Cerebrospinal fluid studies in the Rett syndrome: biogenic amines and beta-endorphins.

The etiology of the Rett syndrome (RS) is unknown. Reduced function of biogenic amines has been described. Symptoms of central apnea, hyperventilation, hypothermia, peripheral analgesia, muscle rigidity, myoclonic jerks, hand stereotypy and seizures occur in RS and have been suggested as a result of elevated central beta-endorphins. It was hypothesized that a dysfunctional modulation of endogenous opiate systems and biogenic amines may be present. Cerebrospinal fluid (CSF) from 12 girls with RS was studied for beta-endorphin immunoreactivity, and biogenic amines. Lactates and pyruvate levels w

Functional evaluation techniques in mitochondrial disorders.

Most of the mitochondrial disorders affect the brain and muscle metabolism with variable degrees of impairment. The diagnostic usefulness of the following physiological and imaging methods, which also offer functional information that can be used for follow-up, is critically assessed: lactate levels, exercise testing, phosphorus magnetic resonance spectroscopy (MRS), proton MRS, functional magnetic resonance imaging, positron emission tomography, optical tissue oximetry and single photon emission tomography. Knowledge of the advantages, shortcomings and results of each method in the evaluation

Genetic Relationships Between Coronet Score and Type Traits Related to Hoof and Body Size in Holstein Cattle in Japan.

Hoof disorders in dairy cows reduce productivity and increase culling risk, causing economic losses and animal welfare concerns. The coronet score is a 5-point indicator of hoof health (1 = healthy; 5 = severe) that may be useful in breeding programs. However, genetic relationships between coronet score and type traits remain unclear. We estimated genetic correlations between coronet score and type traits related to hoof and body size in Holstein dairy cows. Data from 24,174 first-lactation Holstein cows were analyzed. Coronet scores were obtained from test-day records, whereas type traits wer

Obesity during pregnancy: contemporary evidence and clinical implications.

PURPOSE OF REVIEW: Obesity during pregnancy is a growing global health concern with implications for maternal, fetal, and intergenerational outcomes. This review highlights pathophysiological mechanisms, clinical risks, and evidence-based management strategies across the preconception, antenatal, and postpartum periods. RECENT FINDINGS: Maternal obesity amplifies pregnancy-associated insulin resistance, disrupts adipokine balance, and promotes inflammation, placental hormone dysregulation, and aberrant mTOR signaling. These mechanisms increase risks of gestational diabetes, hypertensive disor

Hypoprolactinemia: Biology, Clinical Relevance, and Diagnostic Challenges.

BACKGROUND: Prolactin (PRL) is a pleiotropic hormone, traditionally associated with lactation, but now recognised for its role in reproduction, metabolism, and neuroendocrine and immune regulation. While the clinical features and consequences of hyperprolactinemia are well known, hypoprolactinemia is poorly understood. RESULTS: The causes of PRL deficiency range from genetic mutation to acquired pituitary diseases and rare isolated disorders. Its diagnostic evaluation is often challenging, due to the lack of standardised lower reference limits. Nonetheless, growing evidence links hypoprolacti

Comparison of Enzyme Activity in Order to Describe the Metabolic Profile of Dairy Cows during Early Lactation.

Enzymatic diagnostics have practical applications in diseases of the liver, heart, pancreas, muscles, blood, and neoplastic diseases. This study aimed to compare enzyme activity to describe dairy cows' metabolism during early lactation. Based on their general health symptoms, the cows were assigned to one of three groups: acidotic, healthy and ketotic. Samples of milk, blood and rumen fluid were collected at 12 ± 5 days postpartum. Ketotic cows were characterized by the highest malondialdehyde (MDA, 76.098 nM/mL), glutathione reductase (GluRed, 109.852 U/L), superoxide dismutase (SOD, 294.22 U

Alcohol consumption before pregnancy causes detrimental fetal development and maternal metabolic disorders.

Alcohol consumption before or during pregnancy poses serious health risks to the fetus; however, the underlying mechanisms involved remain obscure. Here, we investigated whether ethanol consumption before pregnancy affects maternal or fetal health and whether pharmacological inhibition of CYP2E1, a major ethanol oxidation enzyme, by 4-methylpyrazole (4-MP) has therapeutic effects. We found that ethanol consumption (5%) 2 weeks before pregnancy resulted in a decrease in the number of viable fetuses and abnormal fetal development, and these effects were accompanied by impaired maternal glucose h

Methyl Donor Deficiency during Gestation and Lactation in the Rat Affects the Expression of Neuropeptides and Related Receptors in the Hypothalamus.

The micronutrients vitamins B9 and B12 act as methyl donors in the one-carbon metabolism involved in transmethylation reactions which critically influence epigenetic mechanisms and gene expression. Both vitamins are essential for proper development, and their deficiency during pregnancy has been associated with a wide range of disorders, including persisting growth retardation. Energy homeostasis and feeding are centrally regulated by the hypothalamus which integrates peripheral signals and acts through several orexigenic and anorexigenic mediators. We studied this regulating system in a rat m

The role of folic acid and selenium against oxidative damage from ethanol in early life programming: a review.

There are disorders in children, covered by the umbrella term "fetal alcohol spectrum disorder" (FASD), that occur as result of alcohol consumption during pregnancy and lactation. They appear, at least in part, to be related to the oxidative stress generated by ethanol. Ethanol metabolism generates reactive oxygen species and depletes the antioxidant molecule glutathione (GSH), leading to oxidative stress and lipid and protein damage, which are related to growth retardation and neurotoxicity, thereby increasing the incidence of FASD. Furthermore, prenatal and postnatal exposure to ethanol in d

Administration of histidine to female rats induces changes in oxidative status in cortex and hippocampus of the offspring.

