On the effect of lateral stretch on the deformation energetics of biological membranes and the lipid dynamics within.

A broad range of cellular functions involve transient or persistent changes in the morphology of lipid membranes, from the organellar to the molecular scale. By and large, the thermodynamics of these remodeling processes remain to be understood. Molecular Dynamics simulations enhanced by advanced sampling methods are uniquely suited to examine and quantitate these phenomena. Here, we focus on the cellular process known as mechanosensation and use the Multi-Map simulation method to quantify how applied lateral tension impacts the energetics of both global and localized membrane perturbations in

In vitro/In vivo Evaluation of Elastic Nano-lipid Carriers of Aztreonam for Pneumonic Lungs.

BACKGROUND: The liposomal inhalation of drugs has been extensively researched for the invasion of biofilms and macrophages in the infected lungs. OBJECTIVE: The present study aimed to explore the in vitro and in vivo effects of elastic nano-lipid vesicles of aztreonam for pulmonary delivery. METHODS: Elastic nano lipid vesicles of the drug were successfully prepared with the commonly abundant lung phospholipids (LIPOID S PC-3, LIPOID PC 16:0/16:0), cholesterol, and span 20. Four such formulations were evaluated for their physicochemical properties, in vitro diffusion, and cytotoxicity. The

Synthesis and characterization of novel poly(ethylene glycol)-lipid conjugates suitable for use in drug delivery.

Liposomal formulations have been used to encapsulate and deliver a wide variety of therapeutic and diagnostic agents. Their circulation can be prolonged by the addition of neutral, hydrophilic polymers such as poly(ethylene glycol) (PEG) to the outer surface. An extended circulation lifetime allows them to take advantage of the enhanced permeability and retention effect (EPR), resulting in increased delivery to target sites. Incorporation of PEG also prevents aggregation and aids in the formation of uniform, small mono-disperse particles. This is often accomplished with the use of PEG-lipid co

Structure and orientation of a voltage-sensor toxin in lipid membranes.

Amphipathic protein toxins from tarantula venom inhibit voltage-activated potassium (Kv) channels by binding to a critical helix-turn-helix motif termed the voltage sensor paddle. Although these toxins partition into membranes to bind the paddle motif, their structure and orientation within the membrane are unknown. We investigated the interaction of a tarantula toxin named SGTx with membranes using both fluorescence and NMR spectroscopy. Depth-dependent fluorescence-quenching experiments with brominated lipids suggest that Trp30 in SGTx is positioned approximately 9 A from the center of the b

Site-directed spin-labeling analysis of reconstituted Mscl in the closed state.

The mechanosensitive channel from Escherichia coli (Eco-MscL) responds to membrane lateral tension by opening a large, water-filled pore that serves as an osmotic safety valve. In an attempt to understand the structural dynamics of MscL in the closed state and under physiological conditions, we have performed a systematic site-directed spin labeling study of this channel reconstituted in a membrane bilayer. Structural information was derived from an analysis of probe mobility, residue accessibility to O(2) or NiEdda and overall intersubunit proximity. For the majority of the residues studied,

Isolation and characterization of bovine-milk derived extracellular vesicles using a modified aqueous two-phase system.

Extracellular vesicles (EVs) are lipid bilayer-bound nanoparticles secreted by nearly all cells, with notable applications in intercellular communication and therapeutic delivery. However, conventional EV isolation techniques, such as ultracentrifugation and size exclusion chromatography, often face challenges like high cost, specialized equipment requirements, and low yield. In this study, we present a modified aqueous two-phase system (ATPS) as an alternative method for isolating EVs from raw bovine milk. This approach is scalable, cost-effective, and requires minimal specialized equipment,

[Pathogenesis of asthenozoospermia in males: Progress in research].

Asthenozoospermia is one of the major causes of male infertility, with complex pathogenesis, which involves abnormalities in sperm morphology, signaling pathways, sperm motility-related energy metabolism, and other factors. Currently, extracellular vesicles (EV) in the seminal plasma have been found with a positive effect on the spermatogenic process by releasing such self-encapsulated molecules as DAN, RNA and proteins into target cells through direct fusion with the structure similar to the lipid bilayer on the surface, and thereby regulating their function. This article reviews the research

Immune cell activation produces locally scrambled foci of plasma membrane lipids.

Most eukaryotic cells maintain a large disparity in lipid compositions between the cytosolic and external leaflets of the plasma membrane (PM) bilayer. This lipid asymmetry is maintained by energy-consuming flippase enzymes that selectively drive phospholipids into the cytosolic leaflet, often against large concentration gradients. Scramblases, activated by intracellular Ca2+ or apoptotic signaling, shuttle phospholipids down their concentration gradient to release lipid asymmetry. Such scrambling is typically evidenced by exposure of phosphatidylserine (PS) to the external leaflet and is asso

Multicomponent Lipid Nanoparticles for RNA Transfection.

Despite the wide variety of available cationic lipid platforms for the delivery of nucleic acids into cells, the optimization of their composition has not lost its relevance. The purpose of this work was to develop multi-component cationic lipid nanoparticles (LNPs) with or without a hydrophobic core from natural lipids in order to evaluate the efficiency of LNPs with the widely used cationic lipoid DOTAP (1,2-dioleoyloxy-3-[trimethylammonium]-propane) and the previously unstudied oleoylcholine (Ol-Ch), as well as the ability of LNPs containing GM3 gangliosides to transfect cells with mRNA and

Spin-label oximetry at Q- and W-band.

