Genetically encoded calcium indicators (GECIs) enable imaging techniques to monitor changes in intracellular calcium in targeted cell populations. Their large signal-to-noise ratio makes GECIs a powerful tool for detecting stimulus-evoked activity in sensory neurons. GECIs facilitate population-level analysis of stimulus encoding with the number of neurons that can be studied simultaneously. This population encoding is most appropriately done in vivo. Dorsal root ganglia (DRG), which house the soma of sensory neurons innervating somatic and visceral structures below the neck, are used most ext

The P2X(3) receptor is a ligand-gated cation channel activated by the binding of extracellular adenosine 5'-triphosphate (ATP), an agent that has been suggested to have a role in the nociceptive pathway after tissue and nerve injury. After peripheral nerve injury, both down regulation and up regulation of the P2X(3) receptor in sensory ganglion neurons have been observed. The purpose of this study was to examine the precise regulation of P2X(3) mRNA expression in primary sensory neurons after nerve injury. We used two nerve injury models in the rat, the transection of the tibial and common per

BACKGROUND: Cleft lip and/or palate (CLP) is one of the most prevalent congenital craniofacial anomalies, with a global incidence of approximately 1 in 700-1000 live births. Surgical management varies substantially across centers in technique, timing, and perioperative care, which complicates evidence-based standardization.
OBJECTIVE: To synthesize high-level evidence from systematic reviews and meta-analyses on the surgical and perioperative management of CLP in children aged 0-6 years, with emphasis on technique comparisons, key clinical outcomes, and analgesic protocols.

BACKGROUND: Trigeminal neuralgia (TN) is a neuropathic facial pain disorder that frequently mimics odontogenic and other orofacial pain conditions, leading many patients to initially seek dental care. This study aimed to characterize the clinical features and diagnostic pathways of patients with TN presenting to a tertiary orofacial pain clinic.
METHODS: This single-center retrospective study included newly diagnosed patients with TN who attended a tertiary orofacial pain clinic between 2024 and 2025. Data regarding their demographic characteristics, pain features, pain intensity, trigger fac

An ultrasound-guided trigeminal nerve block using a coronoid approach was performed in a male neutered Jack Russell Terrier, aged 12 years, undergoing orbital exenteration for a retrobulbar mass. The previously reported temporal approach for this block was contraindicated as the location of the mass risked intersecting the tumour with the needle. With the dog anaesthetized and in left lateral recumbency, a microconvex transducer was placed caudal to the orbital ligament to obtain a transverse view of the pterygopalatine fossa. The angle of the probe was adjusted until the frontal bone and sphe

BACKGROUND: Complications during endoscopic sinus surgery often result from unfavorable anatomy. The prevalence rates of such anatomic findings vary tremendously in the literature, in part due to the small, homogenous populations previously studied.
OBJECTIVE: To characterize the prevalence of sinonasal anatomic variants among ethnic groups and genders seen at an academic medical center.
METHODS: This is a retrospective cross-sectional study of adult subjects who underwent CT imaging of the sinuses from January 2019 to November 2020 at a tertiary academic setting. CT scans were analyzed syst

Inhaled manganese (Mn) can enter the olfactory bulbs via the olfactory epithelium, and can then be further transported trans-synaptically to deeper brain structures. In addition to olfactory neurons, the nasal cavity is innervated by the maxillary division of the trigeminal nerve that projects to the spinal trigeminal nucleus. Direct uptake and transport of inhaled metal particles in the trigeminal system has not been investigated previously. We studied the uptake, deposition, and clearance of soluble Mn in the trigeminal system following nose-only inhalation of environmentally relevant concen

Rat trigeminal neurons innervating tooth pulps were retrogradely labelled with fluorogold and analysed enzyme- and immunohistochemically for their content of substance P, calcitonin gene-related peptide, fluoride-resistant acid phosphatase, GM 1 ganglioside, carbonic anhydrase and neurofilament protein. The data showed that both small, medium-sized and large trigeminal neurons were labelled after fluorogold deposition in maxillary molar pulps, with a majority of the cells being medium-sized and large. Less than 2% of the pulpal neurons showed substance P-like immunoreactivity. Fifty-six per ce

