Efficacy and tolerability of folate-aminopterin therapy in a rat focal model of multiple sclerosis.

BACKGROUND: Activated macrophages in the experimental model of multiple sclerosis (MS) express folate receptor-β (FR-β), representing a promising target for the treatment of MS. Here, we both evaluated the efficacy of a novel folate-aminopterin construct (EC2319) in a rat focal model of multiple sclerosis (MS) and investigated the utility of 68Ga-labeled 1,4,7-triazacyclononane-1,4,7-triacetic acid-conjugated folate (68Ga-FOL) for assessing inflammatory lesions. In addition, we investigated whether FR-β is expressed in the brain of patients with MS. METHODS: Focal delayed-type hypersensitivit

Anti-neutrophil cytoplasmic antibodies in a rat model of trinitrobenzenesulphonic acid-induced liver injury.

BACKGROUND: In sera from patients with autoimmune liver diseases, e. g. primary sclerosing cholangitis (PSC) and autoimmune hepatitis, anti-neutrophil cytoplasmic antibodies (ANCAs) can be found. Until now, no animal model of ANCA induction in liver disease has been described. In this study, we describe a novel rat model of acute liver injury and subsequent ANCA production. MATERIALS AND METHODS: The hapten reagent 2,4,6-trinitrobenzenesulphonic acid (TNBS) was injected into the portal vein of female Lewis rats. Two experimental groups were studied: group A (TNBS/ethanol) received different T

Novel presenilin 1 mutation (S170F) causing Alzheimer disease with Lewy bodies in the third decade of life.

BACKGROUND: Cases of early-onset Alzheimer disease (AD) with an autosomal dominant inheritance pattern (familial AD [FAD]) are rare but have greatly advanced our understanding of the molecular pathogenesis of AD. We describe herein a kindred with very early-onset FAD (age, <40 years) with unusual pathological features and a novel mutation in the presenilin 1 (PSEN1) gene (S170F) and review the existing literature on very early-onset FAD. OBJECTIVE: To analyze the neuropathological and genetic features of a family with onset of AD in the third decade of life. DESIGN, SETTING, AND PARTICIPANTS

Treatment of a segmental nerve defect in the rat with use of bioabsorbable synthetic nerve conduits: a comparison of commercially available conduits.

BACKGROUND: The use of biodegradable synthetic nerve conduits for the reconstruction of segmental nerve defects has been extensively reported in both animal and human studies, with a majority of studies evaluating sensory nerve recovery. However, few studies have compared these nerve conduits for functional motor recovery. The purpose of this study was to compare three commercially available, synthetic, bioabsorbable nerve conduits and autograft with respect to compound muscle action potentials, maximum isometric tetanic force, wet muscle weight, and nerve histomorphometry. METHODS: Eighty Le

Gnotobiotic transgenic mice reveal that transmission of Helicobacter pylori is facilitated by loss of acid-producing parietal cells in donors and recipients.

Helicobacter pylori is acquired during childhood, but its mode of transmission remains unclear. A genotyped H. pylori isolate (Hp1) that expresses two classes of adhesins was introduced into the stomachs of three types of germ-free FVB/N mice to model factors that may affect spread of H. pylori in humans. Normal mice represented human hosts with normal gastric acid production. Transgenic animals expressing human alpha-1,3/4-fucosyltransferase in their gastric pit cells represented humans with normal acid production and the commonly encountered Lewis(b) histo-blood group receptor for the bacter

Cholinergic and other neurotransmitter mechanisms in Parkinson's disease, Parkinson's disease dementia, and dementia with Lewy bodies.

It is now 30 years since the beginning of intensive efforts to understand the neurotransmitter biochemistry of dementia as exemplified by Alzheimer's disease and such studies have led to the development of rational treatment strategies, which are continuing to benefit patients. However, as studies became more sophisticated and clinicians rediscovered an interest in dementia, because of the potential for symptomatic treatment, it has become clear that there are several different neurodegenerative conditions that gives rise to dementia syndromes and that each has distinct neurochemical pathology

Enzymatic PFAS destruction coupled with lanthanum fluoride recovery.

