[The clinical characteristics of 2 outbreaks of trichinelliasis].

In 1988-1989 two outbreaks of trichinosis were recorded. The first case was group disease developed in 3 members of the family. The source of infection was rissoles rolled in cabbage leaves [correction of goloubets] stuffed with minced pork. The second outbreak affected 13 persons who had eaten home-made pork sausage and fat. All the patients displayed fever, weakness, muscular pain, subcutaneous fat edema, predominantly of the face. The patients suffered from dry mouth, thirst, deteriorated appetite, exanthema, eosinophilia, more occasionally, nausea, vomiting, abdominal pain and diarrhea. Tr

Reinfection of Somali children with Trichuris trichiura after chemotherapy: relevance of immunostimulation.

The intestinal helminth status of an age-stratified sample (6 to 20 years old) from a Somalian community has been assessed and the typical pattern of highly aggregated parasite distribution found. A reinfection study on a sample of 40 children (treated and untreated with a pentapeptide identical to the active site of the thymic hormone thymopoietin) seemed to indicate that immunological factors play a significant role in modulating the population dynamics of infection in endemic communities.

The effect of riboflavin on the hematologic changes in iron supplementation of schoolchildren.

A supplementation trial was carried out in 101 children, 6-12 years of age, in 3 primary schools in a rural area. Their hemoglobin level and PCV (mean +/- SD) were 11.64 +/- 1.21 g/dl and 0.356 +/- 0.028 respectively, 74% of them were anemic and the hemoglobin level were correlated with the MCHC (P < .01) . Fifty-one per cent of them had hookworm infection and all those with hemoglobin levels below 10 g/dl had hookworm infection, but there was no difference in mean hemoglobin level between those with hookworm infection and those without. The children were divided into 3 groups: Group I compris

Activity of antifilarial drugs on microfilaremia in the treatment of loiasis: a systematic review.

BACKGROUND: Loiasis, caused by the nematode/filaria Loa loa, presents a major health burden in Central and West Africa. Despite the growing recognition of loiasis' medical significance, current antifilarial drugs remain inadequate in terms of efficacy and safety, particularly for individuals with hypermicrofilaremia. This systematic review aims to evaluate the efficacy of antifilarial treatment regimens for reducing L. loa microfilaremia and provide guidance on treatment strategies. METHODS: A systematic review was conducted to evaluate the efficacy of antifilarial treatment regimens on reduc

Current status of drug therapy for alveolar echinococcosis.

Alveolar echinococcosis (AE) is a chronic zoonotic parasitic disease caused by infection with Echinococcus multilocularis. AE is associated with a high mortality rate and poses a significant threat to human health. The primary treatment for AE is surgical resection of the lesions; however, owing to its long incubation period and insidious disease progression, many patients are diagnosed only after the onset of complications such as liver cirrhosis, jaundice, and portal hypertension, which preclude curative surgical intervention. For patients who are unwilling or unable to undergo surgery, life

Insights into the Role of Compendial/Biorelevant Media on the Supersaturation Behaviour of Drug Combination (Drug-Drug Interaction) and Precipitation Inhibition by Polymers.

Combination drug therapy (CDT) plays an immense role in the treatment of various diseases such as malaria, hypertension, cancer, HIV-AIDS, helminthiasis, and many more. However, in vitro drug-drug interaction (DDI) is not well reported for better efficacy of CDT. In DDI one drug may enhance the precipitation of other drugs thereby reducing the advantage of CDT. Herein, we report DDI in terms of in vitro precipitation of drugs with albendazole and mebendazole. This may be the first report to propensate the possibility of either drug precipitation in the combination. These drugs are categorized

Soil-Transmitted Helminthiasis in Children from a Rural Community Taking Part in a Periodic Deworming Program in the Peruvian Amazon.

Children in the Peruvian Amazon Basin are at risk of soil-transmitted helminths (STH) infections. This study aimed to determine the prevalence of STH infection in children from a rural Amazonian community of Peru and to elucidate epidemiological risk factors associated with its perpetuation while on a school-based deworming program with mebendazole. Stool samples of children aged 2-14 years and their mothers were analyzed through direct smear analysis, Kato-Katz, spontaneous sedimentation in tube, Baermann's method, and agar plate culture. A questionnaire was administered to collect epidemiolo

Drug Screening for Discovery of Broad-spectrum Agents for Soil-transmitted Nematodes.

Soil-transmitted nematodes (STNs), namely hookworms, whipworms, and ascarids, are extremely common parasites, infecting 1-2 billion of the poorest people worldwide. Two benzimidazoles, albendazole and mebendazole, are currently used in STN mass drug administration, with many instances of low/reduced activity reported. New drugs against STNs are urgently needed. We tested various models for STN drug screening with the aim of identifying the most effective tactics for the discovery of potent, safe and broad-spectrum agents. We screened a 1280-compound library of approved drugs to completion agai

Discovery of genomic intervals that underlie nematode responses to benzimidazoles.

