Analysis of differential effects of host plants on the gut microbes of Rhoptroceros cyatheae.

As an indispensable part of insects, intestinal symbiotic bacteria play a vital role in the growth and development of insects and their adaptability. Rhoptroceros cyatheae, the main pest of the relict plant Alsophila spinulosa, poses a serious threat to the development of the A. spinulosa population. In the present study, 16S rDNA and internal transcribed spacer high-throughput sequencing techniques were used to analyze the structure of intestinal microbes and the diversity of the insect feeding on two different plants, as well as the similarities between the intestinal microorganisms of R. cy

Phenotype-genotype association for the identification of resistance to stem borers (Diatraea spp., Lepidoptera: Crambidae) in sugarcane.

Sugarcane (Saccharum spp.) is a major crop for sugar production, but its productivity is affected by stem borers (Diatraea spp.). These insects cause significant losses by damaging the stem and disrupting the transport of photosynthates and nutrients. Although biological control has been effective in Colombia, the identification of genetic resources for the resistance would provide complementary long-term crop protection. This study aimed to identify markers associated with the resistance to insects. For this purpose, a population of 348 genotypes was evaluated by infesting 30 stalks per genot

Cell-penetrating recombinant peptides for potential use in agricultural pest control applications.

Several important areas of interest intersect in a class of peptides characterized by their highly cationic and partly hydrophobic structure. These molecules have been called cell-penetrating peptides (CPPs) because they possess the ability to translocate across cell membranes. This ability makes these peptides attractive candidates for delivery of therapeutic compounds, especially to the interior of cells. Compounds with characteristics similar to CPPs and that, in addition, have antimicrobial properties are being investigated as antibiotics with a reduced risk of causing resistance. These CP

Sex and stripping: The key to the intimate relationship between Wolbachia and host?

Wolbachia pipientis is known to infect only arthropods and nematodes (mainly filarial worms). A unique feature shared by the two Phyla is the ability to replace the exoskeleton, a process known as ecdysis. This shared characteristic is thought to reflect a common ancestry. Arthropod moulting is induced by the steroid hormone 20-hydroxyecdysone (20E) and a role for ecdysteroids in nematode ecdysis has also been suggested. Removing Wolbachia from filarial worms impairs the host's development. From analyses of the genome of Wolbachia harbored by the filarial nematode Brugia malayi and that of its

Host PGRP gene expression and bacterial release in endosymbiosis of the weevil Sitophilus zeamais.

Intracellular symbiosis (endosymbiosis) with gram-negative bacteria is common in insects, yet little is known about how the host immune system perceives the endosymbionts and controls their growth and invasion without complete bacterial clearance. In this study, we have explored the expression of a peptidoglycan recognition protein gene of the weevil Sitophilus zeamais (wPGRP); an ortholog in Drosophila (i.e., PGRP-LB) was recently shown to downregulate the Imd pathway (A. A new scorpion venom toxin paralytic to insects that affects Na+ channel activation. Purification, structure, antigenicity and mode of action.

A new toxin, BotIT2, with a unique mode of action on the isolated giant axon of the cockroach Periplaneta americana and DUM (dorsal unpaired median) neurons, has been purified from the venom of the scorpion Buthus occitanus tunetanus. Its structural, antigenic and pharmacological properties are compared to those of three other groups of neurotoxins found in Buthidae scorpion venoms. Like excitatory, depressant and alpha-type insect-selective neurotoxins, BotIT2 is toxic to insects, but shows the following common and distinctive characteristics. (a) As alpha-type toxins, BotIT2 lack strict sele

Leishmaniasis acquired in the Iraqi Theater of Operations: lessons learned.

Between August 2002 and February 2004, the Department of Defense identified 522 cases of cutaneous leishmaniasis in military personnel. This commentary examines reasons why there were so many cases of leishmaniasis during this conflict as compared with Operation Desert Storm. Lessons learned can be applied to reduce the risk to US personnel during future conflicts. Among the factors to be considered are environment, exposure, vector control, and the failure to deliver insect repellent to deployed US personnel.

[Evaluation of the effectiveness of deltamethrin (K-othrine CE) in the control of Lutzomyia longipalpis (Diptera: Psychodidae), in the municipality of Santa Rita, Paraíba, Brazil].

Deltamethrin in emulsion concentrate was applied in doses of 12.5, 25.0 and 50.0mg/m2, in households near the littoral of the State of Paraíba, where sporadic cases of visceral leishmaniasis were reported. There was a remarkable decrease in populations of several arthropods, including Lutzomyia longipalpis. This insect, however, was found again in some treated walls, 14 and 21 days after spraying. Statistical analysis indicated significant control for only two four-weeks periods, with extreme irregularity in the quantity captured.

Synergistic activity of Bacillus thuringiensis toxins against Simulium spp. larvae.