Spin-lattice relaxation times (T₁s) of small water-soluble spin-labels in the aqueous phase as well as lipid-type spin-labels in membranes increase when the microwave frequency increases from 2 to 35 GHz (Hyde, et al., J. Phys. Chem. B 108 (2004) 9524-9529). The T₁s measured at W-band (94 GHz) for the water-soluble spin-labels CTPO and TEMPONE (Froncisz, et al., J. Magn. Reson. 193 (2008) 297-304) are, however, shorter than when measured at Q-band (35 GHz). In this paper, the decreasing trends at W-band have been confirmed for commonly used lipid-type spin-labels in model membranes. It is conc

Ultrastructure of skin from Refsum disease with emphasis on epidermal lamellar bodies and stratum corneum barrier lipid organization.

Classic Refsum disease (RD) is a rare, autosomal recessively-inherited disorder of peroxisome metabolism due to a defect in the initial step in the alpha oxidation of phytanic acid (PA), a C16 saturated fatty acid with four methyl side groups, which accumulates in plasma and lipid enriched tissues (please see van den Brink and Wanders, Cell Mol Life Sci 63:1752-1765, 2006). It has been proposed that the disease complex in RD is in part due to the high affinity of phytanic acid for retinoid X receptors and peroxisome proliferator-activated receptors. Structurally, epidermal hyperplasia, increas

Proteins that bind and move lipids: MsbA and NPC1.

Membrane proteins that bind and transport lipids face special challenges. Since lipids typically have low water solubility, both accessibility of the substrate to the protein and delivery to the desired destination are problematical. The amphipathic nature of membrane lipids, and their relatively large molecular size, also means that these proteins must possess substrate-binding sites of a different nature than those designed to handle small polar molecules. This review considers two integral proteins whose function is to bind and transfer membrane lipids within or across a membrane. The first

Bicelles stabilize a compact conformation of opsin with enhanced α-helical packing.

Bicelles form a disc-like planar lipid bilayer surrounded by short-chain lipids at the peripheral rim. Therefore, they are well-suited to study purified transmembrane proteins in a more native-like environment. In this study, we investigated the physico-chemical properties of bicelles using transmission electron microscopy, dynamic light scattering, and fluorescence spectroscopy. The G protein-coupled receptor rhodopsin served as a prototypical membrane protein, which we reconstituted into bicelles having an average diameter of 11.6 ± 0.6 nm that increased to 14.9 ± 0.7 nm upon incorporation o

Influence of tail structure on thermotropic phase behavior, stability, supramolecular organization, and vesicle properties of nonionic β-alaninol-based amphiphiles: Homologs of N-acylethanolamines.

The hydrophobic tail structure of lipids or amphiphiles plays a critical role in governing the self-assembly, structure, dynamics, and function. In the present work, β-alaninol-based amphiphiles bearing cholesterol, saturated, unsaturated, and branched fatty acid-based tails were synthesized in N-acyl and N-cholesteroyl forms and systematically investigated to elucidate how tail architecture controls physicochemical properties. Results revealed that variations in amphiphile tail structure influenced pronounced differences in phase transition behavior and supramolecular packing. Amphiphiles bea

Mechanistic Control of Cell Adhesion at Ligand-free Soft Membrane Interfaces.

Cell adhesion at soft interfaces is governed by a complex interplay between interaction forces and interfacial dynamics whose relative contributions remain difficult to disentangle. Here, we investigate the adhesion of yeast cells (Saccharomyces cerevisiae) on supported lipid bilayers with controlled composition, including systems containing the glycolipid ohmline (OHM), which serve as well-defined model membrane interfaces. By combining QCM-D measurements with interfacial characterization, including dilatational rheology, we show that adhesion proceeds through two kinetically distinct regimes…

"Interfacial Activation" High-Fidelity Sensing Strategy: Two-Photon Fluorescence Imaging of Transmembrane H2S Release.

Precise elucidation of dynamic chemical events at the cell membrane interface is vital for understanding fundamental biological processes. However, current fluorescent tools often fail to confine signal generation strictly to the membrane, leading to background interference that compromises imaging fidelity. To address this, we propose an "Interfacial Activation" strategy that transforms the cell membrane from a passive anchoring point to an active signal trigger. This is realized using NACP, an amphiphilic two-photon fluorophore that remains completely nonemissive in aqueous media via aggrega

Formation of tubules and helical ribbons by ceramide phosphoethanolamine-containing membranes.

Ceramide phosphoethanolamine (CPE), a major sphingolipid in invertebrates, is crucial for axonal ensheathment in Drosophila. Darkfield microscopy revealed that an equimolar mixture of bovine buttermilk CPE (milk CPE) and 1,2-dioleoyl-sn-glycero-3-phosphocholine (diC18:1 PC) tends to form tubules and helical ribbons, while pure milk CPE mainly exhibits amorphous aggregates and, at low frequency, straight needles. Negative staining electron microscopy indicated that helices and tubules were composed of multilayered 5-10 nm thick slab-like structures. Using different molecular species of PC and C