Per- and polyfluoroalkyl substances (PFAS) must be treated in large volumes to reach nanogram-per-litre regulatory limits, yet destructive technologies remain confined to the laboratory, while separation methods only concentrate the problem into hazardous waste. This study integrates three functions, enzymatic C-F bond cleavage, fluoride capture by Lewis-acidic metal-organic frameworks, and capacitive electro-swing regeneration, into a single hierarchical architecture that acts as a continuous PFAS-to-fluoride converter. Trametes versicolor laccase and its ABTS mediator are co-immobilised with

Metabolic Remodeling of the Parkinson's Disease Frontal Cortex Revealed by LC-MS/MS Metabolomics.

Parkinson's disease (PD) is a progressive neurodegenerative disorder traditionally defined by dopaminergic neuronal loss and Lewy body pathology; however, increasing evidence indicates that metabolic dysfunction contributes to both motor and non-motor manifestations of disease. While metabolomics studies in PD have largely focused on peripheral biofluids or subcortical brain regions, metabolic remodeling within cortical regions critical for cognition remains poorly characterized. Here, we applied LC-MS/MS-based untargeted metabolomics to post-mortem frontal cortex tissue from PD and neurologic

Construction of a series of pH stable Ca-based MOFs, their CO2 adsorption and catalytic activity.

In this study, three different solvent systems have been employed to investigate the effect of reaction parameters on the synthesis of four alkaline earth metal-based MOFs namely [Ca(0.5 1,4-phenyl diacetic acid)2(H2O)DMF]∞ (Ca-MOF-1), [Ca(1,4-naphthalene dicarboxylate)DMF]∞ (Ca-MOF-2), [Ca2(0.5 1,2,4,5-benzene tetracarboxylate)2(H2O)3DMF]∞ (Ca-MOF-3) and [Ca2(2,6-naphthalene dicarboxylate)2(H2O)6]∞ (Ca-MOF-4). The crystal structures of these four MOFs have been resolved through single crystal X-ray analysis and the bulk phase purity of these MOFs was assessed using PXRD and FT-IR analysis. To

Chitosan nanomedicine containing RGD peptide and PAD4 inhibitor based on phenyl boronate coupling inhibition of primary tumor growth and lung metastasis.

Stimulus-responsive nanodrugs have been extensively studied and their structural changes in the cells are important for controlled intracellular drug release. Histone citrullination of peptidylarginine deiminase 4 (PAD4) regulates the expression of tumor suppressor genes. In our previous study, compounds such as YW3-56 (356) were developed as potent PAD4 inhibitors with excellent anti-tumor activity in vitro and in vivo. To enhance the antitumor activity and improve the bioavailability, we further optimized the structure by modifying the phenylboronic acid moiety to the PAD4 inhibitor (4B). Ta

αS Oligomers Generated from Interactions with a Polyunsaturated Fatty Acid and a Dopamine Metabolite Differentially Interact with Aβ to Enhance Neurotoxicity.

It is increasingly becoming clear that neurodegenerative diseases are not as discrete as originally thought to be but display significant overlap in histopathological and clinical presentations. For example, nearly half of the patients with Alzheimer's disease (AD) and synucleinopathies such as Parkinson's disease (PD) show symptoms and pathological features of one another. Yet, the molecular events and features that underlie such comorbidities in neurodegenerative diseases remain poorly understood. Here, inspired to uncover the molecular underpinnings of the overlap between AD and PD, we inve

Ginkgolic acid promotes autophagy-dependent clearance of intracellular alpha-synuclein aggregates.

The accumulation of intracytoplasmic inclusion bodies (Lewy bodies) composed of aggregates of the alpha-synuclein (α-syn) protein is the principal pathological characteristic of Parkinson's disease (PD) and may lead to degeneration of dopaminergic neurons. To date there is no medication that can promote the efficient clearance of these pathological aggregates. In this study, the effect on α-syn aggregate clearance of ginkgolic acid (GA), a natural compound extracted from Ginkgo biloba leaves that inhibits SUMOylation amongst other pathways, was assessed in SH-SY5Y neuroblastoma cells and rat p

Analyzing the catalytic role of active site residues in the Fe-type nitrile hydratase from Comamonas testosteroni Ni1.

