Rice cold tolerance: Physiological responses, molecular regulatory networks, and improvement strategies.

Low-temperature stress is a critical abiotic constraint that severely impairs rice yield and quality, posing a substantial threat to global food security-especially as rice, a staple food for more than half of the world's population, is inherently susceptible to cold stress. This review systematically summarizes the typical phenotypic and physiological impairments induced by cold stress across key rice growth stages, and elaborates on the multi-layered molecular regulatory networks underpinning rice cold tolerance, which integrate the entire process of cold signal perception, intracellular tra

Sudarshan Kriya yogic breathing in the treatment of stress, anxiety, and depression: part I-neurophysiologic model.

Mind-body interventions are beneficial in stress-related mental and physical disorders. Current research is finding associations between emotional disorders and vagal tone as indicated by heart rate variability. A neurophysiologic model of yogic breathing proposes to integrate research on yoga with polyvagal theory, vagal stimulation, hyperventilation, and clinical observations. Yogic breathing is a unique method for balancing the autonomic nervous system and influencing psychologic and stress-related disorders. Many studies demonstrate effects of yogic breathing on brain function and physiolo

Salt stress and the wheat curl mite (Aceria tosichella) infestation reprograms barley nitrogen metabolism via nitric oxide signaling.

Salinity (NaCl) and wheat curl mite infestation profoundly alter nitric oxide (NO) metabolism in barley, with NaCl dose-dependent responses indicating that nitrogen metabolism is fine-tuned through NO-signaling pathways. Nitric oxide (NO) serves as a multifaceted regulator in plants' responses to environmental stress, acting as both a signaling molecule and a protective agent, while also exhibiting potential harmful effects. Our study investigated NO metabolism in barley (Hordeum vulgare L.) plants under salinity (50 mM and 100 mM NaCl) and wheat curl mite (WCM) infestation, exposed to either

Integrative genomic and multi-omics analyses identify oxidative stress-related pathways and druggable targets for tinnitus.

BACKGROUND: Tinnitus is a complex auditory perceptual disorder often accompanied by neuroinflammatory responses and metabolic abnormalities. Increasing evidence suggests that persistent oxidative stress, together with its interactions with immune regulation and energy metabolism, contributes to the pathophysiology of tinnitus. However, the molecular mechanisms by which oxidative stress drives tinnitus development remain to be systematically elucidated. METHODS: We integrated GWAS meta-analysis with DNA methylation, gene expression, and protein QTLs for 1,844 oxidative stress-related genes. Ca

Chronic stress as a modifier of cancer biology: Mechanisms linking neuroendocrine signaling to tumor initiation, progression, metastasis, and recurrence.

Chronic stress is a systemic disruptor of physiological homeostasis that can influence cancer biology by altering neuroendocrine, immune, metabolic, and stromal processes. While the association between stress and cancer has long been hypothesized, increasing evidence has begun to clarify molecular and cellular mechanisms through which chronic stress may contribute to tumor initiation, progression, and metastasis. This review synthesizes preclinical and clinical evidence indicating that stress-induced dysregulation of catecholamines and glucocorticoids can engage interconnected signaling networ

Endocannabinoid system modulation in bruxism: a neurobiological hypothesis and translational model of ECS-targeted intervention.

Bruxism is a multifactorial motor behavior of predominantly central origin, characterized by repetitive masticatory muscle activity and associated with dysregulation of dopaminergic, serotonergic, GABAergic, and glutamatergic pathways involved in motor control, emotional regulation, and stress responsivity. The endocannabinoid system (ECS) has emerged as a key homeostatic neuromodulator capable of integrating these neurotransmitter systems, thereby influencing pain processing, sleep-wake dynamics, and motor output. This article develops a neurobiological hypothesis based on a narrative integra

Aging as Loss of Biological Self-Correction: A Geroscience Framework for Longevity, Inflammaging, and Restored Resilience.

Longevity science has shifted from the pursuit of lifespan extension toward preservation of healthspan, resilience, and functional independence. The hallmarks of aging provide a powerful descriptive structure, but they do not by themselves specify how diverse molecular defects translate into loss of adaptive function. We propose a complementary conceptual framework in which aging is viewed as progressive impairment of biological self-correction: the distributed capacity to sense consequential perturbations, scale an appropriate response, repair or remove damage, terminate the response, and rec

Cell-Nonautonomous Regulation of Proteostasis in Aging and Disease.

The functional health of the proteome is determined by properties of the proteostasis network (PN) that regulates protein synthesis, folding, macromolecular assembly, translocation, and degradation. In eukaryotes, the PN also integrates protein biogenesis across compartments within the cell and between tissues of metazoans for organismal health and longevity. Additionally, in metazoans, proteome stability and the functional health of proteins is optimized for development and yet declines throughout aging, accelerating the risk for misfolding, aggregation, and cellular dysfunction. Here, I desc

NtMYB78 enhances salt tolerance in tobacco by coordinating stress responsive transcription and lignin deposition.