Dental injection needle migration is a rare complication of orthodontal procedures. When these needles fracture, they typically dislodge into the cervical space or the facial musculature. Migration into the cranial vault is difficult because of the obstacle created by the skull base. We report a rare case of intracranial migration of an anesthetic injection needle through the foramen ovale. A 59-yr-old man underwent the extraction of a right maxillary molar. The distal end of a 25-gauge injection needle broke into his pterygoid musculature, causing him pain while chewing. Vascular imaging obta

Directed isolated face grooming following unilateral chronic constriction injury to the infraorbital nerve (IoN-CCI) is a unique measure of spontaneous neuropathic pain. Variability between rats and the limited duration of the increased face grooming behavior has hampered its usefulness. We studied three possible sources of variability: variations in surgery, pre-existing differences in nocifensive behavior between the rats and variation in time. Three different types of IoN lesion were performed: loose ligation (CCI), tight ligation (CCI-T) and partial tight ligation (PTL, Seltzer method); th

Endothelins, acting through specific endothelin ET(A) and/or ET(B) receptors, participate in nociceptive processing in models of cancer, inflammatory and neuropathic pain. The present study investigated which cell types express endothelin receptors in the trigeminal ganglion, and the contribution of mechanisms mediated by endothelin ET(A) and ET(B) receptors to orofacial heat hyperalgesia induced by unilateral constriction of the infraorbital nerve (CION). Both receptor types were identified by immunohistochemistry in the trigeminal ganglion, ET(A) receptors on small-sized non-myelinated and m

BACKGROUND: Infraorbital neuralgia, one of the rare causes of facial pain, lacks systematic treatment guidelines because few studies on the topic have been published. We previously found that 42°C percutaneous nondestructive pulsed radiofrequency (PRF) treatment could achieve satisfactory pain relief for infraorbital neuralgia patients. However, patients who responded poorly to PRF had no other ideal treatment options until now. Recently, standard PRF combined with 60°C continuous radiofrequency (CRF) was successfully performed on trigeminal neuralgia patients and achieved a promising effectiv

OBJECTIVE: This study was undertaken to analyze the results of a novel surgical method-the endoscopic prelacrimal recess approach (PLRA)-in patients with tumors involving the pterygopalatine fossa (PPF) and infratemporal fossa (ITF). The surgical technique and indications for this approach are also discussed.
METHODS: The authors analyzed data from 7 cases involving patients who underwent resection of PPF and ITF tumors by means of the endoscopic PLRA from 2004 to 2013. Preoperative and postoperative imaging studies were available in all cases and were reviewed. The surgical specimens were al

PURPOSE: Perceived taste intensity and taste quality identification on localized regions of the palate and tongue were examined for 9 patients before orthognathic surgery and again at 1 to 2 and 6 to 9 months after surgery. Taste function would be at risk on the palate after maxillary Le Fort I osteotomy (LFI) and on the tongue after mandibular sagittal split osteotomy (SSO) because of potential damage to peripheral nerves conducting afferent chemosensory information from these regions.
PATIENTS AND METHODS: Three patients had LFI and SSO, 1 had LFI only, and 5 had SSO only. Patients rated ta

PURPOSE: To study different types of extension of sphenoid sinus pneumatization detected by CT and examine their correlation with other anatomical variants.
METHODS: Sphenoid sinus pneumatization was characterized (780 hemi-sinuses) on CT scans. Its incidence and correlations with related anatomical variants were evaluated. Dimensions of surgical window were measured in the lateral type, where pneumatization extends beyond a line connecting Vidian (pteregoid) canal (VC) and foramen rotundum (FR).
RESULTS: The lateral recess of the sphenoid sinus was pneumatized in 27.3% of patients with a me