A strictly conserved active site arginine residue (αR157) and two histidine residues (αH80 and αH81) located near the active site of the Fe-type nitrile hydratase from Comamonas testosteroni Ni1 (CtNHase), were mutated. These mutant enzymes were examined for their ability to bind iron and hydrate acrylonitrile. For the αR157A mutant, the residual activity (k cat = 10 ± 2 s(-1)) accounts for less than 1% of the wild-type activity (k cat = 1100 ± 30 s(-1)) while the K m value is nearly unchanged at 205 ± 10 mM. On the other hand, mutation of the active site pocket αH80 and αH81 residues to alani

Microbiota-Brain Interactions in Dementia: Role of Diet in Neuroinflammation.

Aging is one of the risk factors for dementia, a group of neurocognitive disorders characterized by a progressive decline in cognitive function. Alzheimer's disease (AD) is the most prevalent, followed by vascular dementia (VaD). Other subtypes, such as frontotemporal dementia (FTD) and Lewy body disease (LBD), are less common but exhibit distinctive symptoms related to motor deficits, behavioral changes, and neuropsychiatric disturbances. Each type of dementia is associated with specific pathological features, such as amyloid plaques in AD and tau tangles in FTD. Recent research highlights th

Support-Rebalanced Cu-Ce-V/γ-Al2O3 Catalysts for Durable Fixed-Bed HCl Oxidation: Redox-Acid-Oxygen Rebalancing for Chlorine Recycling.

Catalytic oxidation of waste HCl to Cl2 enables chlorine recycling, but its practical implementation requires shaped catalysts that combine high productivity with long-term chemical and mechanical stability. Herein, a spherical Cu-Ce-V/γ-Al2O3 catalyst was developed by integrating a redox-active Cu-Ce-V oxide ensemble with mechanically robust γ-Al2O3 pellets for fixed-bed HCl oxidation. At a catalyst-bed temperature of 420 ± 5 °C under an undiluted HCl/O2 feed, the catalyst achieved an HCl conversion of approximately 78.2% and a Cl2 space-time yield of 17.04 mol∙kgact-1∙h-1. Olefin vs paraffin selectivity: A multivariate analysis of LDPE pyrolysis products over H3PO4, steam and ZnCl2 treated oat husk biochar.

The catalytic pyrolysis of low-density polyethylene (LDPE) waste into high value hydrocarbons offers a sustainable route for circular polymer economy implementation. This study applies multivariate statistical analysis and mechanistic evaluation to compare the selectivity of H3PO4, steam, and ZnCl2 treated oat husk biochar catalysts in steering LDPE pyrolysis towards olefin rich or paraffin rich products. Gas chromatograph equipped mass spectrometer (GC-MS) data was analyzed using principal component analysis (PCA) to identify compositional clustering and catalyst specific signatures. PCA reve

A topological model of the interaction between alpha-synuclein and sodium dodecyl sulfate micelles.

Human alpha-synuclein is a 140-amino acid protein of unknown function abundantly expressed in the brain and found in Lewy bodies, a characteristic feature of Parkinson's disease. Alpha-synuclein is random in water under physiological conditions, but the first approximately 100 residues interact with SDS micelles or acidic phospholipid small unilamellar vesicles and adopt an ordered conformation. The rest of the molecule remains disordered in the bulk of the solution. The conformation of the N-terminal portion of the molecule in lipids was described as an extended helix [Ramakrishnan, M., Jense

Type I collagen is the autoantigen in experimental autoimmune anterior uveitis.

This study was undertaken to identify and characterize the Ag responsible for the induction of experimental autoimmune anterior uveitis (EAAU). Melanin-associated Ag isolated from bovine iris and ciliary body was digested with the proteolytic enzyme V8 protease to solubilize the proteins and the pathogenic protein was purified to homogeneity. Lewis rats were sensitized to various fractions and investigated for the development of anterior uveitis and an immune response to the purified Ag. The uveitogenic Ag had a mass of 22 kDa (SDS-PAGE) and an isoelectric point of 6.75. The N-terminal amino a

Analysis of uveitogenic sites in phosducin molecule.

PURPOSE: Phosducin, a retinal photoreceptor protein, induces experimental autoimmune uveitis (EAU). In this study, we attempted to determine the numbers of uveitogenic sites in phosducin using synthetic peptides. METHODS: Antigen peptides were synthesized according to the amino acid sequence of the rat-derived phosducin with a peptide-synthesizer and purified by reversed-phase HPLC. First, 13 peptides covering the entire sequence of phosducin were synthesized, and each was injected into the hind footpad of Lewis rats for immunization, and induction of EAU was examined clinically and histologi