Soil salinization is a major abiotic stress limiting plant growth and crop productivity. MYB transcription factors play central roles in plant stress responses. Here, we investigated NtMYB78 using overexpression (OE) and knockout (KO) lines. Expression analysis revealed that NtMYB78 is strongly induced by salt stress. Phenotypic evaluation revealed that OE lines exhibited enhanced germination, biomass accumulation, and survival under salinity, whereas KO lines were highly sensitive. Physiological analyses indicated that NtMYB78 mitigates membrane damage and reactive oxygen species (ROS) accumu

Multi-omics profiling identifies triciribine as a biomarker-guided antimetabolite therapy for acute lymphoblastic leukemia.

Acute lymphoblastic leukemia (ALL) exhibits molecular and metabolic heterogeneity that limits therapeutic efficacy. Although antimetabolites remain central to ALL treatment, determinants of therapeutic sensitivity and resistance remain incompletely understood. Here, we investigated the activity and molecular response landscape of the purine analog triciribine (TCN) across diverse ALL models using integrated phosphoproteomics, thermal proteome profiling, and quantitative proteomics. TCN showed potent cytotoxicity across ALL cell lines and a drug response profile distinct from canonical PI3K-Akt

ATF4 Signaling in HIV-1 Infection: Viral Subversion of a Stress Response Transcription Factor.

Cellular integrated stress response (ISR), the mitochondrial unfolded protein response (UPRmt), and IFN signaling are associated with viral infections. Activating transcription factor 4 (ATF4) plays a pivotal role in these pathways and controls the expression of many genes involved in redox processes, amino acid metabolism, protein misfolding, autophagy, and apoptosis. The precise role of ATF4 during viral infection is unclear and depends on cell hosts, viral agents, and models. Furthermore, ATF4 signaling can be hijacked by pathogens to favor viral infection and replication. In this review, w…

Fermentation with aspergillus costiformis enhances the anti-migratory activity of Aquilaria sinensis leaves with possible involvement of MAPK-related pathways.

Aquilaria sinensis leaves (ALs) are an underutilized plant resource rich in polyphenols and flavonoids. Microbial fermentation has been proposed as an effective strategy to enhance the functional value of plant materials. However, its impact on the bioactivity and mechanisms of ALs remains unclear. This study aimed to evaluate the effects of fermentation with Aspergillus costiformis AHHC on the chemical composition and biological activities of ALs, and to elucidate the molecular mechanisms underlying the fermentation-induced functional changes. Non-targeted metabolomic analysis was performed t

Multi-omics analysis to uncover constitutive priming and dynamic metabolic reprogramming conferring white rust resistance in Brassica juncea.

White rust, caused by Albugo candida, is one of the most devastating diseases of Indian mustard (Brassica juncea), causing yield losses of up to 90%. Durable resistance sources within cultivated Brassica germplasm remain limited. In this study, near-isogenic lines (NILs) of B. juncea cv. Varuna harbouring resistance from an East European source (Donskaja-IV, possessing a single CC-NB-LRR protein-coding R gene) was used to investigate the molecular basis of resistance through integrated transcriptomic and metabolomic analyses at 48 and 96 hours post-inoculation (hpi). A p53-independent pathway regulates nucleolar segregation and antigen translocation in response to DNA damage induced by UV irradiation.

The nucleolus is the site of ribosomal gene transcription, processing of rRNA transcripts and maturation of preribosomal particles. Recent studies have shown that nucleoli are also involved in processes as diverse as aging, proliferation control, stress response and mitotic regulation. The proliferation-dependent nucleolar antigen pKi-67 is a sensitive marker of both proliferative activity and nucleolar integrity. We show that staining for the nucleolar-associated antigen pKi-67 is lost from nucleoli during growth arrest following UV irradiation. Surprisingly, before cells enter growth arrest,

Rationally Designed Molecules Synergistically Modulate Multifaceted Aβ Toxicity, Microglial Activation, and Neuroinflammation.

Synergistic modulation of multifaceted toxicity is the key to tackle multifactorial Alzheimer's disease (AD). The etiology of AD includes amyloid β (Aβ) amyloidosis, metal ion dyshomeostasis, reactive oxygen species (ROS), oxidative stress, mitochondrial damage, and neuroinflammation. We rationally designed multifunctional modulators by integrating pharmacophores for metal chelation, antioxidant and anti-inflammatory properties, and modulation of Aβ42 aggregation on the naphthalene monoimide (NMI) scaffold. The in vitro and cellular studies of NMIs revealed that M3 synergistically modulates me