Histidinemia is an inherited metabolic disorder biochemically characterized by high concentrations of histidine in biological fluids. Usually affected patients are asymptomatic although some individuals have mental retardation and speech disorders. Considering the high prevalence of histidinemia and the scarce information on the effects of maternal histidinemia on their progeny, we investigated various parameters of oxidative stress in brain cortex and hippocampus of the offspring from female rats that received histidine (0.5 mg/g of body weight) in the course of pregnancy and lactation. At 21

Investigation on the effect of supplementing rumen-protected forms of methionine and choline on health situation and reproductive performance of Holstein dairy cows.

Twenty Holstein dairy cows in their first and second lactation were used from 4-week prepartum through 20-week postpartum. The aim was to investigate the effect of feeding different levels of ruminally protected methionine and choline on health situation and reproductive indices of Holstein dairy cows. Cows were randomly assigned to receive one of the following treatments: 18 g day(-1) of rumen protected methionine (RPM), 60 g day(-1) of rumen protected choline (RPC), 18 g day(-1) of RPM + 60 g day(-1) of RPC and neither supplement (control). The treatments significantly affected services per

Anti-ulcer treatment during pregnancy induces food allergy in mouse mothers and a Th2-bias in their offspring.

The treatment of dyspeptic disorders with anti-acids leads to an increased risk of sensitization against food allergens. As these drugs are taken by 30-50% of pregnant women due to reflux and heartburn, we aimed here to investigate the impact of maternal therapy with anti-acids on the immune response in the offspring in a murine model. Codfish extract as model allergen was fed with or without sucralfate, an anti-acid drug, to pregnant BALB/c mice during pregnancy and lactation. These mothers developed a codfish-specific allergic response shown as high IgG1 and IgE antibody levels and positive

Indoor air pollution and childhood asthma: effective environmental interventions.

Exposure to indoor air pollutants such as tobacco smoke and dust mites may exacerbate childhood asthma. Environmental interventions to reduce exposures to these pollutants can help prevent exacerbations of the disease. Among the most important interventions is the elimination of environmental tobacco smoke from the environments of children with asthma. However, the effectiveness of reducing asthmatic children's exposure to environmental tobacco smoke on the severity of their symptoms has not yet been systematically evaluated. Dust mite reduction is another helpful environmental intervention. T

Prevention of food allergies.

Any rational approach to the prevention of food allergies must be based on the diversity of mechanisms which are involved in their pathogenesis. In addition to the IgE-mediated disorders which appear to be responsible for the largest number of cases, other immunologic (e.g., immune complex) and non-immunologic (e.g., enzymatic deficiencies) mechanisms appear to be involved. A preventive program should begin with a recognition that a potential problem exists and the identification of the individual at risk. This is accomplished by a careful documentation of IgE or other allergic reactivity in t

Challenge studies in anxiety disorders.

In psychiatry, the use of pharmacological challenges in panic disorder is unique in that the clinical phenomenon of central interest (i.e., the panic attack) can be provoked readily and assessed in the clinical laboratory setting. During the past 20 years pharmacological challenge studies have increased our knowledge concerning the neurobiology of panic disorder remarkably and may ultimately result in novel and more causal treatment strategies. Moreover, the differences in sensitivity to certain panicogens such as serotonergic agents, lactate, carbon dioxide and cholecystokinin tetrapeptide ar

H4K12 Lactylation Impairs Dopaminergic Neuron Energy Metabolism via the HIF-1α/NDUFS1 Pathway in Parkinson's Disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic (DA) neurons in the substantia nigra, with mitochondrial dysfunction central to its pathology. Recent studies have linked lactate-accumulating in PD due to mitochondrial dysfunction-to disease mechanisms through lactylation, a novel post-translational modification. However, the specific pathogenic role of lactylation in PD remains unclear. Here, H4K12 was identified as a key histone lactylation site and its role in PD pathogenesis was investigated. Plasma lactate and global histone la

An Unusual Presentation of Metformin-Associated Lactic Acidosis: A Case Report.

Metformin-associated lactic acidosis (MALA) is a rare but serious complication of metformin therapy, typically occurring in patients with underlying risk factors, such as renal impairment. While metformin is commonly prescribed for managing type 2 diabetes, its association with lactic acidosis is infrequent. We present the case of a 73-year-old woman with type 2 diabetes and atrial fibrillation (AFib) who developed MALA following acute kidney injury (AKI). The patient presented with symptoms of malaise, dizziness, abdominal pain, nausea, and vomiting, along with signs of severe metabolic acido

Management of multiple vertebral fractures during lactation in a patient with osteogenesis imperfecta type I following twin delivery.

Osteogenesis imperfecta (OI) is an uncommon bone disorder caused by mutations in type I collagen involved in bone matrix leading to increased fracture risk. There are several sub-categories within OI, with OI type I being the most common and mildest form. Women with OI considering pregnancy need to be aware of bone loss and fracture risk, particularly with lactation. We report the first case of a female with twin pregnancy and OI type I who presented with multiple vertebral fractures following delivery and postpartum lactation. Following endocrine review, she weaned breast-feeding but represen

Hypoparathyroidism in Pregnancy and Lactation: Current Approach to Diagnosis and Management.

BACKGROUND: Hypoparathyroidism is an uncommon endocrine disorder. During pregnancy, multiple changes occur in the calcium-regulating hormones, which may affect the requirements of calcium and active vitamin D during pregnancy in patients with hypoparathyroidism. Close monitoring of serum calcium during pregnancy and lactation is ideal in order to optimize maternal and fetal outcomes. In this review, we describe calcium homeostasis during pregnancy in euparathyroid individuals and also review the diagnosis and management of hypoparathyroidism during pregnancy and lactation. METHODS: We searche
