Association of physical activity with insulin resistance: A cross-sectional mediation analysis of the systemic immune-inflammation index in NHANES.

Higher levels of physical activity (PA) are associated with lower insulin resistance (IR), but the underlying mechanisms remain incompletely understood. Systemic inflammation may represent one pathway linking PA to IR. This cross-sectional study included 9088 adults aged ≥ 18 years from the 2005 to 2016 National Health and Nutrition Examination Survey. IR was assessed using the triglyceride-glucose index, total PA was calculated as metabolic equivalent minutes per week (MET-min/week), and the systemic immune-inflammation index (SII) was used as an inflammatory marker. Survey-weighted multivari

Elucidation of a potent pro-resolving mediator of inflammation resolution via human neutrophil-vascular endothelial cell interactions.

The acute inflammatory response is a highly coordinated programmed sequence that enables neutrophils to transmigrate from venules into tissues. Ideally self-limited, the active resolution phase produces specialized molecules that stimulate resolution and prevent collateral tissue damage from excessive neutrophil infiltration. The superfamily of pro-resolving molecules is termed specialized pro-resolving mediators including the essential polyunsaturated fatty acid-derived lipoxins, resolvins, protectins, and maresins. Given the intimate interactions between leukocytes and endothelial cells in i

Macrophage HDAC3 drives obesity and adipose tissue metabolic maladaptation through GDF3-mediated inflammation and chromatin remodeling.

Chronic inflammation is a hallmark of obesity and its associated metabolic disorders. Adipose tissue macrophages (ATMs) play a crucial role in maintaining tissue homeostasis and orchestrating metabolic inflammation. Importantly, the regulation of proinflammatory gene translation is critical for macrophage activation, a process that has been closely linked to the onset of insulin resistance and type 2 diabetes. Histone deacetylase 3 (HDAC3) is a contributing factor of inflammatory gene expression; however, its precise role in modulating adipose tissue inflammation and type 2 diabetes remains po

Pinellia ternata lectin induces mitochondrial damage and inflammation in RAW264.7 cells via mannose receptor-mediated endocytosis.

A lectin from Pinellia ternata tubers (PTL, 12 kDa) with potent pro-inflammatory activity binds to the plasma membrane and translocates into the cytoplasm. However, its cellular entry route and inflammatory toxicity mechanisms remain unclear. Here, confocal microscopy showed that fluorescein isothiocyanate (FITC)-labeled PTL (50 μg/mL) accumulated on the cell surface and was endocytosed by RAW264.7 cells time-dependently. Pharmacological inhibition revealed that PTL uptake was mediated by the mannose receptor C-type 2 (MRC2) and predominantly via clathrin-mediated endocytosis. After endocytosi

Caspase-3/GSDME-Mediated Trophoblast Pyroptosis and Reciprocal Macrophage Polarization Contribute to Inflammation in Early-Onset Preeclampsia.

Early-onset preeclampsia (EOPE) is associated with excessive apoptosis and inflammation, but the mechanistic link between these processes remains enigma. Here, we report elevated circulating pro-apoptotic proteins in EOPE patients at early pregnancy, along with concurrent CASP3 activation and GSDME cleavage in EOPE placentas. Using multiple trophoblast cell lines, we demonstrate that trophoblast cells, which highly express GSDME, undergo a shift from apoptosis to CASP3-dependent pyroptosis, driving inflammation. Notably, pyroptotic trophoblasts further induce pro-inflammatory macrophage polari

Gut microbiota mediates the antidepressant-like effects of xiaochaihutang: mechanisms involving inhibition of inflammation and enhancement of barrier function.

Depression is a common mental disorder that seriously impairs the psychological and physical health of patients. Extensive research has indicated that patients with depression exhibit disturbances in their gut microbiota. Xiaochaihutang (XCHT) is a traditional herbal recipe widely used in China, and previous studies have found it to possess antidepressant properties. However, whether XCHT can modulate depression-associated gut microbiota dysbiosis remains unclear. In this research, we investigated the impact of XCHT on the microbiota-gut-brain axis in depressed rats and elucidated the potentia

Chondrocytes reprogram chromatin in hypoxic microenvironments to activate CADM1-AS1/HDAC1 complex-mediated anti-inflammation signals.

Maintaining chondrocyte homeostasis is crucial for the effective treatment of osteoarthritis. Chondrocytes reside in a hypoxic environment under physiological conditions. This study, conducted exclusively in male mice, examines the association between epigenetics and chondrocyte homeostasis and observes significant alterations in histone acetylation in chondrocytes under hypoxic conditions. We identify a key enhancer element that regulates the long non-coding RNA CADM1-AS1, which is essential for cartilage homeostasis. This work elucidates a mechanism of epigenetic modulation that contributes

Klebsiella pneumoniae infection induces an S100A8/A9-mediated autocrine loop in human airway epithelium to amplify inflammation.

Klebsiella pneumoniae pneumonia is marked by an excessive inflammatory response that drives lung injury. The initial stages of infection and the mechanisms driving hyper-inflammation at the airway epithelial barrier are not fully defined. The alarmins S100A8/A9 are potent inflammatory mediators, and studies have indicated their importance in the host response to K. pneumoniae. Here, we show that infection with live K. pneumoniae directly induces the transcription and secretion of the S100A8/A9 heterodimer in a primary human bronchial epithelial (HBE) cell model. Furthermore, the infection sens

A Receptor-Mediated Endolysosomal Recycling Chimera Enables Catalytic TNF-α Degradation for Sustained Treatment of Mucosal Inflammation.

Mucosal inflammatory disorders such as dry eye disease (DED) and inflammatory bowel disease (IBD), driven by TNF-α-mediated immune-epithelial crosstalk, remain therapeutically challenging. Conventional inhibitors often suffer from poor mucosal permeability and transient efficacy with 1:1 binding. Although extracellular protein degradation (eTPD) technologies can eliminate pathogenic proteins, most of them operate stoichiometrically, limiting dosage efficiency and durability. Here, we report a receptor-mediated endolysosomal recycling chimera (RECYC) platform that enables catalytic, recyclable

Dectin-1 Drives Diabetic Retinopathy via Inducing Microglia-Mediated Inflammation and Blood-Retinal Barrier Breakdown.

Diabetic retinopathy (DR), a prevailing manifestation among diabetic patients, occurs as a major sight-threatening disorder. Dectin-1, as an innate immune receptor, has been notified as a critical modulator of diabetes mellitus. In this context, the implication of Dectin-1 in the process of DR that is still a conundrum will be addressed here. The diabetic mouse model was established by intraperitoneal injection of streptozotocin (STZ), and human microglia cells (HMC3) were subjected to high glucose (HG) to create cellular models of diabetes. Glucose level and body weight were recorded in mice.

CX3CL1-mediated neuronal "rescue signal" influences inflammation progression following spinal cord injury via microglia.

Spinal cord injury (SCI) is a refractory diseases that leads to lasting impairments in sensory and motor functions. Following nerve injury, CX3CL1, a neuron membrane protein, is released. Research suggests that CX3CL1 neurons possess a unique signaling mechanism for rescue. The objective of this study was to investigate the neuroprotective mechanism of CX3CL1 after spinal cord injury. Our findings indicate that injured neurons release CX3CL1, which binds to microglia and activates the NF-κB pathway. This activation downregulates IKB and P65 phosphorylation, thereby controlling the progression
