inefficiencies, altered reward and punishment learning, and broader executive dysfunction that can disrupt daily functioning. Depression has also been linked to differences in sensory perception, sensitivity, and processing speed. However, how depression affects sensorimotor decision-making under uncertainty remains comparatively understudied. In this study, we examined how individuals with depression process and weigh prior information relative to c

Investigating new opportunities to combat stigma in healthcare through a "universal" approach: findings from a longitudinal study of healthcare workers in Mombasa, Kenya.

Stigma plays a significant role in health and healthcare. To date, anti-stigma efforts have primarily focused on specific individual domains (e.g., mental health, HIV), resulting in fragmented efforts to address many different "stigmas." Theorists now advocate for shifting toward a universal framework to understand and respond to stigma. To empirically test the potential utility of stigma as a universal concept, we analyzed data from 453 healthcare workers participating in a prospective cohort in Mombasa, Kenya. Across four waves comprising 925 points of data (June 2023-December 2024), we foun

From inflammatory phenotypes to clinical decisions: a stratified diagnostic and therapeutic framework for CIP.

BACKGROUND: Checkpoint inhibitor pneumonitis (CIP) is an uncommon but potentially life-threatening immune-related adverse event that may interrupt effective anticancer therapy and require prolonged immunosuppression. Its clinical presentation, radiographic appearance, inflammatory biology, and response to corticosteroids vary substantially, while infection, radiation pneumonitis, tumor progression, pulmonary embolism, and pre-existing interstitial lung disease may produce overlapping findings. Conventional severity grading remains important for communication and triage but does not fully captu

Causal-Augmented Source-Free Domain Adaptation With Scale-Free Transformer for Schizophrenia Classification.

Brain functional networks (BFNs) derived from multi-site fMRI data have been widely explored using transformer-based models to extract discriminative connectivity features for the diagnosis of psychiatric disorders, such as schizophrenia (SZ). However, existing transformer-based methods ignore physiological priors like the scale-free property, causing the attention to fail to capture the intrinsic topology of BFNs. Additionally, multi-site heterogeneity makes source-free domain adaptation (DA) essential in clinical practice where data sharing is restricted. However, existing methods mainly rel

Core symptoms of PTSD across four millennia: a phenomenological and nosographic analysis - from ancient Mesopotamian texts to modern psychiatric classifications.

Post-traumatic stress disorder (PTSD) represents a transcultural recurrent response to extreme events, documented across approximately four millennia of human history. Although formally codified in Diagnostic and Statistical Manual of Mental Disorders (DSM)-III (1980), descriptions of post-traumatic symptomatology already appear in Mesopotamian cuneiform texts (2027-2003 BCE) and continue through classical literature and modern clinical observation. This historical-critical analysis assesses the phenomenological continuity of trauma-related symptoms across epochs and cultures, reconstructing t

Real-time control of urban drainage system for flood and combined sewer overflow mitigation with a novel recurrent deep reinforcement learning framework.

Urban drainage systems (UDS) are facing increasing challenges such as flooding and combined sewer overflows (CSOs) due to urbanization, aging infrastructure, and intensifying rainfall patterns. Deep reinforcement learning (DRL) has gained attention for enabling more intelligent and adaptive control strategies in UDS management, as it allows agents to learn optimal policies through interaction with dynamic environments. However, conventional DRL methods assume the next state of environment depends solely on the current state and action. In real-time UDS control, the history of environmental sta

Beyond correlation and causation: epistemic caution, anomalous findings, and COVID-19 vaccine safety.

The COVID-19 pandemic exposed science's adaptive strengths and epistemic limits. In fact, in the pandemic, mRNA vaccines are attributed to have saved millions of lives. The maxim "correlation does not imply causation" is an essential safeguard against premature causal inference. However, its use may become epistemically restrictive when the absence of established causality is treated as sufficient reason not to investigate (severe) vaccine adverse events. This article examines the conditions under which a valid cautionary principle may have contributed to premature epistemic closure. Thomas Ku

Tipping the scales: the predictive utility of the PCE-ACE ratio for criminogenic and wellbeing outcomes in a general adult population.

BACKGROUND: Adverse Childhood Experiences (ACEs) and Positive Childhood Experiences (PCEs) are each independently associated with a range of adult outcomes, including mental health, substance use, and criminal justice involvement. However, few studies have examined how the balance between these experiences influences outcomes. This study explores the predictive utility of a PCE:ACE ratio. Unlike previous measures of resiliency and risk protection scales that treat risk and protective factors as parallel dimensions, the ratio is population-level heuristic intended to capture the relative balanc

Curriculum-Based Reinforcement Learning for Autonomous UAV Navigation in Unknown Curved Tubular Conduits.

Autonomous drone navigation in confined tubular environments remains a major challenge due to the constraining geometry of the conduits, the proximity of the walls, and the perceptual limitations inherent to such scenarios. We propose a reinforcement learning (RL) approach enabling a drone to navigate unknown three-dimensional tubes without any prior knowledge of their geometry, relying solely on local observations from a Light Detection and Ranging (LiDAR) sensor and a conditional visual detection of the tube center. In contrast, the Pure Pursuit algorithm, used as a deterministic baseline, b

From Slice to Sequence: Autoregressive Tracking Transformer for Consistent 3-D Lymph Node Detection in CT Scans.

Lymph node (LN) assessment is an essential task in the routine radiology workflow, providing valuable insights for cancer staging and treatment planning. Identifying scatteredly-distributed and low-contrast LNs in 3D CT scans is highly challenging, even for experienced clinicians. Previous lesion and LN detection methods demonstrate the effectiveness of 2.5D approaches (i.e., using 2D backbone with multi-slice inputs), leveraging pretrained 2D model weights and showing improved accuracy as compared to separate 2D or 3D detectors. However, slice-based 2.5D detectors do not explicitly model inte

Machine learning approaches for predicting preventive maintenance costs of expressways in Xinjiang.

With the expansion of high-speed road mileage and the aging of road networks, maintenance workloads have increased dramatically. Accurately forecasting preventive maintenance costs is a practical approach to rationally control maintenance expenditure, enhance capital utilization efficiency, and ensure the long-term stable operation of the road network. However, the current predictive model for preventive maintenance costs fails to adequately account for variations in types of preventive maintenance measures, limiting their ability to capture the actual cost variation patterns under different e
