We studied plasma melatonin profiles by radioimmunoassay in nine patients suffering from human african trypanosomiasis and six healthy controls matched according to the age and the photoperiodic conditions. In order to establish a method for the estimation of death time (DT) from measuring melatonin (MT) contents in pineal bodies (PBs) and biological fluids, 85 cadavers were investigated--44 dead in Sapporo (N 43 degrees 4', E 141 degrees 21') and 41 in Tokyo (N 35 degrees 39', E 139 degrees 44'). The paper deals with the central regulation of the circadian rhythm of the melatonin in the pineal gland by light according to the literature. Examples of the effects of melatonin and their inclusion in the total endocrine complexity are pointed out. Possibilities for a therapy of chronobiological diseases with bright light resulting from animal experiments are discussed in connection with requirements for a future application to the therapy of human beings.

Normal control subjects were examined on three separate occasions with light of sufficient intensity to suppress nocturnal plasma melatonin concentrations. One hour of light was given at each of the following times: (a) 2100-2200h; (b) midnight to 0100h; (c) 0400-0500h. Melatonin synthesis was just becoming apparent at 2100h. Serotonin as well as melatonin introduced into the 3-rd cerebral ventricle in doses of 120 mug blocked induction of ovulation in rabbits; it was also observed that the ovulation-blocking effect of serotonin was much more effective than that of melatonin. The inhibitory effect of both indolamines on this process was more evident in summer than in spring. The role of serotonin and melatonin as mediators blocking the release of hypothalamic gonadotropin-releasing hormones (GRH) and the participation of these amines in the regulation of seasonal sexual rhythms are discussed.
