A voxel-based morphometry study of grey matter loss in fragile X-associated tremor/ataxia syndrome.

Fragile X-associated tremor/ataxia syndrome is a neurodegenerative disorder that primarily affects older male premutation carriers of the fragile X mental retardation gene. Although its core symptoms are mainly characterized by motor problems such as intention tremor and gait ataxia, cognitive decline and psychiatric problems are also commonly observed. Past radiological and histological approaches have focused on prominent neurodegenerative changes in specific brain structures including the cerebellum and limbic areas. However, quantitative investigations of the regional structural abnormalit

Brain Correlates of the Alcohol Use Disorder Pharmacotherapy Response: A Systematic Review of Neuroimaging Studies.

BACKGROUND: Although Alcohol Use Disorder (AUD) is highly prevalent worldwide, treating this condition remains challenging. Further, potential treatments for AUD do not fully address alcohol-induced neuroadaptive changes. Understanding the effects of pharmacotherapies for AUD on the human brain may lead to tailored, more effective treatments, and improved individual clinical outcomes. OBJECTIVES: We systematically reviewed the literature for studies investigating pharmacotherapies for AUD that included neuroimaging-based treatment outcomes. We searched the PubMed, Scielo, and PsycINFO databas

Cerebellar-limbic neurocircuit is the novel biosignature of physio-cognitive decline syndrome.

Both physical and cognitive deficits occur in the aging process. We operationally defined the phenomenon as physio-cognitive decline syndrome (PCDS) and aimed to decipher its corresponding neuroanatomy patterns and neurocircuit. High resolution 3T brain magnetic resonance imaging (MRI) images from a community-dwelling longitudinal aging cohort were analysed. PCDS was defined as weakness (handgrip strength) and/or slowness (gait speed) concomitant with impairment in any cognitive domain (defined by 1.5 standard deviation below age, sex-matched norms), but without dementia or disability. Among 1

Basal ganglia/limbic striatal and thalamocortical involvement in craving and loss of control in alcoholism.

The authors explore the possible role of basal ganglia/limbic striatal and thalamocortical circuits in craving and loss of control in alcohol abuse and dependence. Alcoholics may suffer from a defect in the neuronal systems within basal ganglia/limbic striatal and thalamocortical neuronal circuits, especially within the striatoaccumbal-ventral pallidal portion of this circuit or its dopaminergic nigrotegmental modulation. Alcoholic craving may result from a neurophysiologically driven obsession resulting from overactivity within the fronto-thalamic neuronal loop, and loss of control of alcohol

Effects of sex, pubertal stage, and pubertal hormones on frontal and limbic responses to fearful and happy faces during early adolescence.

Adolescence is a key stage of brain development marked by heightened emotional reactivity, with variability across individuals and sexes. Puberty onset also initiates a cascade of changes that influence how the brain responds to social-emotional information. To more fully characterize the role of sex and puberty in the brain's response to social-emotional cues during early adolescence, we tested the main and interactive effects of sex, pubertal stage, and pubertal hormone levels on neural activation to negative and positive facial expressions. Combining baseline and 2-year follow-up data from

Fronto-limbic white matter microstructure, behavior, and emotion regulation in survivors of pediatric brain tumor.

PURPOSE: After treatment, pediatric brain tumor survivors (PBTS) face emotional and behavioral challenges, perhaps due to tumor or treatment-related changes in brain structures involved in emotion regulation, including those with fronto-limbic connections. We hypothesized that relative to healthy controls (HCs), PBTS would exhibit greater difficulties with behavior and emotional functioning, and display reduced mean fractional anisotropy (mFA) in white matter tracts with fronto-limbic connections including the cingulum bundle (CB), inferior fronto-occipital fasciculus (IFOF), and uncinate fasc

Preliminary evidence of white matter abnormality in the uncinate fasciculus in generalized social anxiety disorder.

BACKGROUND: Individuals with generalized social anxiety disorder (GSAD) exhibit exaggerated amygdala reactivity to aversive social stimuli. These findings could be explained by microstructural abnormalities in white matter (WM) tracts that connect the amygdala and prefrontal cortex, which is known to modulate the amygdala's response to threat. The goal of this study was to investigate brain frontal WM abnormalities using diffusion tensor imaging (DTI) in patients with social anxiety disorder. METHODS: A Turboprop DTI sequence was used to acquire diffusion tensor images in 30 patients with GSA

Visual associative agnosia and optic aphasia. A single case study and a review of the syndromes.

The case is presented of a patient who showed visual naming disturbances caused by a left occipital infarction. His performance on tests of visual naming, of recognition not requiring a verbal response, and of verbal-visual matching demonstrated a wide range of qualitatively different errors, including complete inability to recognize the object, access to partial semantic knowledge, and mere name finding difficulty. On the basis of the present case and of a review of the recent literature, the clinical distinction between visual associative agnosia and optic aphasia and the relation of these d

Neuroleptic-like disruption of the conditioned avoidance response requires destruction of both the mesolimbic and nigrostriatal dopamine systems.

An examination of the ability to learn an active avoidance response was made in rats subjected to 6-hydroxydopamine (6-OHDA) lesions of the individual terminal areas of the midbrain dopamine (DA) system or a lesion to all these terminal regions in one group. Lesions were made by infusing 8 micrograms (base) of 6-OHDA in 2 microliter of vehicle into the following forebrain regions (each region representing a separate group of rats); frontal cortex, nucleus accumbens, corpus striatum and a double lesion of nucleus accumbens and corpus striatum. A separate group of rats received a smaller 6-OHDA

Symptom-related damage patterns in mesial temporal lobe epilepsy with hippocampal sclerosis.