Species of Simulium spread diseases in humans and animals such as onchocerciasis and mansonelosis, causing health problems and economic loses. One alternative for controlling these insects is the use of Bacillus thuringiensis serovar israelensis (Bti). This bacterium produces different dipteran-active Cry and Cyt toxins and has been widely used in blackfly biological control programs worldwide. Studies on other insect targets have revealed the role of individual Cry and Cyt proteins in toxicity and demonstrated a synergistic effect among them. However, the insecticidal activity and interaction

A high-precision force plate method for long-term mass monitoring of fruit flies under controlled temperature and humidity.

Long-term monitoring of body mass in small insects provides critical insights into physiological regulation, metabolism, and behaviour, yet remains technically challenging owing to their extremely low mass and sensitivity to environmental fluctuations. Conventional microbalances and load cells offer high sensitivity but may have limitations in long-term stability or compatibility with controlled environmental conditions. Here, we present an optical, high-precision, force plate-based method that enables continuous, long-term monitoring of body mass in individual fruit flies under controlled tem

Toxicity and potential utility of ivermectin and moxidectin as xenointoxicants against the common bed bug, Cimex lectularius L.

The recent resurgence of the common bed bug Cimex lectularius L. throughout western industrialized nations has been facilitated in part by the insect becoming pesticide-resistant. Novel control strategies, including xenointoxication, should be considered to combat C. lectularius. Ivermectin, a U.S. Food and Drug Administration (FDA)-approved treatment for several human parasites, and the antiparasitic drug moxidectin, currently being explored in human clinical trials, were evaluated for efficacy against C. lectularius. Results showed that C. lectularius fed on ivermectin or moxidectin blood co

Insect-Specific Viruses: An overview and their relationship to arboviruses of concern to humans and animals.

The group of Insect-specific viruses (ISVs) includes viruses apparently restricted to insects based on their inability to replicate in the vertebrates. Increasing numbers of ISVs have been discovered and characterized representing a diverse number of viral families. However, most studies have focused on those ISVs belonging to the family Flaviviridae, which highlights the importance of ISV study from other viral families, which allow a better understanding for the mechanisms of transmission and evolution used for this diverse group of viruses. Some ISVs have shown the potential to modulate arb

Application of quantitative PCR for quantization of densovirus genome.

Densovirus is classified as invertebrate virus belonging to the subfamily Densovirinae of Parvoviridae family. This group of viruses infects only insects and several densoviruses have been isolated from indigenous mosquitoes and mosquito cell lines. A number of mosquitoes, especially Aedes aegypti and Ae. albopictus are important vectors of viruses, which are the major causes of dengue hemorrhagic fever and yellow fever in humans. As densoviruses do not cause any pathology in humans, these viruses have been proposed to be a potential vector for use in biological control of mosquitoes and insec

Identification of G protein-coupled receptors in the pea aphid, Acyrthosiphon pisum.

GPCRs play crucial roles in the growth, development and reproduction of organisms. In insects, a large number of GPCRs have been reported for Holometabola but not Hemimetabola. The recently sequenced pea aphid genome provides us with the opportunity to analyze the evolution and potential functions of GPCRs in Hemimetabola. 82 GPCRs were identified from the representative model hemimetabolous insect Acyrthosiphon pisum, 37 of which have ESTs evidence, and 73 are annotated for the first time. A striking difference between A. pisum, Drosophila melanogaster and Tribolium castaneum is the duplicati

Selective recognition between embryonic afferent neurons of grasshopper appendages in vitro.

Selective affinity between afferent neurons has been proposed as a major mechanism underlying the assembly of the insect peripheral nervous system during development. Afferent insect neurons establish adhesive interactions in vivo that are resistant to proteolytic degradation by elastase and independent of the basal lamina. We have tested whether afferent neurons express selective affinity for one another under more simplified and controlled conditions in vitro. We report here that (1) afferent neurons from dissociated embryonic tissue selectively aggregate within 80 min when incubated with ro

Control and surveillance of African trypanosomiasis. Report of a WHO Expert Committee.

Current estimates indicate that 60 million people are at risk of infection with human African trypanosomiasis or sleeping sickness, with about 300,000 new cases each year. However, less than 4 million people are under surveillance and only 10% of new cases are diagnosed and treated. This report of a WHO Expert Committee reviews current epidemiological information on African trypanosomiasis and its vectors, and evaluates recent advances in drug treatment and the development of tools for the control and surveillance of the disease. The report provides examples of treatment schedules, vector cont

[Control of Triatoma sordida in rural peri-domestic environment of the Porteirinha municipality, Minas Gerais, Brazil].

Triatoma sordida, a predominantly peridomestic species, is currently the triatomine species most frequently collected in Brazil. To evaluate the effectiveness of control activities against this species, a survey to determine infestation rates was carried out in October 1993 in 12 rural localities in the municipality of Porteirinha, State of Minas Gerais, Brazil. In the survey, 772 T. sordida were captured, of which 3.6% were infected with Trypanosoma cruzi. Of the 406 dwellings investigated, 34.9% were infested, and 27.6% of 695 peridomiciliar ecotopes contained T. sordida. The area around the