Parasitic nematodes impose a debilitating health and economic burden across much of the world. Nematode resistance to anthelmintic drugs threatens parasite control efforts in both human and veterinary medicine. Despite this threat, the genetic landscape of potential resistance mechanisms to these critical drugs remains largely unexplored. Here, we exploit natural variation in the model nematodes Caenorhabditis elegans and Caenorhabditis briggsae to discover quantitative trait loci (QTL) that control sensitivity to benzimidazoles widely used in human and animal medicine. High-throughput phenoty

Monitoring the efficacy of drugs for neglected tropical diseases controlled by preventive chemotherapy.

In the last decade, pharmaceutical companies, governments and global health organisations under the leadership of the World Health Organization (WHO) have pledged large-scale donations of anthelmintic drugs, including ivermectin (IVM), praziquantel (PZQ), albendazole (ALB) and mebendazole (MEB). This worldwide scale-up in drug donations calls for strong monitoring systems to detect any changes in anthelmintic drug efficacy. This review reports on the outcome of the WHO Global Working Group on Monitoring of Neglected Tropical Diseases Drug Efficacy, which consists of three subgroups: (i) soil-t

Human cystic echinococcosis: epidemiologic, zoonotic, clinical, diagnostic and therapeutic aspects.

This review represents an updated scenario on the transmission cycle, epidemiology, clinical features and pathogenicity, diagnosis and treatment, and prevention and control measures of a cestode parasite Echincoccus granulosus (E. granulosus) infection causing cystic echinococcosis (CE) in humans. Human CE is a serious life-threatening neglected zoonotic disease that occurs in both developing and developed countries, and is recognized as a major public health problem. The life cycle of E. granulosus involves a definitive host (dogs and other canids) for the adult E. granulosus that resides in

Ascaris lumbricoides: an overview of therapeutic targets.

A. lumbricoides is the largest of the common nematode parasites of man and has been associated with intestinal pathology, respiratory symptoms and malnutrition in children from endemic areas. Current anthelmintic treatments have proven to be safe. However, a reduced efficacy of single dose drugs has been reported. In veterinary practice, anthelmintic drug resistance is an irreversible problem. Thus, research and development of sensitive tools for early detection of drug resistance as well as new anthelmintic approaches are urgently needed. In this review, we summarized data providing informati

In vitro metacestodicidal activities of genistein and other isoflavones against Echinococcus multilocularis and Echinococcus granulosus.

Echinococcus multilocularis and Echinococcus granulosus metacestode infections in humans cause alveolar echinococcosis and cystic echinococcosis, respectively, in which metacestode development in visceral organs often results in particular organ failure. Further, cystic hydatidosis in farm animals causes severe economic losses. Although benzimidazole derivatives such as mebendazole and albendazole are being used as therapeutic agents, there is often no complete recovery after treatment. Hence, in searching for novel treatment options, we examined the in vitro efficacies of a number of isoflavo

Hydatid cyst of the liver-criteria for the selection of appropriate treatment.

The appropriate treatment of hydatid cysts of the liver is determined by several factors, namely the patient, the cyst, the therapeutic resources and the physician. Characteristics of cysts, can be described by ultrasonography (US). Based on US images, we can classify hydatid cysts, according the evolutionary phase of the larval parasite and to choose the most appropriate therapeutic approach. US is also important to evaluate the efficacy of the treatment. Concerning the therapeutic methods, surgery had long been the only treatment available for the hydatid cyst of the liver. Beginning the 197

Defective mucosal immunity and normal systemic immunity of Mongolian gerbils, Meriones unguiculatus, to reinfection with Strongyloides venezuelensis.

The systemic and local protective activity of Mongolian gerbils was examined after re-infection with Strongyloides venezuelensis. Mongolian gerbils were unable to expel S. venezuelensis adult worms from the intestine for over ten weeks after a primary infection. Therefore, immune animals were prepared by treating with mebendazole four weeks after a primary infection and then they were challenged by different maturation stages of the parasite; subcutaneous inoculation with the infective larvae (L3) obtained by faecal culture, oral administration of L3 obtained from the lungs of rats three days

Orphan Medications: An Innovative and Promising Approach against Leishmaniasis.

Over the decades, management of protozoan disease – ‘leishmaniasis’ has been considerably challenging on account of inadequacy in drug efficacy, restricted drug availability, unattainability of treatment by impoverished, and the disease’s endemicity in several countries. At present, ≈ 90 countries are affected by leishmaniasis where visceral leishmaniasis (VL) cases reported are ≈ 30,000 and 1 million cases of cutaneous leishmaniasis (CL) (World Health Organization, 2023). Currently no licensed vaccines are commercially available and therefore, advancement in the area of ‘Orphan Drugs’ offers
