Delayed limbal mesenchymal stem cell secretome therapy attenuates NF-κB, IL-8, and MMP-9 expression following phacoemulsification-induced corneal endothelial damage.

BACKGROUND: The limbal mesenchymal stem cell secretome (LMSC-S) contains anti-inflammatory cytokines that suppress extracellular matrix (ECM) degradation in phacoemulsification-damaged corneal endothelial cells. This study aimed to evaluate the effect of LMSC-S on post- phacoemulsification ECM degradation. METHODS: Thirty-six eyes from 32 New Zealand white rabbits (Oryctolagus cuniculus) were included. The normal group (N) comprised neither exposed nor treated eyes. Control group 1 (C1) served as the control for treatment group 1 (T1), where LMSC-S was administered simultaneously with phacoemu

RSPO3 Promotes Proliferation and Self-Renewal of Limbal Epithelial Stem Cells Through a WNT/β-Catenin-Independent Signaling Pathway.

PURPOSE: R-spondin3 (RSPO3), a mammalian-specific amplifier of WNT signaling pathway, maintains the homeostasis of various adult stem cells. However, its expression at the limbus and the effect on limbal epithelial stem cells (LESCs) remains unclear. We investigated the impact of RSPO3 on the proliferation and self-renewal of LESCs and explored its molecular mechanisms. METHODS: The expression of four RSPO subtypes at the limbus were detected. Co-cultured with RSPO3 in vitro, the cell outgrowth area and cell density of human LESCs (hLESCs) were measured, along with EdU assay and evaluation of

Development and characterization of a preclinical mouse model of alkali-induced limbal stem cell deficiency.

PURPOSE: Limbal stem cell deficiency (LSCD) secondary to ocular surface alkali burn is a blinding condition that features corneal conjunctivalization. Mechanistic insights into its pathophysiology are lacking. Here, we developed a mouse model that recapitulates human disease to comprehensively delineate the clinicopathological features of a conjunctivalized cornea. METHODS: LSCD was induced in the right eyes of 6-8-week-old C57BL/6 male and female mice (n = 151) by topical administration of 0.25N sodium hydroxide on the cornea. Uninjured left eyes served as controls. Clinical, histological, p

Inhibition of TGFβ cell signaling for limbal explant culture in serumless, defined xeno-free conditions.

Outgrowths of limbal epithelium by explant culture are used to treat limbal stem cell deficiency (LSCD). The explant culture medium is always complemented with serum, a complex solution which includes TGFβ. Since TGFβ is a cytostatic effector for epithelial proliferation we examined its effect on these cultures. Limbal biopsies were set on explant culture in DMEM/F12 with 5 ng/ml EGF and cholera toxin (ChT), ITS, and 5% FBS, henceforth SHEM or a) SHEMSB=SHEM plus SB431542 an inhibitor of TGFβ signaling; b) sfSHEM = SHEM with FBS replaced by 0.05% Albumax II; and c) sfSHEMSB and sfSHEMA83 = sfS

Effects of activated omental cells on rat limbal corneal alkali injury.

Omental cells (OCs) are shown to help wound healing. The purpose of this study is to investigate if OCs improve cornea repair after alkali injury by subconjunctival injection of activated OCs in rats. Forty eight hours after limbal corneal alkali injury, fresh isolated OCs were injected subconjunctivally into the recipient rat's eye. Prior to the injury and at 0, 4 and 8 days after injury, the eyes were examined using slit lamp biomicroscopy. Corneal opacification and corneal neovascularization were graded in a masked fashion. The inflammatory response to the injury was evaluated by counting n

An Optimized Mechanical Peeling Protocol for Mouse Corneal Endothelium.

Corneal endothelium is a single layer of polygonal cells responsible for maintaining stromal dehydration and corneal transparency. Isolating corneal endothelium from murine models is exceptionally challenging without an established protocol, which historically limits the utility of mouse models to study corneal endothelial disease. Herein, a detailed method is presented to obtain a highly pure population of corneal endothelial cells via mechanical peeling of the endothelial sheet from both young and aged mice. Under a dissecting microscope, the corneal dome, including the limbus, was dissected