OBJECTIVE: This study was undertaken to elucidate the neuroanatomical and network substrates underlying diverse seizure symptoms in mesial temporal lobe epilepsy with hippocampal sclerosis (mTLE-HS) by applying multivariate support vector regression (SVR) to individualized structural symptom mapping and atrophy network mapping (ANM). METHODS: We retrospectively enrolled 216 patients with mTLE-HS and 460 healthy controls. Six non-mutually exclusive seizure symptoms were assessed. We performed left-right flipping of all images of right-sided mTLE-HS patients along the midline before processing

Plasma volume fraction in drug-resistant epilepsy: A DCE-MRI marker of microvascular pathology.

OBJECTIVES: Microvascular remodeling and blood-brain barrier dysfunction (BBBD) are increasingly recognized as contributors to epilepsy. However, commonly used vascular imaging markers are often state-dependent and lack spatial specificity. We aimed to (1) validate plasma volume fraction (vₚ) derived from dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) as a sensitive marker of microvascular changes; (2) characterize the vₚ alterations in patients with drug-resistant epilepsy (DRE); and (3) assess the spatial relationship between vₚ abnormalities and BBBD. METHODS: We analyzed D

Brain connectivity signatures of cognitive impairment in temporal lobe epilepsy identified by robotic assessment.

BACKGROUND: Subjects with temporal lobe epilepsy (TLE) often experience cognitive impairment in different domains. Currently, the mechanisms underlying neuropsychological dysfunction in TLE remain poorly understood. The main objective is to characterize the multivariate relationship between brain connectivity patterns and cognitive impairment detected by robotic testing in subjects with TLE. METHODS: Kinarm robotic technology was used to evaluate motor, cognitive, and sensory domains of healthy controls and individuals with TLE. Structural connectivity (SC) and functional connectivity (FC) we

Neuropathology of centenarians and supercentenarians: An autopsy study.

Studies of centenarians and supercentenarians provide insight into aging and substrates of cognitive impairment at the extreme limits of age. In this study, we examined pathologic findings in 15 centenarians (99-109 years) and 7 supercentenarians (111-115 years) who underwent autopsy at our institution during a 25-year period between 2000 and 2025 and compared them with a cohort of 24 older adults (70-90 years) who underwent autopsy during 2024-2025. Of the 22 centenarians and supercentenarians, 15 underwent full autopsies and 7 had brain only examinations. The average age at death of the cent

Characterizing the Prevalence of Psychiatric Conditions and Fear-Potentiated Startle Response in Civilians with a History of Trauma and Seizures.

BACKGROUND: Depression, posttraumatic stress disorder (PTSD), and suicidality are more prevalent among people with seizures, but few studies exist in low-resourced minoritized populations. Existing knowledge of the overlapping neurocircuitry between seizure activity in limbic regions (eg, medial temporal lobe epilepsy) and PTSD also suggests that people with seizure histories may exhibit PTSD-like alterations in their fear-potentiated startle (FPS) responses. However, this hypothesis has yet to be systematically tested. Here, we leveraged a large study on stress and trauma to evaluate the prev

Frequency-dependent seizure-suppressing effects of optogenetic activation of septal inhibitory cells in mesial temporal lobe epilepsy.

Mesial temporal lobe epilepsy (MTLE) is characterized by recurring focal seizures that arise from limbic areas and are often refractory to pharmacological interventions. We have reported that optogenetic stimulation of PV-positive cells in the medial septum at 0.5 Hz exerts seizure-suppressive effects. Therefore, we compared here these results with those obtained by optogenetic stimulation of medial septum PV-positive neurons at 8 Hz in male PV-ChR2 mice (P60-P100) undergoing an initial, pilocarpine-induced status epilepticus (SE). Optogenetic stimulation (5 min ON, 10 min OFF) was performed f

Cognitive and motor impulsivity in the healthy brain, and implications for eating disorders and obesity: A coordinate-based meta-analysis and systematic review.

Alterations in the impulse-control balance, and in its neural bases, have been reported in obesity and eating disorders (EDs). Neuroimaging studies suggest a role of fronto-parietal networks in impulsive behaviour, with evaluation and anticipatory processes additionally recruiting meso-limbic regions. However, whether distinct facets of cognitive and motor impulsivity involve common vs. specific neural correlates remains unclear. We addressed this issue through Activation Likelihood Estimation (ALE) meta-analyses of fMRI studies on delay discounting (DD) and go/no-go (GNG) tasks, alongside con

Modulation of in vitro epileptiform activity by optogenetic stimulation of parvalbumin-positive interneurons.

GABAA signaling is surprisingly involved in the initiation of epileptiform activity since increased interneuron firing, presumably leading to excessive GABA release, often precedes ictal discharges. Field potential theta (4-12 Hz) oscillations, which are thought to mirror the synchronization of interneuron networks, also lead to ictogenesis. However, the exact role of parvalbumin-positive (PV) interneurons in generating theta oscillations linked to epileptiform discharges remains unexplored. We analyzed here the field responses recorded in the CA3, entorhinal cortex (EC), and dentate gyrus (DG
